Active preparations of Na+,K(+)-ATPase containing three types of catalytic isoforms were isolated from the bovine brain to study the structure and function of the sodium pump. Na+,K(+)-ATPase from the brain grey matter was found to have a biphasic kinetics with respect to ouabain inhibition and to consist of a set of isozymes with subunit composition of alpha 1 beta 1, alpha 2 beta m and alpha 3 beta m (where m = 1 and/or 2). The alpha 1 beta 1 form clearly dominated. For the first time, glycosylation of the beta 1-subunit of the alpha 1 beta 1-type isozymes isolated from the kidney and brain was shown to be different. Na+,K(+)-ATPase from the brain stem and axolemma consisted mainly of a mixture of alpha 2 beta 1 and alpha 3 beta 1 isozymes having identical ouabain inhibition constants. In epithelial and arterial smooth muscle cells, where the plasma membrane is divided into functionally and biochemically distinct domains, the polarized distribution of Na+,K(+)-ATPase is maintained through interactions with the membrane cytoskeleton proteins ankyrin and spectrin (Nelson and Hammerton, 1989; Lee et al., 1996). We were the first to show the presence of the cytoskeleton protein tubulin (beta 5-isoform) and glyceraldehyde-3-phosphate dehydrogenase in a high-molecular-weight complex with Na+,K(+)-ATPase in brain stem neuron cells containing alpha 2 beta 1 and alpha 3 beta 1 isozymes. Consequently, the influence of not only subunit composition, but also of glycan and cytoskeleton structures and other plasma membrane-associated proteins on the functional properties of Na+,K(+)-ATPase isozymes is evident.  相似文献

Antiamoebin I (Aam-I) is a membrane-active peptaibol antibiotic isolated from fungal species belonging to the genera Cephalosporium, Emericellopsis, Gliocladium, and Stilbella. Antiamoebin I has the amino acid sequence: Ac-Phe(1)-Aib-Aib-Aib-Iva-Gly-Leu-Aib(8)-Aib-Hyp-Gln-Iva-Hyp-Aib-Pro-Phl(16). By using the uniformly (13)C,(15)N-labeled sample of Aam-I, the set of conformationally dependent J couplings and (3h)J(NC) couplings through H-bonds were measured. Analysis of these data along with the data on magnetic nonequivalence of the (13)C(beta) nuclei (Deltadelta((13)C(beta))) in Aib and Iva residues allowed us to draw the univocal conclusion that the N-terminal part (Phe(1)-Gly(6)) of Aam-I in MeOH solution is in fast exchange between the right-handed and left-handed 3(10)-helical conformations, with an approximately equal population of both states. An additional conformational exchange process was found at the Aib(8) residue. The (15)N-NMR-relaxation and CD-spectroscopy measurements confirmed these findings. Molecular modeling and Monte Carlo simulations revealed that both exchange processes are correlated and coupled with significant hinge-bending motions around the Aib(8) residue. Our results explain relatively low activity of Aam-I with respect to other 15-amino acid residue peptaibols (for example, zervamicin) in functional and biological tests. The high dynamic 'propensity' possibly prevents both initial binding of the antiamoebin to the membrane and subsequent formation of stable ionic channels according to the barrel-stave mechanism.  相似文献

The blood of 6 newborn boys with haemolytic disease of newborns and of 3 healthy newborn boys was examined. In all 949 metaphase plates were analysed. In all the investigated plates the karyotype was masculine, 46,XY [correction of 46HY]. The mitotic index is lowered considerably in sick children that suggests inhibition of their cellular immunity. In these conditions, the K and L lymphocytes of donor blood, introduced in substituting blood transfusion, can interact with the sensitized erythrocytes in accordance with the type of reaction "the transplant against the host" causing the increase in the level of bilirubin after the operation. It is expedient to study the possibility of realization of substituting blood transfusion using the leucocyte-free blood.  相似文献

Rhizobium leguminosarum bv. trifolii strain TA1 is an aerobic, motile, Gram-negative, non-spore-forming rod that is an effective nitrogen fixing microsymbiont on the perennial clovers originating from Europe and the Mediterranean basin. TA1 however is ineffective with many annual and perennial clovers originating from Africa and America. Here we describe the features of R. leguminosarum bv. trifolii strain TA1, together with genome sequence information and annotation. The 8,618,824 bp high-quality-draft genome is arranged in a 6 scaffold of 32 contigs, contains 8,493 protein-coding genes and 83 RNA-only encoding genes, and is one of 20 rhizobial genomes sequenced as part of the DOE Joint Genome Institute 2010 Community Sequencing Program.  相似文献

Conditions of limited proteolysis of the protease Lon from Escherichia coli that provided the formation of fragments approximately corresponding to the enzyme domains were found for studying the domain functioning. A method of isolation of the domains was developed, and their functional characteristics were compared. The isolated proteolytic domain (LonP fragment) of the enzyme was shown to exhibit both peptidase and proteolytic activities; however, it cleaved large protein substrates at a significantly lower rate than the full-size protease Lon. On the other hand, the LonAP fragment, containing both the ATPase and the proteolytic domains, retained almost all of the enzymatic properties of the full-size protein. Both LonP and LonAP predominantly form dimers unlike the native protease Lon functioning as a tetramer. These results suggest that the N-terminal domain of protease Lon plays a considerable role in the process of the enzyme oligomerization.  相似文献

Large geographic range, wide habitat specificity and broad range of prey (including a number of pests of protected crops) suggest that a predatory ladybird Cheilomenes propinqua can be considered as potential agent for biological control in greenhouses. We investigated the influence of day length (10, 12 and 14 hr), temperature (20 and 24°C) and diet (the green peach aphid Myzus persicae and eggs of the grain moth Sitotroga cerealella) on the rate of maturation, fecundity and induction of reproductive diapause in C. propinqua females of a laboratory population originated from Alexandria, Egypt. The proportion of diapausing females (i.e. those with poorly developed ovaries and well-developed fat body) varied from 5% to 70% being higher at short day, low temperature and feeding on the grain moth eggs. This diapause, however, was not very stable: more than 20% of females kept under diapause-inducing conditions started to lay eggs during 110 days, although their pre-oviposition period was about 5 times longer than that of females which matured at the same temperature but at the long day and on the natural diet. Although not very stable, reproductive diapause significantly increased survival of starving females. Such a short-term reproductive diapause can be considered as an adaptation to mild and short-term subtropical winter. The results of our study suggest that C. propinqua mass rearing will be more intensive at the combination of high temperature, natural food (aphids) and long day (14 hr), whereas individuals intended for long-term storage should be reared under moderate temperature (20°C) and short-day (10 hr) conditions and should be fed on factitious food (the grain moth eggs).  相似文献
