The identification of macrophyte reference communities represents a significant challenge in Mediterranean zones, where few aquatic taxa exist compared to other regions of the world. In the highly seasonal rivers of Portugal, we explored the contribution of bryophytes to enhance the compositional and ecological characterization of reference conditions. We performed partitioning, classification, and ordination analyses and obtained eight groups of bryophytic assemblages explained by water availability and quality. Groups A, B, and H assemblages are type-specific of the inland areas of the northern eco-region, reflecting the neutral minerotrophic currents with summer low currents. Groups C and D are type-specific of acidic oligotrophic shifting currents dependent of precipitation events in the mountainous streams of the North Atlantic eco-region. Groups E, F, and G are widespread in continental northern, central, and southern zones in neutral and highly mineralized watercourses with seasonal dryness. We propose bryophytes as a tool in the characterization of the reference conditions for two main reasons: (i) many of the recorded taxa are capable of withstanding the natural seasonal desiccation, and (ii) the type-specific bryophytes have perennial life-strategies and can be assessed at any time of year as a support for other type-specific macrophytes in compositional assessments of reference conditions.  相似文献

One of the most important structures for an individual’s survival is a refuge, especially for species subject to significant predatory pressure or living in environments affected by severe climatic conditions. We studied how wood mice (Apodemus sylvaticus), inhabiting a Mediterranean environment, use nests and the factors that could be determining this use. Radio-tracking of 16 mice allowed us to detect 24 diurnal nests, which were used with a high fidelity rate (1.75 nests/animal). Nest sites that had been identified using radio-tracking were matched with 24 randomly selected non-nest sites (located within an area defined by the sum of all individual home ranges), and all sites were characterized according to their macro- and micro-habitat parameters. Using a generalized linear model, we tested three hypotheses: (1) a nest’s use is influenced by its proximity to food patches; (2) the degree of sheltering provided by each site influences its use; and (3) nest use is constrained by a combination of food- and shelter-related features. Females had higher nest fidelity than males, and the degree of sheltering provided by each site had a positive significant influence on its use. Nests offering higher protection (e.g., under thick vegetation), good drainage (e.g., sleep slopes), and lower human and predatory disturbance (far from orchards) were most often used. The proximity to food patches seemed to negatively influence nest use. Results indicate the need to preserve patches of natural dense vegetation near riparian habitats, which is in agreement with the focus of mammalian protection plans in Mediterranean agricultural landscapes. These patches are important areas for Apodemus sylvaticus, an acorn disperser species, which is preyed on by raptors and mammalian carnivores.  相似文献

Cells from animals, plants and single cells are enclosed by a barrier called the cell membrane that separates the cytoplasm from the outside. Cell layers such as epithelia also form a barrier that separates the inside from the outside or different compartments of multicellular organisms. A key feature of these barriers is the differential distribution of ions across cell membranes or cell layers. Two properties allow this distribution: 1) membranes and epithelia display selective permeability to specific ions; 2) ions are transported through pumps across cell membranes and cell layers. These properties play crucial roles in maintaining tissue physiology and act as signaling cues after damage, during repair, or under pathological condition. The ion-selective self-referencing microelectrode allows measurements of specific fluxes of ions such as calcium, potassium or sodium at single cell and tissue levels. The microelectrode contains an ionophore cocktail which is selectively permeable to a specific ion. The internal filling solution contains a set concentration of the ion of interest. The electric potential of the microelectrode is determined by the outside concentration of the ion. As the ion concentration varies, the potential of the microelectrode changes as a function of the log of the ion activity. When moved back and forth near a source or sink of the ion (i.e. in a concentration gradient due to ion flux) the microelectrode potential fluctuates at an amplitude proportional to the ion flux/gradient. The amplifier amplifies the microelectrode signal and the output is recorded on computer. The ion flux can then be calculated by Fick’s law of diffusion using the electrode potential fluctuation, the excursion of microelectrode, and other parameters such as the specific ion mobility. In this paper, we describe in detail the methodology to measure extracellular ion fluxes using the ion-selective self-referencing microelectrode and present some representative results.  相似文献

Plasmodium ovale and Plasmodium malariae, two of the four human malaria parasites, are usually found at very low prevalence in East Asia, even in areas with intense malaria transmission. In this article, Fumihiko Kawamoto, Qing Liu, Marcelo Ferreira and Indah Tantular review data obtained in recent field surveys, using alternative diagnostic methods such as acridine orange staining and PCR-based methods, to evaluate the prevalence of these two malaria species in East Asia. They argue that these species might be much more prevalent in East Asia than reported previously. In addition, they discuss the implications of sequence variations found in the small subunit ribosomal RNA genes of the two species targeted by diagnostic PCR and compare morphological criteria for speciation of malaria parasites stained with Giemsa and acridine orange.  相似文献

Charcot-Marie-Tooth (CMT) disease is one of the most common heritable neuromuscular disorders, affecting 1 in every 2500 people. Mutations in LITAF have been shown to be causative for CMT type 1C disease. In this paper we explore the subcellular localization of wild type LITAF and mutant forms of LITAF known to cause CMT1C (T49M, A111G, G112S, T115N, W116G, L122V and P135T). The results show that LITAF mutants A111G, G112S, W116G, and T115N mislocalize from the late endosome/lysosome to the mitochondria while the mutants T49M, L122V, and P135T show partial mislocalization with a portion of the total protein present in the late endosome/lysosome and the remainder of the protein localized to the mitochondria. This suggests that different mutants of LITAF will produce differing severity of disease. We also explored the effect of the presence of mutant LITAF on wild-type LITAF localization. We showed that in cells heterozygous for LITAF, CMT1C mutants T49M and G112S are dominant since wild-type LITAF localized to the mitochondria when co-transfected with a LITAF mutant. Finally, we demonstrated how LITAF transits to the endosome and mitochondria compartments of the cell. Using Brefeldin A to block ER to Golgi transport we demonstrated that wild type LITAF traffics through the secretory pathway to the late endosome/lysosome while the LITAF mutants transit to the mitochondria independent of the secretory pathway. In addition, we demonstrated that the C-terminus of LITAF is necessary and sufficient for targeting of wild-type LITAF to the late endosome/lysosome and the mutants to the mitochondria. Together these data provide insight into how mutations in LITAF cause CMT1C disease.  相似文献

Understanding native communities is a crucial step for the management of biological nitrogen fixation, since they may be either a source of efficient strains or a limiting factor when efficient strains need to be introduced. This work aimed to evaluate the density, diversity and efficiency of Leguminosae nodulating bacterial (LNB) communities and their component strains in soils under various land use systems (LUSs): pristine forest, agriculture, pasture, agroforestry, young secondary forest, and old secondary forest,. The LNB communities were trapped from these soils by using the promiscuous host siratro under controlled conditions. We also studied their relationships with physical and chemical attributes of the soil. Agroforestry and agriculture soil samples induced the highest number of nodules in siratro, while forest soil samples induced the lowest number of nodules. No relationship was found between LNB and Leguminosae species diversity in the LUSs. The soil chemical variables that were most related to differences in nodule number and shoot dry matter weight of plants inoculated with soil suspensions of the LUSs were, respectively: Ca2+, Mg2+, base saturation, exchangeable bases and Cu2+; and pH, cation exchange capacity, B, Cu2+ and clay. Although, LNB communities from all LUSs were efficient under controlled and similar conditions, they were found to be composed of strains with variable efficiency: inefficient, efficient, highly efficient and superior efficiency. Efficient strains occurred at the highest frequency in all LUSs. The isolated strains presented similar and new sequences that were phylogenetically related to well known LNB genera in α-and β-Proteobacteria. Unusual genera in these branches, as well as in other branches, which are probably endophytic bacteria, were also isolated from nodules. These data support siratro as a useful trap species to study the LNB biodiversity of diverse ecosystems in tropical soils. The fact that the highest diversity and nodulation were seen in managed systems such as agriculture and agroforestry suggests a high resilience of LNB communities to changes in land use after deforestation in a region where large forest areas are still preserved and can be a source of propagules.  相似文献

Community structure and diet of roving herbivorous reef fishes were analysed in 13 study sites around the five islands of the Abrolhos Archipelago, north-eastern Brazil (17°58' S; 38°42' W). This area has been part of the Abrolhos Marine National Park since 1983. Abundances and diets of fishes within the families Scaridae, Acanthuridae and Kyphosidae were compared among groups of sites differing in benthic community structure and exposure regime. The abundance of roving herbivorous fishes was higher in shallower sites than in deeper sites. At all study sites, observations of total herbivorous reef fish community structure revealed that 64% of fishes were acanthurids, 33% were scarids and only 4% were kyphosids. This pattern was predominant in all study sites. The majority of fishes examined in this study had the bulk of their diet based on both algae (35–90%) and detritus (35–65%). Among groups of algae, filamentous algae were the most commonly consumed as the majority of roving herbivorous fishes in Abrolhos feed as scrapers and excavators. The exceptions were the kyphosids, which had a diet consisting primarily of macroalgae (mainly Phaeophyta), and Acanthurus coeruleus that consumed mainly turf algae and very little detritus. The data collected indicate that roving herbivorous fishes in the Abrolhos Archipelago are more abundant at calm sites where detritus and delicate algae, their major food resources, are more abundant. The results presented here, along with underwater observations, suggest that the abundance and diet of these three fish families are associated with substratum cover.  相似文献
