The behaviour of pectic substances in aqueous solution has been studied by the light-scattering technique. Pectic substances were found to be present as high molecular aggregates having strong, intermolecular interactions, and their behaviour depends upon the nature of the carbohydrate chain. Additives can cause dissociation of the polysaccharide aggregates, resulting, in some cases, in particles having similar weight-average ( w) and number-average ( n) molecular weights.  相似文献

Galacturonan, the main constituent of the backbone (core) of the comaruman macromolecule, a pectin from the marsh cinquefoil Comarum palustre L., was obtained on partial acid hydrolysis of the pectin. Using atomic force microscopy and methylation analysis of the galacturonan, the backbone of the comaruman macromolecule was shown to contain branches as side chains consisting of α-1,4-linked residues of D-galactopyranosyl uronic acid attached to the 2-and 3-positions of the galacturonic acid residues of the core, in addition to linear regions of α-1,4-D-galacturonan. An O-specific polysaccharide from the lipopolysaccharide Yersinia pseudotuberculosis 1A serovar has been isolated and characterized. This compound was shown to contain residues of paratose, 6-deoxy-D-manno-heptose, D-galactose and 2-amino-2-deoxy-D-glucose in equimolar ratios. Using methylation studies, partial acid hydrolysis and 13C NMR spectroscopy, the following structure was proposed for the repeating unit of the O-specific polysaccharide: (Formula: see text).  相似文献

Sources of carbohydrate nutrition such as sucrose, glucose, and galactose, with the exception of arabinose, were shown to influence positively callus growth and polysaccharide (pectin silenan and acidic arabinogalactan) biosynthesis. Galactose was found to cause a stimulatory effect on yield and productivity of arabinogalactan. Low concentrations of sucrose failed to support the cell growth and polysaccharide biosynthesis. Increasing sucrose concentrations led to biomass accumulation but failed to enhance efficiency of the substrate utilization. The optimal medium for the campion cell culture growth was found to be one containing 30 g/liter of sucrose or a mixture of sucrose with glucose (in 15 g/liter). Increasing sucrose concentrations in the medium from 30 to 100 g/liter failed to significantly influence the polysaccharide yields while the polysaccharide productivity per liter of the medium grew due to promotion of culture productivity in biomass. Variations of the carbon sources in the nutrient media were shown to influence insignificantly the biochemical characteristics of arabinogalactan and silenan while an increase in the sucrose concentration to 50-100 g/liter led to a diminution of the galacturonic acid content in silenan and to changes in contents of the neutral monosaccharide residues in silenan and arabinogalactan.  相似文献

Two fractions that included acid arabinogalactan and pectin were extracted from the callus culture of duckweed plants (Lemna minor L.) with water and ammonium oxalate. Residues of galactose and arabinose in the 2.0-2.5:1 ratio were the major constituents of acid arabinogalactan. The pectin fraction contained primarily residues of glucuronic acids, galactose, and arabinose. The percentage of arabinogalactan and pectin was similar. The yield of polysaccharide fractions did not depend on the method for their isolation. Extraction with water, treatment of the biomass with an aqueous solution of formalin and diluted hydrochloric acid, and extraction with an aqueous solution of ammonium oxalate allowed us to obtain the highest-purity pectin polysaccharide.  相似文献

A method for analysing 32P in small aqueous samples by measuring the ?erenkov radiation is described. Centering problems with small sample volumes are eliminated by placing the sample in a polystyrene tube in the scintillation vial. The detection efficiency is high, 54.5 ± 0.6% (2 SD) at a sample volume of 25 μl. The reproducibility is good and independent of the sample volume. The detection efficiency of 32P in polyacrylamide gel is shown to be as good as in water.  相似文献

A branched chain octose, 3,6-dideoxy-4-C-(L-glycero-1'-hydroxyethyl)-D-xylo- hexose, was isolated from the lipopolysaccharide of Yersinia frederiksenii, serovar O: 16.29 and identified as yersiniose A from Y. pseudotuberculosis, serovar VI. Mild hydrolysis of the lipopolysaccharide with acetic acid afforded a rhamnan. Structural features of the trisaccharide repeating unit were elucidated on the basic of 13C NMR spectral data, methylation studies and periodate oxidation. Polysaccharide fractions SAcI and SAcII were isolated from callus tissues of rowan tree stems. The SAcI fraction was shown to contain compounds belonging to the arabinogalactan II group. The SAcII fraction, called sorban, comprised pectic polysaccharides composing the protopectin complex of the cell wall callus. The SAcII fraction was found to contain a large amount of galacturonic acid residues and a set of neutral sugars characteristic of rhamnogalacturonan I. The composition and properties allowed a suggestion that the sorban backbone is mainly formed by 1,4-α-D-galactopyranosyluronic acid residues, while the neutral sugars are represented by 1,4-linked glucopyranose and xylopyranose residues, 1,5-linked arabinofuranose, 1,6-linked galactopyranose and mannofuranose residues as well as terminal glucopyranose and xylopyranose residues. The callus growth was shown to be associated with nearly a constant content of galacturonic acid and neutral sugar residues in sorban (fraction SAcII).  相似文献

Nestbox provision is a technique used to increase nest-site availability for secondary cavity-nesting birds. However, little is known about the demographic consequences of nestbox provision in different habitat types. To assess how nestbox provision affects the density of hole-nesting birds simultaneously in two contrasting habitats, we compared the breeding density of Great Tits along transects without nestboxes with that in transects where nestboxes were provided. Although the initial density of breeders was considerably higher in the deciduous habitat than in the coniferous habitat, provision of nestboxes increased density by a similar number of additional pairs in each habitat type. Thus, the provision of nestboxes in managed coniferous forests may be as effective in increasing the breeding opportunities of cavity nesters as in deciduous stands. Moreover, previous research showed that pairs in deciduous habitat with nestboxes have consistently lower breeding success than those in coniferous habitat with nestboxes. It is possible that the addition of nestboxes in the preferred habitat increased density to such an extent that density-dependent effects became apparent.  相似文献
