Study of the kinetics of thermoinactivation of lysyl-tRNA-synthetase from the liver of x-irradiated rats

A study was made of the kinetics of thermoinactivation of lysyl-tRNA-synthetase isolated from rat liver at early times of radiation damage development after the effect of a minimum absolutely lethal X-radiation dose (0.21 C/kg). The thermostability of a dimer form of the enzyme was shown to be higher than that of a monomer. It was established that substrates had a stabilizing effect on the enzyme during thermoinactivation. On the basis of the data obtained from the studies in the kinetic properties of the enzyme and the thermoinactivation a conclusion is made that lysyl-tRNA-synthetase is stabilized during subunit aggregation. The thermostability of the enzyme was decreased by irradiation.     

Teratogenic and goitrogenic activity of propineb and propylenethiourea in the rat

The teratogenic and goitrogenic effects of Propineb, dithiocarbamate pesticide and Propylenthiourea (PLTU), its metabolite and degradation product have been studied. The aim of this study was to show the possible correlation between the two activities. Female Sprague-Dawley rats were treated with Propineb and PLTU starting from 6th to 16th day of pregnancy. The functional state of maternal and foetal thyroid, the toxicity of products versus dams and embryotoxic and teratogenic effects were examined. The observed goitrogenic effect may be compared to that reported in the previous studies of the authors, if considering time of sacrifice. In fact, the lesion quickly rises and as rapidly regresses when treatment is stopped. The foetal thyroid has not been affected by the product administered to the dams. PLTU showed a clear teratogenic activity at doses that did not show any maternal toxicity (45 and 90 mg/k).     

Changes in Epstein-Barr virus antibody titers associated with aging

Antibody titers to the Epstein-Barr virus (EBV), early antigen (EA) IgG, and virus capsid antigen (VCA) IgG and IgA, were measured in 44 geriatric subjects to determine if the depression in cellular immunity known to be associated with aging affects the expression of latent EBV. Similar assays were performed on plasma obtained from a young adult (medical student) population as a control group. We found that 89% of the geriatric samples were positive for EA IgG, and 83% of the plasma obtained from medical students were positive for EA IgG. One hundred percent of the geriatric subjects were positive for VCA IgG, and 87% of the medical students were positive for VCA IgG. Seven percent of the medical student blood samples were positive for VCA IgA; in contrast, 36% of the blood samples obtained from the geriatrics subjects were positive. Significant differences were also found in the geometric mean titers (GMT) of antibodies to EBV antigens; the GMT to EBV EA and VCA were significantly higher in the geriatric group. The data suggest that there may be some loss of control over latent EBV by the cellular immune response in geriatric individuals.     

Novel silylated steroids as aromatase inhibitors

Androst-4-en-3-one analogs incorporating a trimethylsilyl or a trimethylsilylmethyl group at C-1, C-2 or C-19 were prepared and evaluated as inhibitors of aromatase. Only 10-[1-hydroxy-2-(trimethylsilyl)ethyl]estr-4-ene-3,17-dione inhibited human placental aromatase. Enzyme kinetic analysis revealed competitive inhibition [apparent dissociation constant (Ki) of 562 +/- 12 nM] associated with marginal time-dependent inhibition.     

The effect of different hyperglucagonemic states on monooxygenase activities and isozymic pattern of cytochrome P-450 in mouse

The continuous infusion of a low dose of glucagon (35 g/kg/d, for 5 d) constitutes, in view of glucose-6-phosphatase and phosphoenolpyruvate carboxykinase activities , a reliable experimental model of hyperglucagonemia. By conjunction of monooxygenase assays and immunoquantitation of specific isozymes of cytochrome P-450, the actual inducing ability of glucagon has been shown and it might explain some of the modifications of the drug metabolizing system in diabetic mice. The isozymic pattern of cytochrome P-430 of liver microsomes from diabetic mice appears very different from that produced by classical inducers.

Retention of cadmium in organs of the rat after a single dose of labeled cadmium-3-phytate

Because of the low safety factor estimated for the normal content of Cd in human foods, it is important to establish the influence of food constituents such as phytate on the bioavailability of this toxic metal. We studied the retention of radioactive¹⁰⁹Cd administered to rats as a chloride or a phytate in a single dose by stomach tube. The animals were fed either a normal rat chow containing 0.29% of phytate or a low phytate diet containing less than 0.1% phytate. Highly elevated levels of¹⁰⁹Cd were found only in the animals that were supplied with¹⁰⁹Cd as a chloride and had been fed the low phytate diet. In the animals supplied with¹⁰⁹Cd as a phytate, which had also received the low phytate diet, the levels of¹⁰⁹Cd in the intestine were as high as those in the group mentioned before, but the retentions in all other tissues resembled those of the respective groups fed the normal chow. The findings indicate that phytate is responsible for a considerable decrease in the intestinal absorption of Cd. Furthermore, it appears to exert an influence on the kinetics of Cd retention in the intestine.     

NADPH-specific and NADH-specific xylose reduction is catalyzed by two separate enzymes in Pachysolen tannophilus

Summary Pachysolen tannophilus contains — in addition to an NADPH-linked xylose reductase — a separate NADH-linked one, in this respect differing from the yeast Pichia stipitis. Both enzyme proteins can conveniently be separated from each other by either ion exchange chromatography or chromatofocusing.     

Binding and degradation of proteoglycans by cultured arterial smooth muscle cells. II. Binding sites of proteoglycans on the cell surface

Exogenous proteoglycans stained for electron microscopy with colloidal gold and/or cuprolinic blue bind to the surface of cultured arterial smooth muscle cells at two different sites. (I) About 20% of the proteoglycans adsorbed to the cells from the culture medium interact as monomeric and multimeric proteoglycans with smooth or coated membrane areas. (II) The bulk of exogenous proteoglycans exhibits high affinity binding to cell membrane-associated 10 nm fibrils containing or being closely associated with fibronectin and to collagen. It is suggested that the self association of proteoglycans and their binding to the cell membrane and to cell surface-associated fibronectin and collagen are important for maintaining an appropriate micro-environment for the cultured cells.
