Contemporary techno-scientific and medical developments are restructuring social interactions and the very processes by which individual subjectivity is formed. This essay elaborates on the experiential and ethical impact of such transformations from the perspective of people who, in ordinary and unexpected ways, act science and technology out. We carried out ethnographic research in an HIV/AIDS Testing and Counseling Center (CTA) in northeastern Brazil, combining participant observation with epidemiological analyses and clinical survey. We found a high demand for free testing by low-risk clients, largely working and middle class, experiencing anxiety and complaining of AIDS-like symptoms. Most of the clients were sero-negative and many returned for a second and third testing. We understand this to be a new techno-cultural phenomenon and call it imaginary AIDS. Throughout this essay, we describe CTA's routine practices, place these practices in historical, political, economic and cross-cultural perspective, and analyze the subjective data we collected from the clients of our pilot study. We explore how clinical epidemiological expertise and HIV testing technology are integrated into new forms of bio-politics aimed at specific marketable and disease-free populations, and on the affective absorption of bio-technical truth and the engendering of a technoneurosis in this testing center. 相似文献

Theoretical considerations have shown that the five possible overlapping
reading-frame configurations differ significantly in their coding
flexibility and thus in their information content (Siegel and Fitch 1980;
Smith and Waterman 1980). Contrary to expectation, the overlapping frame
configuration allowing the greatest coding flexibility is rarely seen,
whereas one of the most constraining is common. We point out here that this
overlapping reading-frame paradox and an observed but unexplained
preference in coding regions for a pyrimidine-purine at codon boundaries
(Shepherd 1981; Jones and Kafatos 1982; Smith et al. 1983) are intimately
linked. The codon boundary preference, which may be related to translation
efficiency or accuracy, places constraints on the evolution of overlapping
coding regions. These considerations may help identify actual coding
regions in DNA sequences. We have analyzed five sequenced (enteric)
bacterial insertion sequences for codon boundary incidences and
reading-frame configurations and find that they are consistent with these
proposed constraints.

An anchored restriction-mapping approach applied to the genetic analysis of the Anopheles gambiae malaria vector complex 1 总被引:1,自引:0,他引:1
We introduce here a simple approach for rapidly determining restriction
maps for a number of regions of a genome; this involves "anchoring" a map
with a rare restriction site (in this case the seldom-cutting EagI)
followed by partial digestion of a frequent-cutting enzyme (e.g., Sau 3A).
We applied this technology to five species of the Anopheles gambiae
complex. In a single Southern blot we obtained about a 15-kb restriction
map each for the mtDNA, rRNA gene, and a scnDNA region for each of five
species. Phylogenetic analyses of these regions yield trees at odds with
the more traditional chromosome inversion-based trees. The value of the
approach for systematic purposes is the ease with which several large,
independent regions of the genome can be quickly assayed for molecular
variation.

In this study, we examined the effects of angiotensin II (AngII) in a genetic in vitro PD model produced by alpha-synuclein (alpha-syn) overexpression in the human neuroglioma H4 cell line. We observed a maximal decrease in alpha-syn-induced toxicity of 85% and reduction in inclusion formation by 19% when cultures were treated with AngII in the presence of the angiotensin type 1 (AT1) receptor antagonist losartan and AT2 receptor antagonist PD123319. When compared to AngII, the AT4 receptor agonist AngIV was moderately effective in protecting H4 cells against alpha-syn toxicity and did not significantly reduce inclusion formation. Here we show that AngII is protective against genetic, as well as neurotoxic models of PD. These data support the view that agents acting on the renin-angiotensin-system (RAS) may be useful in the prevention and/or treatment of Parkinson's disease. 相似文献

The aggregation, deposition, and dysfunction of alpha-synuclein (aSyn) are common events in neurodegenerative disorders known as synucleinopathies. These include Parkinson''s disease, dementia with Lewy bodies, and multiple system atrophy. A growing body of knowledge on the biology of aSyn is emerging and enabling novel hypotheses to be tested. In particular, the hypothesis that aSyn is secreted from neurons, thus contributing to the spreading of pathology not only in the brain but also in other organs, is gaining momentum. Nevertheless, the precise mechanism(s) of secretion, as well as the consequences of extracellular aSyn species for neighboring cells are still unclear. Here, we review the current literature and integrate existing data in order to propose possible mechanisms of secretion, cell dysfunction, and death. Ultimately, the complete understanding of these processes might open novel avenues for the development of new therapeutic strategies. 相似文献
