The relationship between the microclimate within an Oak-Hickory forest and photosynthetic characters of two resident evergreen herbs with contrasting leaf phenologies was investigated on a monthly basis for 1 full year. Heuchera americana has leaf flushes in the spring and fall, with average leaf life spans of 6–7 months. Hexastylis arifolia produces a single cohort of leaves each spring with a leaf life span of 12–13 months. We predicted that among evergreen plants inhabiting a seasonal habitat, a species for which the frequency of leaf turnover is greater than the frequency of seasonal extremes would have a greater annual range in photosynthetic capacity than a species that only produced a single flush of leaves during the year. Photosynthetic parameters, including apparent quantum yield, maximum photosynthetic capacity (Pmax), temperature of maximum photosynthesis, photochemical efficiency of PSII and leaf nitrogen (N) and chlorophyll concentrations, were periodically measured under laboratory conditions in leaves sampled from natural populations of both species. Mature leaves of both species acclimated to changing understory conditions with the mean seasonal differences being significantly greater for Heuchera than for Hexastylis. Area based maximum photosynthetic rates at 25°C were approximately 250% and 100% greater in winter leaves than summer leaves for Heuchera and Hexastylis respectively. Nitrogen concentrations were highest in winter leaves. Chlorophyll concentrations were highest in summer leaves. Low Pmax/N values for these species suggest preferential allocation of leaf nitrogen into non-photosynthetic pools and/or light-harvesting function at the expense of photosynthetic enzymes and electron transport components. Despite the increase in photosynthetic capacity, there was evidence of chronic winter photoinhibition in Hexastylis, but not in Heuchera. Among these ecologically similar species, there appears to be a trade-off between the frequency of leaf production and the balance of photosynthetic acclimation and photoinhibition.  相似文献

The role of electron transport to O2 in mitigating against photoinactivation of Photosystem (PS) II was investigated in leaves of pea (Pisum sativum L.) grown in moderate light (250 mol m–2 s–1). During short-term illumination, the electron flux at PS II and non-radiative dissipation of absorbed quanta, calculated from chlorophyll fluorescence quenching, increased with increasing O2 concentration at each light regime tested. The photoinactivation of PS II in pea leaves was monitored by the oxygen yield per repetitive flash as a function of photon exposure (mol photons m–2). The number of functional PS II complexes decreased nonlinearly with increasing photon exposure, with greater photoinactivation of PS II at a lower O2 concentration. The results suggest that electron transport to O2, via the twin processes of oxygenase photorespiration and the Mehler reaction, mitigates against the photoinactivation of PS II in vivo, through both utilization of photons in electron transport and increased nonradiative dissipation of excitation. Abscisic acid (ABA)-deficient mutants of Arabidopsis do not synthesize the epoxy-xanthophylls antheraxanthin, violaxanthin, or neoxanthin. However, thylakoid membranes from these mutants contain 3-fold more zeaxanthin than wild-type plants. This increase in zeaxanthin occurs as a stoichiometric replacement of the missing violaxanthin and neoxanthin within the pigment-protein complexes of both photosystem I and photosystem II (PSII). The retention of zeaxanthin in the dark by ABA-deficient mutants sensitizes the leaves to the development of nonphotochemical quenching (NPQ) during the first 2 to 4 min following a dark-light transition. However, the increase in pool size does not result in any increase in steady-state NPQ. When we exposed wild-type and ABA-deficient mutants leaves to twice growth irradiance, the mutants developed lower maximal NPQ but suffered similar photoinhibition to wildtype, measured both as a decline in the ratio of variable to maximal fluorescence and as a loss of functional PSII centers from oxygen flash yield measurements. These results suggest that only a few of the zeaxanthin molecules present within the light-harvesting antenna of PSII may be involved in NPQ and neither the accumulation of a large pool of zeaxanthin within the antenna of PSII nor an increase in conversion of violaxanthin to zeaxanthin will necessarily enhance photoprotective energy dissipation.  相似文献

The cellular mechanism by which an injection of sheep red blood cells (SRBC) results in an increased production of B lymphocytes in mouse bone marrow has been studied by adoptive cell transfer and the use of two in vivo assays of bone marrow B-cell genesis. Proliferation of B progenitor cells was examined by immunofluorescent labeling combined with mitotic arrest, while small lymphocyte renewal was measured by [3H]thymidine labeling and radioautography. In C3H/HeJ mice the administration of SRBC resulted in increased proliferation among bone marrow pre-B cells which contained cytoplasmic mu heavy chains but lacked kappa light chains and surface mu chains. The turnover of small lymphocytes also increased. These stimulatory effects were transferred to naive recipient mice by organ fragments and by cell suspensions harvested from the spleens of donor mice injected with SRBC. In contrast, spleen cells and thymus cells from saline-injected donors and thymus cells from SRBC-injected donors had no such stimulatory effects. The results demonstrate that spleen cells mediate the stimulatory effect of SRBC on bone marrow B-lymphocyte production. Spleen cell transfer provides a system to study further the cells and factors involved in the regulation by external environmental agents of the rate of primary B-cell genesis in vivo.  相似文献

Effects of varying light intensity and nitrogen nutrition on photosynthetic physiology and biochemistry were examined in the sun plant Phaseolus vulgaris (common bean) and in the shade plant Alocasia macrorrhiza (Australian rainforest floor species). In both Phaseolus and Alocasia, the differing growth regimes produced large changes in photosynthetic capacity and composition of the photosynthetic apparatus. CO2-saturated rates of photosynthesis were linearly related to leaf nitrogen (N) content in both species but photosynthesis per unit leaf N was markedly higher for Phaseolus than for Alocasia. Photosynthetic capacity was also higher in Phaseolus per unit ribulose 1,5-bisphosphate (RuBP) carboxylase (RuBPCase) protein. The leaf content of RuBPCase was linearly dependent on leaf N content in the two species. However, the proportion of leaf N which was RuBPCase was greater in Phaseolus than in Alocasia and was more sensitive to growth conditions, ranging from 6% of leaf N at low light to 20% at high light. In Alocasia, this range was much less, 6 to 11%. However, chlorophyll content was much more sensitive to light intensity in Alocasia. Thus, the RuBPCase/chlorophyll ratio was quite responsive to N availability and light intensity in both species (but for different reasons), ranging from 6 grams per gram for Phaseolus and 2 grams per gram for Alocasia at high leaf N and 1.5 gram per gram for Phaseolus and 0.5 gram per gram for Alocasia at low leaf N. These large changes in the proportions of components of the photosynthetic apparatus had marked effects on the sensitivity of these species to photoinhibition. These environmental effects also caused changes in the absolute levels of metabolites of the photosynthetic carbon reduction cycle. Concentrations of RuBP and P-glycerate were approximately 2-fold higher in high light-grown than low light-grown Phaseolus and Alocasia when expressed on a leaf area basis. However, if metabolite pool sizes are expressed on the basis of the RuBPCase catalytic site concentration, then they were little affected by the marked changes in leaf makeup. There appears to be fundamental differences between these species in the mechanism of sun-shade adaptation and N partitioning in the photosynthetic apparatus that result in significant differences in the N-use efficiency of photosynthesis between Phaseolus and Alocasia but similar RuBPCase:substrate:product ratios despite these differences.  相似文献

Two CAM species, Kalanchoë daigremontiana Hamet et Perrier and Hoya carnosa (L.) R. Br., were grown under a range of five photon flux area densitites (PFD) and then characterized. Significant acclimation to shade was indicated by progressive decreases in leaf thickness, rates of respiratory O2 uptake, light compensation point, maximum rates of photosynthetic O2 evolution, nocturnal acid accumulation, and δ13C values, and increases in chlorophyll concentration and absolute levels of room temperature (25°C) and 77K fluorescence. Quantum yields (as measured by O2 exchange) and the ratio of variable 77K fluorescence over the maximum yield (Fv/Fm) were relatively constant across the treatments. The only significant deviation from the above characteristics was in H. carnosa grown under full glasshouse PFD, where it apparently experienced photoinhibition. Following a photoinhibitory treatment, K. daigremontiana exhibited increases in the light compensation point and progressively greater reductions in the quantum yield, maximum photosynthetic rate, Fv/Fm, and the variable component of room temperature fluorescence with increasing shade during growth. Thus although Crassulacean acid metabolism plants can adjust to shaded conditions, they are susceptible to photoinhibition when exposed to higher PFD than that experienced during growth.  相似文献

To examine the concept that the genesis of lymphocytes in the bone marrow may be regulated by homeostatic feedback signals from peripheral B lymphocytes or their products, lymphocyte production was measured in mice selectively depleted of B lymphocytes by repeated administration of anti-IgM antibodies from birth. The turnover of small lymphocytes was quantitated radioautographically after DNA labeling by continuous infusion of 3H-thymidine. In the femoral marrow of anti-IgM-treated mice, the number of small lymphocytes was reduced and their turnover time was shorter than in control mice, presumably reflecting the premature elimination from the marrow of maturing cells about to express surface IgM. The absolute number of small lymphocytes being produced per femur in unit time, however, was identical in anti-IgM-treated and control mice. Lymphocyte production in the thymus was also unaffected by anti-IgM suppression whereas in the spleen the turnover of small lymphocytes was reduced due to the lack of young immigrant B lymphocytes from the bone marrow. The results demonstrate that the normal large-scale production of lymphocytes in mouse bone marrow is independent of the magnitude of the peripheral pool of B lymphocytes or the level of circulating immunoglobulins, suggesting the process is not subject to feedback control. Some implications for the genesis and diversity of primary B lymphocytes are discussed.  相似文献

Photosynthetic O2 production and photorespiratory O2 uptake were measured using isotopic techniques, in the C3 species Hirschfeldia incana Lowe., Helianthus annuus L., and Phaseolus vulgaris L. At high CO2 and normal O2, O2 production increased linearly with light intensity. At low O2 or low CO2, O2 production was suppressed, indicating that increased concentrations of both O2 and CO2 can stimulate O2 production. At the CO2 compensation point, O2 uptake equaled O2 production over a wide range of O2 concentrations. O2 uptake increased with light intensity and O2 concentration. At low light intensities, O2 uptake was suppressed by increased CO2 concentrations so that O2 uptake at 1,000 microliters per liter CO2 was 28 to 35% of the uptake at the CO2 compensation point. At high light intensities, O2 uptake was stimulated by low concentrations of CO2 and suppressed by higher concentrations of CO2. O2 uptake at high light intensity and 1000 microliters per liter CO2 was 75% or more of the rate of O2 uptake at the compensation point. The response of O2 uptake to light intensity extrapolated to zero in darkness, suggesting that O2 uptake via dark respiration may be suppressed in the light. The response of O2 uptake to O2 concentration saturated at about 30% O2 in high light and at a lower O2 concentration in low light. O2 uptake was also observed with the C4 plant Amaranthus edulis; the rate of uptake at the CO2 compensation point was 20% of that observed at the same light intensity with the C3 species, and this rate was not influenced by the CO2 concentration. The results are discussed and interpreted in terms of the ribulose-1,5-bisphosphate oxygenase reaction, the associated metabolism of the photorespiratory pathway, and direct photosynthetic reduction of O2.  相似文献

Summary The 13C values of submerged aquatic plants from contrasting but relatively defined habitats, and the 13C values of emergent, floating and submerged leaves of dimorphic aquatic plants, were measured. In many instances the 13C values of dissolved inorganic carbon in the water were also measured. Plant 13C values in the vicinity of-40 to-50 were found in rapidly flowing spring waters with carbonate 13C values of-16 to-21, consistent with the notion that species such as Fontinalis antipyretica almost exclusively assimilate free CO2 via RuP2 carboxylase. Plant 13C values in the vicinity of-10 to-15 in sluggish water with carbonate 13C values of about-5 were observed, consistent with the notion that boundary layer diffusion and/or HCO3 - uptake may determine the 13C value of submerged aquatic plants in these circumstances. Comparisons of 13C values of the same or related species growing in waters of similar carbonate 13C value but different flow rates confirmed this view; more negative 13C values were frequently associated with plants in fast moving water. In Britain, but not in Finland, the 13C values of submerged leaves of dimorphic plants were almost invariably more negative than in aerial leaves. The 13C value of carbonate from chalk streams and in acid springs indicate substantial inputs of respiratory CO2, as opposed to atmospheric carbon. The contributions of these variations in 13C of the carbon source, and of isotope fractionation in diffusion, to the 13C value of submerged parts of dimorphic plants is discussed.  相似文献
