Both LH and FSH play a central role in controlling ovarian function in mammals. However, little is known about the type of ovarian cells that are responsive to LH and FSH in marsupials. We determined, using in situ hybridization, the localization of mRNA encoding the receptors (R) for LH and FSH in ovaries of brushtail possums. The mRNA encoding FSH-R was observed in granulosa cells of healthy follicles containing at least two complete layers of cells. The mRNA encoding LH-R was first observed in granulosa cells at the time of antrum formation. Cells of the theca interna expressed LH-R mRNA but not FSH-R mRNA. Neither FSH-R nor LH-R mRNA was detected in atretic follicles. Both FSH-R and LH-R mRNAs were observed in luteal tissue, but only LH-R mRNA was observed in interstitial cells. Granulosa cells from follicles of various sizes (0.5 to >2 mm in diameter) responded to LH and FSH treatment with an increase in cAMP synthesis. In contrast, luteal tissue did not respond to either FSH or LH treatment. In conclusion, expression of FSH-R in the brushtail possum ovary was similar to that observed in many eutherian mammals. However, active LH-R was expressed in granulosa cells much earlier in follicular development than has been previously observed. In addition, although mRNAs for both FSH-R and LH-R were observed, neither FSH nor LH treatment stimulated cAMP synthesis in luteal tissue.  相似文献

Understanding the mechanisms regulating the diversity and distribution of arthropods is essential to understanding food web interactions and ecosystem functioning. Local arthropod diversity is known to be linked to features of surrounding landscapes, including the area of human‐developed land. Yet, how such landscape moderation of diversity affects processes within local sites remains understudied. We report on a study that 1) measured the impacts of human development surrounding old‐field habitats of arthropods on arthropod food web structure within those habitats and 2) determined if these shifts were associated with cascading impacts on the plant community. We sampled the arthropod community in 16 old‐fields that span an urban‐rural gradient throughout southern New England, USA. In each field, we also established paired mesocosms enclosing vegetation, one of which allowed arthropod herbivory while the other excluded such interactions, to isolate impacts of arthropod herbivory on three functional groups of plants: grasses, goldenrod and non‐goldenrod forbs. Biomass of both herbivorous and predatory arthropods were positively related to the proportion of natural area surrounding a field early in the growing season (June). This relationship persisted later into the season for predatory arthropods (through July), but not for herbivorous arthropods. We found no evidence that the biomass of predators was related to the abundance of herbivorous arthropods in a field; or that biomass of herbivores was correlated to change in plant biomass between the two types of mesocosms. We did, however, find that in fields with low predator abundance there was greater herbivory on grasses (nutritious host), but that in high predator fields goldenrod was increasingly impacted (safe host), as is predicted by past work in old‐field ecosystems. The findings support the generalizability of landscape moderated biodiversity to non‐agricultural systems and suggests that observed shifts in food webs have implications for community and ecosystem dynamics.  相似文献

How ocean acidification affects marine life is a major concern for science and society. However, its impacts on encrusting biofouling communities, that are both the initial colonizers of hard substrata and of great economic importance, are almost unknown. We showed that community composition changed significantly, from 92% spirorbids, 3% ascidians and 4% sponges initially to 47% spirorbids, 23% ascidians and 29% sponges after 100 days in acidified conditions (pH 7.7). In low pH, numbers of the spirorbid Neodexiospira pseudocorrugata were reduced ×5 compared to controls. The two ascidians present behaved differently with Aplidium sp. decreasing ×10 in pH 7.7, whereas Molgula sp. numbers were ×4 higher in low pH than controls. Calcareous sponge (Leucosolenia sp.) numbers increased ×2.5 in pH 7.7 over controls. The diatom and filamentous algal community was also more poorly developed in the low pH treatments compared to controls. Colonization of new surfaces likewise showed large decreases in spirorbid numbers, but numbers of sponges and Molgula sp. increased. Spirorbid losses appeared due to both recruitment failure and loss of existing tubes. Spirorbid tubes are comprised of a loose prismatic fabric of calcite crystals. Loss of tube materials appeared due to changes in the binding matrix and not crystal dissolution, as SEM analyses showed crystal surfaces were not pitted or dissolved in low pH conditions. Biofouling communities face dramatic future changes with reductions in groups with hard exposed exoskeletons and domination by soft‐bodied ascidians and sponges.  相似文献

The RecG protein of Escherichia coli is a structure-specific DNA helicase that targets strand exchange intermediates in genetic recombination and drives their branch migration along the DNA. Strains carrying null mutations in recG show reduced recombination and DNA repair. Suppressors of this phenotype, called srgA, were located close to metB and shown to be alleles of priA. Suppression depends on the RecA, RecBCD, RecF, RuvAB, and RuvC recombination proteins. Nine srgA mutations were sequenced and shown to specify mutant PriA proteins with single amino acid substitutions located in or close to one of the conserved helicase motifs. The mutant proteins retain the ability to catalyze primosome assembly, as judged by the viability of recG srgA and srgA strains and their ability to support replication of plasmids based on the ColE1 replicon. Multicopy priA+ plasmids increase substantially the recombination- and repair-deficient phenotype of recG strains and confer similar phenotypes on recG srgA double mutants but not on ruvAB or wild-type strains. The multicopy effect is eliminated by K230R, C446G, and C477G substitutions in PriA. It is concluded that the 3'-5' DNA helicase/translocase activity of PriA inhibits recombination and that this effect is normally countered by RecG.  相似文献

Ehrlichia chaffeensis, transmitted from Amblyomma americanum ticks, causes human monocytic ehrlichiosis. It also infects white-tailed deer, dogs and several other vertebrates. Deer are its reservoir hosts, while humans and dogs are incidental hosts. E. chaffeensis protein expression is influenced by its growth in macrophages and tick cells. We report here infection progression in deer or dogs infected intravenously with macrophage- or tick cell-grown E. chaffeensis or by tick transmission in deer. Deer and dogs developed mild fever and persistent rickettsemia; the infection was detected more frequently in the blood of infected animals with macrophage inoculum compared to tick cell inoculum or tick transmission. Tick cell inoculum and tick transmission caused a drop in tick infection acquisition rates compared to infection rates in ticks fed on deer receiving macrophage inoculum. Independent of deer or dogs, IgG antibody response was higher in animals receiving macrophage inoculum against macrophage-derived Ehrlichia antigens, while it was significantly lower in the same animals against tick cell-derived Ehrlichia antigens. Deer infected with tick cell inoculum and tick transmission caused a higher antibody response to tick cell cultured bacterial antigens compared to the antibody response for macrophage cultured antigens for the same animals. The data demonstrate that the host cell-specific E. chaffeensis protein expression influences rickettsemia in a host and its acquisition by ticks. The data also reveal that tick cell-derived inoculum is similar to tick transmission with reduced rickettsemia, IgG response and tick acquisition of E. chaffeensis.  相似文献

The T4D bacteriophage gene 28 product is a component of the central plug of the tail baseplate, as shown by the following two independent lines of evidence. (i) A highly sensitive method for radioactive labeling of only tail baseplate plug components was developed. These labeled plug components were incorporated by a complementation procedure into new phage particles and were analyzed by radioautography after sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Three new structural proteins were found in addition to the three known tail plug proteins (i.e., gP29, gP27, and gP5). One of the three newly identified components had a molecular weight of 24,000 to 25,000 and appeared to be a product of T4D gene 28. (ii) Characterization of mutants of Escherichia coli bacteriophage T4D which produced altered gene 28 products also indicated that the gene 28 product was a viral tail component. T4D 28ts phage particles produced at the permissive temperature had altered heat labilities compared with parent T4D particles. We isolated a single-step temperature revertant of T4D 28ts and found that it produced phage particles which phenotypically resembled the original T4D particles. Since the properties of the phage baseplate components usually determine heat lability, these two changes in physical stability after two sequential single mutations in gene 28 supported the other evidence that the gene 28 product was a viral baseplate component. Also, compared with parent T4D particles, T4D 28ts and T4D 28am viral particles adsorbed at different rates to various types of host cells. In addition, T4D 28ts particles exhibited a different host range than parent T4D particles. This T4D mutant formed plaques with an extremely low efficiency on all E. coli K-12 strains tested. We found that although T4D 28ts particles adsorbed rapidly and irreversibly to the E. coli K-12 strains, as judged by gene rescue experiments, these particles were not able to inject their DNA into the E. coli K-12 strains. On the other hand, the T4D 28ts revertant had a plating efficiency on E. coli K-12 strains that was quite similar to the plating efficiency of the original parent, T4D. These properties of phage particles containing an altered gene 28 product supported the analytical finding that the gene 28 product is a structural component of the central plug of the T4D tail baseplate. They also indicated that this component plays a role in both host cell recognition and viral DNA injection.  相似文献

The effect of age on the total, transferable and percent of transferable embryos was examined in 144 embryo collections from 56 Texas Longhorn cows aged from 3 to 22 years. At 10 years of age, the percent of embryos collected that were transferable declined because of a decline in the viability of the embryos. The total embryos recovered per collection remained the same for all ages, which suggested that the pool of follicles present in the ovary was sufficient to respond to superovulation in a similar manner for all ages studied.  相似文献

Background and aims

Our study quantified variations leaf respiration in darkness (R D) and light (R L), and associated traits along the Franz Josef Glacier soil development chronosequence in New Zealand.

Methods

At six sites along the chronosequence (soil age: 6, 60, 150, 500, 12,000 and 120,000 years old), we measured rates of leaf R D, R L (using Kok method), light-saturated CO2 assimilation rates (A), leaf mass per unit area (M A), and concentrations of leaf nitrogen ([N]), phosphorus ([P]), soluble sugars and starch.

Results

The chronosequence was characterised by decreasing R D, R L and A, reduced [N] and [P] and increasing M A as soil age increased. Light inhibition of R occurred across the chronosequence (mean inhibition = 16 %), resulting in ratios of R L:A being lower than for R D:A. Importantly, the degree of light inhibition differed across the chronosequence, being lowest at young sites and highest at old sites. This resulted in R L:A ratios being relatively constant across the chronosequence, whereas R D:A ratios increased with increasing soil age. Log-log R-A-M A-[N] relationships remained constant along the chronosequence. By contrast, relationships linking rates of leaf R to [P] differed among leaves with low vs high [N]:[P] ratios. Slopes of log-log bivariate relationships linking R L to A, M A, [N] and [P] were steeper than that for R D.

Conclusions

Our findings have important implications for predictive models that seek to account for light inhibition of R, and for our understanding of how environmental gradients impact on leaf trait relationships  相似文献

We used Satellite Relay Data Loggers to obtain the first dive profiles for critically endangered leatherback turtles outside the nesting season. As individuals moved from the Caribbean out into the Atlantic, key aspects of their diving behaviour changed markedly, in line with theoretical predictions for how dive duration should vary with foraging success. In particular, in the Atlantic, where foraging success is expected to be higher, dives became much longer than in the Caribbean. The deepest-ever dive profile recorded for a reptile was obtained in the oceanic Atlantic, with a 54-min dive to 626 m on 26 August 2002. However, dives were typically much shallower (generally <200 m) and shorter (<40 min). These results highlight the suitability of this species for testing models of dive performance.  相似文献

The microbial reduction of metals has attracted recent interest as these transformations can play crucial roles in the cycling of both inorganic and organic species in a range of environments and, if harnessed, may offer the basis for a wide range of innovative biotechnological processes. Under certain conditions, however, microbial metal reduction can also mobilise toxic metals with potentially calamitous effects on human health. This review focuses on recent research on the reduction of a wide range of metals including Fe(III), Mn(IV) and other more toxic metals such as Cr(VI), Hg(II), Co(III), Pd(II), Au(III), Ag(I), Mo(VI) and V(V). The reduction of metalloids including As(V) and Se(VI) and radionuclides including U(VI), Np(V) and Tc(VII) is also reviewed. Rapid advances over the last decade have resulted in a detailed understanding of some of these transformations at a molecular level. Where known, the mechanisms of metal reduction are discussed, alongside the environmental impact of such transformations and possible biotechnological applications that could utilise these activities.  相似文献
