Obtaining of monoclonal antibodies to human growth hormone and their characteristics

The aim of present study was to obtain the hybridomas producing monoclonal antibodies against human growth hormone (Mabs hGH), to investigate the properties of the obtained Mabs and the possibility of their application in immunoanalytical systems. Two hybridomas secreting Mabs against hGH and belonging to the IgGI subclass have been obtained. The cross-reactivity of the Mabs with structurally similar to hGH hormones (hGHbio, hPL, hPRL, bGH, bPRL, pPRL) using indirect IFA has been studied. It has been shown that Mabs hGHI and Mabs hGH2 are directed to common specific antigenic determinant i.e. they have the same epitope specificity and don't react with other structurally related hormones, i.-e. this determinant is unique for hGH. The obtained Mabs hGH2 would find application for determination hGH by immunochemical methods in fractions while the hormone isolation from pituitaries and hGH obtained recombinant DNA methodology. The development of immunosorbents on the basis Mabs hGH2 seems to be perspective. Application of this immunosorbent may give possibility to optimize hormone isolation process.     

Kinetics of hematopoietic stem cells in the bone marrow of hepatectomized mice

Two-thirds of the liver was removed from (CBA X C57BL/6j) F1 female mice. A significant increase of the number of endogenous colonies count in the spleen of partially hepatectomized mice was observed on the 5-th day after the operation. This increase was not associated with the changes in the number of stem cells in the bone marrow as partial hepatectomy at different times after the operation exerted no effect on the number of colony-forming units (CF1) in the bone marrow.     

Y-chromosome variation in Altaian Kazakhs reveals a common paternal gene pool for Kazakhs and the influence of Mongolian expansions

Kazakh populations have traditionally lived as nomadic pastoralists that seasonally migrate across the steppe and surrounding mountain ranges in Kazakhstan and southern Siberia. To clarify their population history from a paternal perspective, we analyzed the non-recombining portion of the Y-chromosome from Kazakh populations living in southern Altai Republic, Russia, using a high-resolution analysis of 60 biallelic markers and 17 STRs. We noted distinct differences in the patterns of genetic variation between maternal and paternal genetic systems in the Altaian Kazakhs. While they possess a variety of East and West Eurasian mtDNA haplogroups, only three East Eurasian paternal haplogroups appear at significant frequencies (C3*, C3c and O3a3c*). In addition, the Y-STR data revealed low genetic diversity within these lineages. Analysis of the combined biallelic and STR data also demonstrated genetic differences among Kazakh populations from across Central Asia. The observed differences between Altaian Kazakhs and indigenous Kazakhs were not the result of admixture between Altaian Kazakhs and indigenous Altaians. Overall, the shared paternal ancestry of Kazakhs differentiates them from other Central Asian populations. In addition, all of them showed evidence of genetic influence by the 13(th) century CE Mongol Empire. Ultimately, the social and cultural traditions of the Kazakhs shaped their current pattern of genetic variation.     

Effect of cytokinins on expression of radish CLE genes

CLE (CLAVATA3/ENDOSPERM SURROUNDING REGION) peptides are peptide phytohormones playing the important role in the regulation of various type meristem development and also in the interaction between plants and parasites and symbionts. At the same time, the interaction of CLE peptides with other phytohormones, participating in these processes, is essentially unstudied. Using real-time RT-PCR, we analyzed expression of some genes encoding CLE peptides in radish (Raphanus sativus var. radicula Pers.) under normal conditions and after treatment with the cytokinin 6-benzyladenine. Even after short-term (30 min) action of cytokinin, expression of group A CLE genes was manifold suppressed, whereas cytokinin did not affect expression of group B CLE genes. Radish lines contrasting in the trait “spontaneous tumor formation” demonstrated similar dynamics of CLE gene expression in response to cytokinin treatment but differed in the levels of their expression. A possibility of interaction between CLE peptides and cytokinins in the spontaneous development of tumors in radish lines is discussed. The important part of the work was the selection of optimal reference genes for the analysis of radish gene expression by real-time RT-PCR method. Most stable expression was observed for the genes of ubiquitin (RsUBQ) and glyceraldehydes-3-phosphate dehydrogenase (RsGAPDH).     

An Assay for Express Screening of Potato Transformants by GFP Fluorescence

An express assay for screening of potato transformants by their GFP fluorescence intensities is developed. In comparison to the widely used methods of transgenic plant screening by PCR, Real-Time RTPCR or Northern-blotting, the GFP fluorescence assay needs no expensive reagents and takes less time. This approach may also be used for nondestructive screening of the T0 transgenic regenerants which can be further grown and used. To prove this assay reliability, the expression of the hGFP gene in the leaves of transgenic potato (cv. Skoroplodny) plants, determined by its mRNA accumulation, was compared to GFP fluorescence intensity in the micro-samples of aseptic plant leaves. The strong correlation between the results of these two methods is the evidence of positive dependence of GFP fluorescence intensity on the target mRNA content.     

A Possible Approach to Comparison of Survival Functions

Special numerical index for comparison of survival functions is proposed. The index is called by directed probabilistic semimetric. The examples of these semimetrics are presented. The procedure of the index estimating on sampled data is considered.     

Method of disinfecting plague-infected fleas in preparation for electron microscopic study

During fixation of fleas infected with plague agent (with preliminary cut heads, limbs and posterior part of the abdomen) in 2.5% glutaric dialdehyde or 2% osmium tetroxide their disinfection was obtained in two days. After double fixation with 2-hour exposure in each of these fixators disinfection of the material was acheived only after additional 2-day maintenance in 70% ethyl alcohol. Fleas, which had been placed into fixators or in 70% ethyl alcohol after double fixation without preliminary dissection, were disinfected completely only in 10 days.
