We studied sexual and habitat-specific variation in herbivores' host exploitation patterns and tested for a hypothesis on sex-specific adaptation in replicated landscapes. The hypothesis has not to our knowledge been previously tested either in a marine environment or for the host exploitation traits of an herbivore. The hypothesis states, first, that populations may show different adaptations due to spatially differing selective environments, and second, that males and females that differ ecologically (e.g. in reproductive behavior, in host use patterns) will respond to selective environments in distinct ways. We investigated possible differences in the host exploitation patterns of the marine generalist crustacean grazer Idotea baltica between the sexes or among populations originating from different habitats: plant assemblages dominated by either a brown alga or an angiosperm species. We determined the preferences for both the structure and nutritive quality of five common host species and compared these to the actual performance on the hosts. We found that performance on the hosts differed between the sexes and differently so in the two habitat types, supporting the hypothesis of sex-specific adaptation between distinct selective environments. Habitat-specific preference for the structural host further supported ecological divergence between habitat types. The different approaches for testing host exploitation resulted in a very different rank order of the hosts, indicating the complexity of the factors involved. The herbivore differentiates for the chemical quality of the hosts, but this alone does not explain host exploitation; rather it supports the theory of enemy-free space as a determinant of host choice. We also discuss the chemical characteristics of the host associated with the exploitation patterns found.  相似文献

The Ret receptor tyrosine kinase is the common signaling receptor for the glial cell line-derived neurotrophic factor (GDNF) family ligands. The Met918Thr mutation leads to constitutive activation of Ret and is responsible for dominantly inherited cancer syndrome MEN2B. Previously, we found that the mice carrying the mutation (MEN2B mice) have profoundly increased tissue dopamine (DA) concentrations in the striatum as well as increased striatal levels of tyrosine hydroxylase (TH) and dopamine transporter. The aim of this study was to characterize the striatal dopaminergic neurotransmission in MEN2B mice and to clarify the mechanisms by which they compensate their over-production of DA. We found that tyrosine hydroxylase activity and DA synthesis are increased in MEN2B mice. Augmented effects of α-methyl-para-tyrosine (αMT, an inhibitor of TH) and tetrabenazine (VMAT2 blocker) on DA levels suggest that also storage of DA is increased in MEN2B mice. There was no difference in the basal extracellular DA concentrations or potassium-evoked DA release between the genotypes. The effects of cocaine and haloperidol were also similar between the genotypes as assessed by in vivo microdialysis. However, with in vivo voltammetry we found increase in stimulated DA release in MEN2B mice and detailed analysis of DA overflow showed that uptake of DA was also enhanced in MEN2B mice. Thus, our data show that enhanced synthesis of DA leading to increased storage and releasable pools in pre-synaptic terminals in MEN2B mice apparently also leads to increased DA release, which in turn is compensated by higher dopamine transporter activity.  相似文献

The macrophage scavenger receptor macrophage receptor with a collagenous structure (MARCO) is expressed in mice by the marginal zone macrophages of the spleen and by macrophages of the medullary cords of lymph nodes, as well as the peritoneal macrophages. MARCO is a relative of scavenger receptor A (SR-A), the more widely expressed prototypic member of the scavenger receptor family. In the present study, we found that genetic ablation of MARCO leads to changes in the organization of the splenic marginal zone, and causes a significant reduction in the size of the resident peritoneal macrophage population, possibly due to changes in adhesion and migration capacity. In mice lacking both MARCO and SR-A these effects are even more apparent. During ontogeny, the appearance and organization of the MARCO-expressing cells in the spleen precedes the appearance of other receptors on macrophages in the marginal zone, such as SIGNR1 and Siglec-1. In the absence of MARCO, a clear delay in the organization of the marginal zone was observed. Similar findings were seen when the reappearance of the various subsets from precursors was studied after depleting macrophages from the adult spleen by a liposome treatment. When challenged with a pneumococcal polysaccharide vaccine, a T-independent type 2 Ag for which an intact marginal zone is crucial, the knockout mice exhibited a clearly impaired response. These findings suggest that both MARCO and SR-A, in addition to being important scavenger receptors, could be involved in the positioning and differentiation of macrophages, possibly through interaction with endogenous ligands.  相似文献

Background

Recent studies have suggested the therapeutic potential of vascular endothelial growth factor (VEGF) gene therapy in ischemic skeletal muscle. However, only limited information is available about the effects of VEGF gene therapy in different regions of ischemic limbs, effects of control adenoviruses, and biodistribution of the transgenes after intramuscular (i.m.) administration. Here we studied angiogenesis and side effects of adenovirus‐mediated VEGF and β‐galactosidase (LacZ) gene transfers in ischemic rabbit hindlimbs.

Methods and results

Ten days after induction of ischemia, rabbits were treated with i.m. injections of saline, LacZ adenovirus (AdLacZ; 2×1010 pfu) or adenovirus encoding mouse VEGF164 (AdVEGF; 2×1010 pfu). In rabbits treated with AdVEGF an increase in serum VEGF164 levels was detected by ELISA three and seven days after the gene transfer. 30 days after the gene transfer a positive effect on capillary density was observed in the thigh region both in rabbits treated with AdVEGF and AdLacZ compared with animals that received saline. On the other hand, AdVEGF and AdLacZ gene transfers had no effect on the capillary density in the calf region on day 30. A positive correlation between the capillary density and the number of collateral arteries was observed in the thigh. Hindlimb and testis edema and excess non‐physiological growth of capillaries were detected as adverse effects of the AdVEGF gene therapy. Biodistribution analysis showed that the transgene was present not only in the target muscle, but also in ectopic tissues seven days after i.m. gene transfer.

Conclusions

The results suggest that a high dose of adenoviral vector encoding either AdVEGF or AdLacZ induces angiogenesis in the rabbit hindlimb ischemia model; i.m. injection of adenovirus leads to the transfection of ectopic organs; and AdVEGF gene transfer induces edema in ischemic skeletal muscle. Spatial recognition cues used in site fidelity in the ant Formica uralensis Ruzsky were studied using outdoor and laboratory arenas. Ant workers visiting symmetrically spaced feeders were colour-marked corresponding to the initial feeder visited during sampling. The effect of manipulating environmental cues on the mean 'spatial specialization' of the population was measured. Site recognition appears to be based on visual landmark/canopy cues. However, ants maintained some fidelity when shielded from these cues, suggesting the involvement of additional cues. When ridding our experimental device of olfactory deposits and shielding visual cues, site fidelity was lost. Idiothetic and/or geomagnetic cues are thought to provide spatial references to visual or olfactory landmarks. Altering nest position relative to the arena and changing the geomagnetic field within the arena in our study, however, did nothing to the site fidelity of visually deprived and non-deprived foragers.
We conclude that site fidelity is developed in a visually structured environment but supplemented by an olfactory backup system that is probably based on discrete home range markings rather than radial odour trails. We demonstrate furthermore that the visual component involved in site location can be stored in the memory of individual F. uralensis foragers during a 6-month hibernation period.  相似文献

Inbreeding depression is a major selective force favoring outcrossing in flowering plants. However, some self-fertilization should weaken the harmful effects of inbreeding by exposing deleterious alleles to selection. This study examines the maintenance of inbreeding depression in the predominantly outcrossing species Pinus sylvestris L. (Scots pine). Open-pollinated and self-fertilized progeny of 23 maternal trees, originating from a natural stand in southern Finland, were grown at two sites. We observed significant inbreeding depression in two of the four life stages measured. Inbreeding depression was largest for seed maturation (δ = 0.74), where seedset in open-pollinated strobili (70.9%) was about four times higher than in selfed strobili (18.3%). Inbreeding depression in postgermination survival (upto an age of 23 years) was also high (δ = 0.62–0.75). No significant differences in height (δ = 0.05) or flowering (δ = 0.14) of the trees after 23 years were observed. Cumulative inbreeding depression was high (δ = 0.90–0.94) and differed significantly among maternal families (range 0.45–1.00). The magnitude of inbreeding depression among the 23 maternal parents was not significantly correlated between early (seed maturation) and later (postgermination survival) life stages, suggesting that its genetic basis varies across the life cycle. Size differences among the progeny types diminished in time due to nonrandom size-specific mortality, causing a decrease in the inbreeding depression estimates for height over time. Our results indicate that Scots pine exhibits high levels of inbreeding depression during both early and later stages of the life cycle. It is argued that self-fertilization in Scots pine is inefficient in purging the genetic load caused by highly deleterious mutations because of the nearly complete loss of selfed individuals over time. The germination and growth of pollen grains of Nicotiana tabacum and N. alata with the anti-microtubule drug oryzalin retarded significantly the movement of the vegetative nucleus (VN) and the generative cell (GC) from the grain to the tube apex but had no effect on pollen tube elongation. In N. tabacum, only 11% and 48% of the pollen tubes treated with oryzalin for 6 h and 12 h, respectively, had the VN and GC in the tube mainly in its middle part. In corresponding control materials, 79% and 99% of pollen tubes contained the VN and GC close to the apex. Indirect immunofluorescence microscopy and related studies of the tubes grown in the presence of oryzalin revealed complete absence of microtubules (MTs) but apparently intact microfilaments (MFs). These results suggested that the movement of VN and GC from the grain into the tube is possible when no MTs but only MFs are present, but the movement is then slow. In control tubes, the parallel orientation of MT bundles and extensions of VN were interpreted to represent the structural organization needed for the MT-dependent movement of VN.  相似文献

Summary To study the regulation of the development of myoid cells, seminiferous tubules of adult and prepubertal rats were grown in organ culture under the influence of testosterone, HCG and cyproterone acetate. Contractility, EM-structure and histochemical activities of alkaline phosphatase and ATPase of the myoid cells were studied in the adult rat tubules at one week intervals up to 5 weeks in culture, in the prepubertal rat tubules at the age of 15 days and after 15 and 21 days in culture. As in vivo, contractions appear in cultured tubules of prepubertal rats at the age of 15 days. Testosterone and HCG increase the percentage of contractile tubules and the number of filaments of the myoid cells. Cyproterone acetate inhibits both functional and structural development and tends to decrease the enzyme activities. In the cultured adult rat tubules cyproterone acetate causes disappearance of contractility within one week, while contractions normally are found for 3 weeks. Testosterone and HCG have no notable effects on the contractility of adult rat tubules, but they lengthen the persistence of alkaline phosphatase activity. It is concluded that the maturation and also the functioning of the myoid cells are subject to androgenic regulation.  相似文献

Production of ligninolytic enzymes by three strains of the white rot fungus Phlebia tremellosa (syn. Merulius tremellosus) was studied in bioreactor cultivation under nitrogen-limiting conditions. The Mn(II) concentration of the growth medium strongly affected the secretion patterns of lignin peroxidase and laccase. Two major lignin peroxidase isoenzymes were expressed in all strains. In addition, laccase and glyoxal oxidase were purified and characterized in one strain of P. tremellosa. In contrast, manganese peroxidase was not found in fast protein liquid chromatography profiles of extracellular proteins under either low (2.4 μM) or elevated (24 and 120 μM) Mn(II) concentrations. However, H2O2- and Mn-dependent phenol red-oxidizing activity was detected in cultures supplemented with higher Mn(II) levels. Mineralization rates of 14C-ring-labelled synthetic lignin (i.e., dehydrogenation polymerizate) by all strains under a low basal Mn(II) level were similar to those obtained for Phanerochaete chrysosporium and Phlebia radiata. A high manganese concentration repressed the evolution of 14CO2 even when a chelating agent, sodium malonate, was included in the medium.  相似文献
