本文用插线法制作局灶性脑缺血/再灌损伤模型,利用激光共聚焦扫描显微镜观察活体脑片细胞内Ca2+的分布及动态变化,结果表明:(1)缺血/再灌时间不同,梗塞面积不同,缺血4小时梗塞面积占同侧半球的16.3%,缺血4小时再灌20小时梗塞面积增加到25.9%,缺血24小时梗塞面积占同侧半球的60.4%。(2)本文首次观察到在缺血4小时纹状体区域的Ca2+变化明显高于皮层,并且再灌后皮层及纹状体区域Ca2+的含量明显增加  相似文献

In order to investigate the mitochondrial genome of Panthera tigris amoyensis, two South China tigers (P25 and P27) were analyzed following 15 cymt-specific primer sets. The entire mtDNA sequence was found to be 16,957 bp and 17,001 bp long for P25 and P27 respectively, and this difference in length between P25 and P27 occurred in the number of tandem repeats in the RS-3 segment of the control region. The structural characteristics of complete P. t. amoyensis mitochondrial genomes were also highly similar to those of P. uncia. Additionally, the rate of point mutation was only 0.3% and a total of 59 variable sites between P25 and P27 were found. Out of the 59 variable sites, 6 were located in 6 different tRNA genes, 6 in the 2 rRNA genes, 7 in non-coding regions (one located between tRNA-Asn and tRNA-Tyr and six in the D-loop), and 40 in 10 protein-coding genes. COI held the largest amount of variable sites (9 sites) and Cytb contained the highest variable rate (0.7%) in the complete sequences. Moreover, out of the 40 variable sites located in 10 protein-coding genes, 12 sites were nonsynonymous.  相似文献

Lung cancer is a serious disease that threatens an affected individual's life. Its pathogenesis has not yet to be fully described, thereby impeding the development of effective treatments and preventive measures. “Cancer driver” theory considers that tumor initiation can be associated with a number of specific mutations in genes called cancer driver genes. Four omics levels, namely, (1) methylation, (2) microRNA, (3) mutation, and (4) mRNA levels, are utilized to cluster cancer driver genes. In this study, the known dysfunctional genes of these four levels were used to identify novel driver genes of lung adenocarcinoma, a subtype of lung cancer. These genes could contribute to the initiation and progression of lung adenocarcinoma in at least two levels. First, random walk with restart algorithm was performed on a protein–protein interaction (PPI) network constructed with PPI information in STRING by using known dysfunctional genes as seed nodes for each level, thereby yielding four groups of possible genes. Second, these genes were further evaluated in a test strategy to exclude false positives and select the most important ones. Finally, after conducting an intersection operation in any two groups of genes, we obtained several inferred driver genes that contributed to the initiation of lung adenocarcinoma in at least two omics levels. Several genes from these groups could be confirmed according to recently published studies. The inferred genes reported in this study were also different from those described in a previous study, suggesting that they can be used as essential supplementary data for investigations on the initiation of lung adenocarcinoma. 环指蛋白RING1能结合DNA并抑制基因的转录.采用酵母双杂交方法从人骨骼肌文库中筛选出了与A型核纤层蛋白(lamin A)结合的RING1蛋白,回复杂交酵母能在缺陷培养基上生长.RING1与绿色荧光蛋白融合载体转染HEK293细胞,激光共聚焦显微观察发现RING1能与带红色荧光蛋白的lamin A蛋白在细胞核周围共定位.免疫共沉淀结果证明RING1与lamin A能够相互作用.结果证明了一个新的lamin A结合蛋白,为揭示lamin A影响基因表达乃至细胞衰老提供了依据.  相似文献

 本文是对海拔930m的17年生海南五针松(Pinus fanzeliana)人工林分生物量和生产力进行了测定和研究。按平均标准木法和样方收获法分别调查了乔木层,灌木层,草本地被物层和枯枝落叶层。据调查数,建立了估测乔木层单株林木各器官干重的回归方程。方程的相关系数和估测精度都较高,具有参考价值.分析结果表明:林分总生物量平均为161.152t/ha,生产力为10630.69kg/(ha·a),其中:乔木层生物量为149.351t/ha,生产力为11095kg/(ha·a),叶面积为199248.734m2/ha,叶面积指数为19.9149m2/m2。  相似文献

Previously, we reported that retigeric acid B (RB), a natural pentacyclic triterpenic acid isolated from lichen, inhibited cell growth and induced apoptosis in androgen-independent prostate cancer (PCa) cells. However, the mechanism of action of RB remains unclear. In this study, we found that using PC3 and DU145 cells as models, RB inhibited phosphorylation levels of IκBα and p65 subunit of NF-κB in a time- and dosage-dependent manner. Detailed study revealed that RB blocked the nuclear translocation of p65 and its DNA binding activity, which correlated with suppression of NF-κB-regulated proteins including Bcl-2, Bcl-x(L), cyclin D1 and survivin. NF-κB reporter assay suggested that RB was able to inhibit both constitutive activated-NF-κB and LPS (lipopolysaccharide)-induced activation of NF-κB. Overexpression of RelA/p65 rescued RB-induced cell death, while knockdown of RelA/p65 significantly promoted RB-mediated inhibitory effect on cell proliferation, suggesting the crucial involvement of NF-κB pathway in this event. We further analyzed antitumor activity of RB in in vivo study. In C57BL/6 mice carrying RM-1 homografts, RB inhibited tumor growth and triggered apoptosis mainly through suppressing NF-κB activity in tumor tissues. Additionally, DNA microarray data revealed global changes in the gene expression associated with cell proliferation, apoptosis, invasion and metastasis in response to RB treatment. Therefore, our findings suggested that RB exerted its anti-tumor effect by targeting the NF-κB pathway in PCa cells, and this could be a general mechanism for the anti-tumor effect of RB in other types of cancers as well.  相似文献

CCL5 (previously called RANTES) is in the CC‐chemokine family and plays a crucial role in the migration and metastasis of human cancer cells. On the other hand, the effect of CCL5 is mediated via CCR receptor. RT‐PCR and flow cytometry studies demonstrated CCR5 but not CCR1 and CCR3 mRNA in oral cancer cell lines, especially higher in those with high invasiveness (SCC4) as compared with lower levels in HSC3 cells and SCC9 cells. Stimulation of oral cancer cells with CCL5 directly increased the migration and metalloproteinase‐9 (MMP‐9) production. MMP‐9 small interfering RNA inhibited the CCL5‐induced MMP‐9 expression and thereby significantly inhibited the CCL5‐induced cell migration. Activations of phospholipase C (PLC), protein kinase Cδ (PKCδ), and NF‐κB pathways after CCL5 treatment was demonstrated, and CCL5‐induced expression of MMP‐9 and migration activity was inhibited by the specific inhibitor of PLC, PKCδ, and NF‐κB cascades. In addition, migration‐prone sublines demonstrate that cells with increasing migration ability had more expression of MMP‐9, CCL5, and CCR5. Taken together, these results indicate that CCL5/CCR5 axis enhanced migration of oral cancer cells through the increase of MMP‐9 production. J. Cell. 为获得大孔树脂纯化岩高兰多酚的最佳工艺,以岩高兰的地上部分为原料,通过考察6种不同类型树脂(HPD-100、X-5、AB-8、D101、HPD-600、NKA-II)的含水率、吸附率和解吸率的大小,筛选出一种最适合纯化岩高兰多酚的树脂。在此基础上,选择对纯化工艺影响较大的4种因素(上样浓度、乙醇浓度、洗脱流速、洗脱体积),进行响应面法分析得到最佳工艺。结果表明:HPD-600型大孔树脂对岩高兰多酚的纯化效果最佳,其最优工艺参数为:上样浓度0.84 mg·mL-1;乙醇浓度62.15%;洗脱流速0.67 mL·min-1;洗脱体积2.71 BV。该条件下,岩高兰多酚的提取率为229.18 mg·g-1,岩高兰多酚的纯度由8.11%提高到22.56%,回收率为67.78%。本研究为岩高兰多酚的纯化工艺提供了新的技术路线,也可为岩高兰提取物的研究和应用提供参考。  相似文献

目的:观察新生SD大鼠原代培养皮层神经元的钙激活钾通道(Kca)在黎芦碱致神经元损伤模型上的激活、抑制效应.方法:采用细胞贴附和内面向外两种膜片钳单通道记录方法记录新生SD大鼠原代培养皮层神经元的Kca电生理活动.结果:黎芦碱在胞外可激活Kca.在有钙浴液内,细胞贴附式,钳制膜电位 30 mV,加入不同浓度黎芦碱(μmol/L:15、25、50、75),通道开放概率由0.005分别增加为0.014±0.003、0.085±0.010、0.132±0.016、0.059±0.006(P<0.01),在50μmol/L以内表现出浓度依赖性.无钙浴液内,细胞贴附式膜片上,钳制膜电位 50 mV,随药物浓度(μmol/L)增加为15、40、60、100时,通道开放概率由0.005分别增加为0.014±0.010、0.113±0.006、0.141±0.004、0 295±0.009(P<0.05).6例内面向外式膜片上,钳制膜电位 40 mV,分别加入黎芦碱25 μmol/L、50μmol/L 3 min后,通道开放概率由0.011±0.008分别增加为0.010±0.010、0.012±0.007(P>0.05).黎芦碱在胞内Kca开放概率,平均开放/关闭时间,电流幅值均无明显变化.结论:黎芦碱通过影响胞内游离钙水平间接调节Kca,在缺血缺氧早期,胞内游离钙增高激活Kca开放.  相似文献
