Healthcare-associated infections (HAIs) are causes of mortality and morbidity worldwide. The prevalence of bacterial resistance to common antibiotics has increased in recent years, highlighting the need to develop novel alternatives for controlling these pathogens. Pitviper venoms are composed of a multifaceted mixture of peptides, proteins and inorganic components. L-amino oxidase (LAO) is a multifunctional enzyme that is able to develop different activities including antibacterial activity. In this study a novel LAO from Bothrops mattogrosensis (BmLAO) was isolated and biochemically characterized. Partial enzyme sequence showed full identity to Bothrops pauloensis LAO. Moreover, LAO here isolated showed remarkable antibacterial activity against Gram-positive and -negative bacteria, clearly suggesting a secondary protective function. Otherwise, no cytotoxic activities against macrophages and erythrocytes were observed. Finally, some LAO fragments (BmLAO-f1, BmLAO-f2 and BmLAO-f3) were synthesized and further evaluated, also showing enhanced antimicrobial activity. Peptide fragments, which are the key residues involved in antimicrobial activity, were also structurally studied by using theoretical models. The fragments reported here may be promising candidates in the rational design of new antibiotics that could be used to control resistant microorganisms.  相似文献

The conversion of natural grasslands to cultivated pastures can have a significant impact on the composition and structure of soil macro- and mesofauna groups, compromising the resilience of these organisms and the ecosystem services they provide. We studied the responses of these groups to increasing levels of soil disturbance across a gradient of four land management practices: Natural grassland (NG), Improved-natural grassland (IG), Perennial-cultivated pasture (PP), and Annual-cultivated pasture (AP). The NG area had a higher abundance of macrofauna, greater dominance of certain groups, particularly Isoptera, and a lower abundance of mesofauna. On the other hand, the IG area had a higher abundance and diversity of macro- and mesofauna, with a lower dominance of specific groups. The PP area had a higher abundance of Coleoptera, Oligochaeta, and Hemiptera, whereas the AP area, despite soil disturbance, showed a higher abundance of mesofauna, particularly mites, Collembola, and Enchytraeidae, but lower diversity. Different grassland management practices have significantly altered the composition and structure of macro- and mesofauna groups, resulting in high dissimilarity between communities. We recommend IG as a more productive and sustainable alternative to the total replacement of natural vegetation (NG) with intensified converted pastures (PP and AP), as it maintains soil fauna diversity and ecosystem services.  相似文献

Mycobacterium tuberculosis shikimate dehydrogenase (MtbSD) catalyzes the fourth reaction in the shikimate pathway, the NADPH-dependent reduction of 3-dehydroshikimate. To gather information on the kinetic mechanism, initial velocity patterns, product inhibition, and primary deuterium kinetic isotope effect studies were performed and the results suggested a steady-state ordered bi-bi kinetic mechanism. The magnitudes of both primary and solvent kinetic isotope effects indicated that the hydride transferred from NADPH and protons transferred from the solvent in the catalytic cycle are not significantly rate limiting in the overall reaction. Proton inventory analysis indicates that one proton gives rise to solvent isotope effects. Multiple isotope effect studies indicate that both hydride and proton transfers are concerted. The pH profiles revealed that acid/base chemistry takes place in catalysis and substrate binding. The MtbSD 3D model was obtained in silico by homology modeling. Kinetic and chemical mechanisms for MtbSD are proposed on the basis of experimental data.  相似文献

High concentrations of cadmium (Cd) in the environment can threaten the local biota and one of its main sources is anthropic activities such as zinc (Zn) mining. Some plant species are able to tolerate high Cd concentrations, using anatomical and physiological strategies to avoid the absorption or accumulation of this element in their biomass. The in vitro assessment of these strategies is an efficient way to control variables external to the experiment. We aimed to investigate the anatomical and physiological changes in Alternanthera tenella exposed to Cd and its potential for accumulation in controlled microenvironmental conditions. We evaluated changes in the leaf and root anatomy, antioxidant system, and biomass of A. tenella grown in a culture medium containing increasing Cd concentrations (0, 50, 100, and 150 μM), in the presence of 1500 μM Zn. Alternanthera tenella was able to accumulate Cd and Zn and these elements competed for absorption by the species. Increase in Cd in the medium led to a progressive thickening of the root tissues, which was also observed on the leaves, albeit only at concentrations below 100 μM Cd. The concentration of 150 μM Cd was toxic to the leaf tissue and stimulated the formation of hydrogen peroxide, interfering with the antioxidant system and reducing plant biomass and the chlorophyll levels. Therefore, in vitro cultivated A. tenella can accumulate Cd and tolerate up to 100 μM Cd by modifying its anatomy and physiology in order to cope with Cd stress.  相似文献

The invasion of the northwestern Atlantic by the Indo-Pacific lionfish has developed extraordinarily fast, and is expected to cause one of the most negative ecological impacts among all marine invasions. In less than 30 years, lionfish have dramatically expanded their distribution range to an area encompassing the eastern coast of the USA, Bermuda, the entire Caribbean region and the Gulf of Mexico. The rapidity of the lionfish spread has raised concerns in other parts of the Atlantic that may be under the reach of the invasion. Despite the anticipation that lionfish would eventually extend their range throughout most of the eastern coast of South America, it had not been recorded in Brazil until now. Here we report the first lionfish appearance for the Brazilian coast and show that the individual collected by us is genetically linked to the invasive Caribbean population. Since small-range endemics are found in several locations in Brazil and are among the species that are most vulnerable to extinction, we recommend urgent control, management and education measures aimed at minimizing the effects of this impending invasion.  相似文献

An understanding of isoniazid (INH) drug resistance mechanism in Mycobacterium tuberculosis should provide significant insight for the development of newer anti-tubercular agents able to control INH-resistant tuberculosis (TB). The inhA-encoded 2-trans enoyl-acyl carrier protein reductase enzyme (InhA) has been shown through biochemical and genetic studies to be the primary target for INH. In agreement with these results, mutations in the inhA structural gene have been found in INH-resistant clinical isolates of M.tuberculosis, the causative agent of TB. In addition, the InhA mutants were shown to have higher dissociation constant values for NADH and lower values for the apparent first-order rate constant for INH inactivation as compared to wild-type InhA. Here, in trying to identify structural changes between wild-type and INH-resistant InhA enzymes, we have solved the crystal structures of wild-type and of S94A, I47T and I21V InhA proteins in complex with NADH to resolutions of, respectively, 2.3A, 2.2A, 2.0 A, and 1.9A. The more prominent structural differences are located in, and appear to indirectly affect, the dinucleotide binding loop structure. Moreover, studies on pre-steady-state kinetics of NADH binding have been carried out. The results showed that the limiting rate constant values for NADH dissociation from the InhA-NADH binary complexes (k(off)) were eleven, five, and tenfold higher for, respectively, I21V, I47T, and S94A INH-resistant mutants of InhA as compared to INH-sensitive wild-type InhA. Accordingly, these results are proposed to be able to account for the reduction in affinity for NADH for the INH-resistant InhA enzymes.  相似文献

While mouthbrooding is not an uncommon parental care strategy in fishes, paternal mouthbrooding only occurs in eight fish families and is little studied. The high cost of paternal mouthbrooding to the male implies a low risk of investment in another male''s offspring but genetic parentage patterns are poorly known for paternal mouthbrooders. Here, we used single-nucleotide polymorphism genetic data to investigate parentage relationships of broods of two mouthbrooders of northern Australian rivers, mouth almighty Glossamia aprion and blue catfish Neoarius graeffei. For N. graeffei, we found that the parentage pattern was largely monogamous with the brooder male as the sire. For G. aprion, the parentage pattern was more heterogeneous including observations of monogamous broods with the brooder male as the sire (73%), polygyny (13%), cuckoldry (6%) and a brood genetically unrelated to the brooder male (6%). Findings demonstrate the potential for complex interrelationships of male care, paternity confidence and mating behaviour in mouthbrooding fishes.  相似文献
