Summary Ultrastructural changes during zoospore formation and aggregation into motile, aggregating zoospores were examined in the colonial green algaPediastrum tetras. Developing zoospores are characterized by irregularly shaped nuclei, presence of peripheral networks of rough endoplasmic reticulum, chloroplasts with tightly apposed thylakoids and dictyosome cisternae which are compressed and reduced in size. A single membrane bound organelle with a fine granular matrix of moderate electron density of diameter ranging from 0.2 m to 0.6 m and associated with chloroplasts, mitochondria and endoplasmic reticulum was found only in adult cells. Although this organelle has the morphology of a microbody, it did not stain with 3,3-diaminobenzidine (DAB) at pH 9.6 or pH 7.6, whereas mitochondrial membranes stained. No DAB staining was observed along the cell wall or the plasma membrane of zoospores, or associated with endoplasmic reticulum, plastid membranes or dictyosomes.  相似文献

This paper presents XBAM (the Extended Branch-Arrow Model), a new model of the development of the retino-tectal topographic mapping as observed in frog, toad, and goldfish visual systems. The updating process employed by XBAM is distributed in nature and depends upon interactions between branches of retinal fibers, the branches and the boundaries of the tectum and grafts, and the branches and the tectal surface. Results of computer simulation of the model are related to experimental data obtained from tectal and retinal graft and lesion studies, and comparisons are also made with other models.  相似文献

Customary procedures used to determine chromosomal inheritance do not disclose many of the chromosomal polymorphisms known to be present, resulting in uninformative families. The sequential printing of individual chromosomes presented here is a technique that has increased the number of informative families in our studies. This technique has revealed previously unseen heritable chromosome differences and allowed the designation of parental origin.  相似文献

Nesting birds must provide a thermal environment sufficient for egg development while also meeting self‐maintenance needs. Many birds, particularly those with uniparental incubation, achieve this balance through periodic incubation recesses, during which foraging and other self‐maintenance activities can occur. However, incubating birds may experience disturbances such as predator or human activity which interrupt natural incubation patterns by compelling them to leave the nest. We characterized incubating mallard Anas platyrhynchos and gadwall Mareca strepera hens’ responses when flushed by predators and investigators in Suisun Marsh, California, USA. Diurnal incubation recesses initiated by investigators approaching nests were 63% longer than natural diurnal incubation recesses initiated by the hen (geometric mean: 226.77 min versus 142.04 min). Nocturnal incubation recesses, many of which were likely the result of predators flushing hens, were of similar duration regardless of whether the nest was partially depredated during the event (115.33 [101.01;131.68] minutes) or not (119.62 [111.96;127.82] minutes), yet were 16% shorter than natural diurnal incubation recesses. Hens moved further from the nest during natural diurnal recesses or investigator‐initiated recesses than during nocturnal recesses, and the proportion of hen locations recorded in wetland versus upland habitat during recesses varied with recess type (model‐predicted means: natural diurnal recess 0.77; investigator‐initiated recess 0.82; nocturnal recess 0.31). Hens were more likely to take a natural recess following an investigator‐initiated recess earlier that same day than following a natural recess earlier that same day, and natural recesses that followed an investigator‐initiated recess were longer than natural recesses that followed an earlier natural recess, suggesting that hens may not fulfill all of their physiological needs during investigator‐initiated recesses. We found no evidence that the duration of investigator‐initiated recesses was influenced by repeated visits to the nest, whether by predators or by investigators, and trapping and handling the hen did not affect investigator‐initiated recess duration unless the hen was also fitted with a backpack‐harness style GPS–GSM transmitter at the time of capture. Hens that were captured and fitted with GPS–GSM transmitters took recesses that were 26% longer than recesses during which a hen was captured but a GPS–GSM transmitter was not attached. Incubation interruptions had measurable but limited and specific effects on hen behavior.  相似文献

Treatment of patients diagnosed as schizophrenic with antipsychotic drugs (neuroleptics) is known to cause occasional unexplained depletion of white blood cells, especially neutrophil granulocytes. It has been known for many years that neuroleptics can interfere with the mitochondrial respiratory chain in vitro. Because there has been a growing interest recently in mitochondrial targeting of drugs, and since a quantitative structure-activity relationship (QSAR) model that predicts mitochondrial accumulation of neuroleptics has been published, we investigated the effects of neuroleptics on white blood cell mitochondria. Venous blood samples were collected from both patients undergoing treatment with neuroleptics and healthy volunteers. The samples were processed for transmission electron microscopy. The resulting images of white blood cells were analyzed using stereology to compare quantitatively mitochondrial morphology in the patient and control groups. We found that in patients, but not in controls, there was swelling of mitochondria and fragmentation of the mitochondrial cristae. There also were fewer mitochondria in patients than in controls, although due to the swelling of the organelles, the volume density of mitochondria in the two groups was not significantly different. Such changes are typical of a toxic insult. Consequently, it seems plausible that, since schizophrenia is not a disease considered to affect white blood cells per se, these changes probably are due to the medication.  相似文献

Ambient air temperatures (T(a)) of <6 degrees C or >29 degrees C have been shown to induce large changes in arterial blood pressure and heart rate in homeotherms. The present study was designed to investigate whether small incremental changes in T(a), such as those found in typical laboratory settings, would have an impact on blood pressure and other cardiovascular parameters in mice and rats. We predicted that small decreases in T(a) would impact the cardiovascular parameters of mice more than rats due to the increased thermogenic demands resulting from a greater surface area-to-volume ratio in mice relative to rats. Cardiovascular parameters were measured with radiotelemetry in mice and rats that were housed in temperature-controlled environments. The animals were exposed to different T(a) every 72 h, beginning at 30 degrees C and incrementally decreasing by 4 degrees C at each time interval to 18 degrees C and then incrementally increasing back up to 30 degrees C. As T(a) decreased, mean blood pressure, heart rate, and pulse pressure increased significantly for both mice (1.6 mmHg/ degrees C, 14.4 beats.min(-1). degrees C(-1), and 0.8 mmHg/ degrees C, respectively) and rats (1.2 mmHg/ degrees C, 8.1 beats.min(-1). degrees C(-1), and 0.8 mmHg/ degrees C, respectively). Thus small changes in T(a) significantly impact the cardiovascular parameters of both rats and mice, with mice demonstrating a greater sensitivity to these T(a) changes.  相似文献

We used mitochondrial/nuclear gene sequence analyses to determine the historical relationships of the endemic species of Todus (Aves: Todidae) from the Caribbean. We collected 1920-bp of nucleotide sequence data from the mitochondrial genes cytochrome b, ATPase 6, ATPase 8, and 591-bp of the single-copy nuclear gene c-mos for all Todus species and representatives of their outgroup taxa (Hylomanes, Barypthengus, Chloroceryle, Ceryle, and Galbula) to reconstruct the evolutionary history (via parsimony and maximum likelihood) of the five Todus species. The substitution rates among the mitochondrial genes were found to be much higher than the substitution rate for the c-mos gene, consequently resulting in higher substitutional saturation for the mitochondrial genes. When we applied weighting schemes to account for the variance in substitutional heterogeneity among the genes then parsimony and likelihood analyses both demonstrate that the genus Todus is monophyletic and closer to the Hylomanes and Barypthengus genera than the Chloroceryle and Ceryle genera. The mitochondrial-gene trees and nuclear-gene trees both show similar results, thus providing support for the relationships among the taxa from loci within two independently evolving genomes. The nuclear gene c-mos was found, therefore, to be a viable nuclear gene candidate for resolving intermediate and deep divergences.  相似文献

Transplantation of spermatogonial stem cells into syngeneic or immunosuppressed recipient mice or rats can result in donor-derived spermatogenesis and fertility. Recently, this approach has been employed to introduce a transgene into the male germline. Germ-cell transplantation in species other than laboratory rodents, if successful, holds great promise as an alternative to the inefficient methods currently available to generate transgenic farm animals that can produce therapeutic proteins in their milk or provide organs for transplantation to humans. To explore whether germ-cell transplantation could result in donor-derived spermatogenesis and fertility in immunocompetent recipient goats, testis cells were transplanted from transgenic donor goats carrying a human alpha-1 antitrypsin expression construct to the testes of sexually immature wild-type recipient goats. After puberty, sperm carrying the donor-derived transgene were detected in the ejaculates of two out of five recipients. Mating of one recipient resulted in 15 offspring, one of which was transgenic for the donor-derived transgene. This is the first report of donor cell-derived sperm production and transmission of the donor haplotype to the next generation after germ-cell transplantation in a nonrodent species. Furthermore, these results indicate that successful germ-cell transplantation is feasible between immunocompetent, unrelated animals. In the future, transplantation of genetically modified germ cells may provide a more efficient alternative for production of transgenic domestic animals.  相似文献

G protein-coupled receptors (GPCRs) can form homodimers/oligomers and/or heterodimers/oligomers. The mechanisms used to form specific GPCR oligomers are poorly understood because the domains that mediate such interactions and the step(s) in the secretory pathway where oligomerization occurs have not been well characterized. Here we have used subcellular fractionation and fluorescence resonance energy transfer (FRET) experiments to show that oligomerization of a GPCR (alpha-factor receptor; STE2 gene product) of the yeast Saccharomyces cerevisiae occurs in the endoplasmic reticulum. To identify domains of this receptor that mediate oligomerization, we used FRET and endocytosis assays of oligomerization in vivo to analyze receptor deletion mutants. A mutant lacking the N-terminal extracellular domain and transmembrane (TM) domain 1 was expressed at the cell surface but did not self-associate. In contrast, a receptor fragment containing only the N-terminal extracellular domain and TM1 could self-associate and heterodimerize with wild type receptors. Analysis of other mutants suggested that oligomerization is facilitated by the N-terminal extracellular domain and TM2. Therefore, the N-terminal extracellular domain, TM1, and TM2 appear to stabilize alpha-factor receptor oligomers. These domains may form an interface in contact or domain-swapped oligomers. Similar domains may mediate dimerization of certain mammalian GPCRs.  相似文献

Arctic ground squirrels (Spermophilus parryii) overwinter in hibernaculum conditions that are substantially below freezing. During torpor, captive arctic ground squirrels displayed ambient temperature (T(a))-dependent patterns of core body temperature (T(b)), metabolic rate (TMR), and metabolic fuel use, as determined by respiratory quotient (RQ). At T(a) 0 to -16 degrees C, T(b) remained relatively constant, and TMR rose proportionally with the expanding gradient between T(b) and T(a), increasing >15-fold from a minimum of 0.0115 +/- 0.0012 ml O(2). g(-1). h(-1). At T(a) 0-20 degrees C, T(b) increased with T(a); however, TMR did not change significantly from T(b) 0 to 12 degrees C, indicating temperature-independent inhibition of metabolic rate. The overall change in TMR from T(b) 4 to 20 degrees equates to a Q(10) of 2.4, but within this range of T(b), Q(10) changed from 1.0 to 14.1. During steady-state torpor at T(a) 4 and 8 degrees C, RQ averaged 0.70 +/- 0.013, indicating exclusive lipid catabolism. At T(a) -16 and 20 degrees C, RQ increased significantly to >0.85, consistent with recruitment of nonlipid fuels. RQ was negatively correlated with maximum torpor bout length. For T(a) values <0 degrees C, this relationship supports the hypothesis that availability of nonlipid metabolic fuels limits torpor duration in hibernating mammals; for T(a) values >0 degrees C, hypotheses linked to body temperature are supported. Because anterior body temperatures differ from core, overall, the duration torpor can be extended in hibernating mammals may be dependent on brain temperature.  相似文献
