Samples from cardiac valves of 31 patients were analyzed by gas chromatography and mass spectrometry. The algorithm of mass spectrometric parameters was developed, which permitted the determination of about 200 known microbial fatty acids, aldehydes and sterols, sufficient for the detection and quantitative determination of more that 170 taxons of clinically significant microorganisms on the genus or species levels. The quantitative and qualitative differences in the composition of microbial markers of endocardial valves in normal and pathological states, particularly in cases of infectious endocarditis, were detected. The participation of 37 microbial taxons in the process was confirmed. The level of endocardium colonization in infectious endocarditis reached from 2 to 7 x 10(9) microbial cells/g of valvular tissue (which exceeded twofold the equivalent concentrations of the marker in the normal state). In terms of quantity, the leading role was played by Cardiobacterium hominis.  相似文献

The importance of the intraspecific typing of salmonellae for effective epidemiological analysis is shown. The methods (phenotypical and genotypical), used for the epidemiological marking of salmonellae, are characterized, their advantages and drawbacks, as well as prospects of their further use, are given. The importance of the standardization of molecular-genetic methods to ensure data exchange between different laboratories and the investigation of international outbreaks of enteric infections is pointed out. The international projects PulseNet and Salmgene of the surveillance of Salmonella infection, based on genotyping of Salmonella isolates, are presented.  相似文献

Spotted sterile male - a new mutation in mice is described (tentative symbol Ssm). White spotting on the belly, legs and tail as well as sterility in heterozygous males Ssm/+ of the B10.M strain are caused by autosomal semidominant gene Ssm. The gene is localized on the 5 chromosome: the frequency of recombination between Ssm and go is 13.6 +/- 1.6%; Ssm is closely linked to Wv. The diheterozygotes Ssm+/+Wv are darkeyed white sterile mice. The deficiency of spermatogenic epithelium cells, emptyness of seminiferous tubules as well as interstitial tissue overgrowing occurred in the testis in sterile males Ssm/+ of B10.M. The fertile hybrid males Ssm/+ are obtained in outcrossing of females Ssm/+ of B10.M with males of YT/Y, CBA/CaY, DBA/2JY, A.CA/Y strains.  相似文献

Effects of hypochlorous acid and of the myeloperoxidase-hydrogen peroxide-chloride system on mono- and polyunsaturated phosphatidylcholines were analyzed by means of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Chlorohydrins and glycols were detected as main products according to the characteristic shift of molecular masses. Mainly mono-chlorohydrins result upon the incubation of HOCl/(-)OCl with 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine, whereas only traces of mono-glycols were detected. 1-Palmitoyl-2-linoleoyl-sn-glycero-3-phosphocholine yielded a complex mixture of products. Mono-chlorohydrins and glycols dominated only at short incubation, while bis-chlorohydrins as well as products containing one chlorohydrin and one glycol moiety appeared after longer incubation. Similarly, a complex product mixture resulted upon incubation of 1-stearoyl-2-arachidonoyl-sn-glycero-3-phosphocholine with hypochlorous acid. Additionally, tris-chlorohydrins, products with two chlorohydrin and one glycol moiety, as well as lysophosphatidylcholines and fragmentation products of the arachidonoyl side chain were detectable. Mono-chlorohydrins of 1-stearoyl-2-oleoyl-sn-glycero-3-phosphocholine were detected after the incubation of the latter phospholipid with the myeloperoxidase-hydrogen peroxide-chloride system at pH 6.0. These chlorohydrins were not observed in the absence of chloride, hydrogen peroxide, or myeloperoxidase as well as in the presence of methionine, taurine, or sodium azide. Thus, mono-chlorohydrins in 1-stearoyl-2-oleoyl-sn-glycero-3-phosphocholine produced by hypochlorous acid from the myeloperoxidase-hydrogen peroxide-chloride system can also be detected by means of MALDI-TOF MS.  相似文献

The monosaccharide composition of cell hydrolysates can be used as a criterion for the chemical differentiation of gram-positive bacteria. The monosaccharide composition of six bacterial species belonging to the genus Bacillus has been determined using gas chromatography, mass spectrometry and computers. The qualitative composition was similar, glucose, galactose, ribose and glucosamine being the main components in all of the species. Some Bacillus species differed in their minor components. Although the monosaccharide composition appeared to be homogeneous, bacteria can be identified in terms of their carbohydrate profile using computers. To this end, the monosaccharide composition of bacterial cells is represented as a two-dimensional data file including the qualitative composition of components and the quantity of each component.  相似文献

A new assay for superoxide radicals is based on the interaction of hydroxylamine (1-oxy-2,2,6,6-tetramethyl-4-oxopiperidine) with superoxide, giving rise to a stable nitroxide radical. Working concentration ranges of hydroxylamine and cells are determined. It was shown that the amount of superoxide generated was proportional to the concentration of nitroxide radicals. The sensitivity and specificity of the proposed assay were compared to chemiluminescence and cytochrome-c reduction.  相似文献

We studied on apple snail neurons the connection between K+ and Na+ concentration gradients, transmembrane difference of potentials, and concentrations of Ca2+ in the external medium. Sensitivity of the resting potential (RP) of neurons to the influence of temperature and to metabolic poisons rose considerably with a decrease of Ca2+ concentration in the solution surrounding a ganglion. An excess of Ca2+ in the external medium did not affect the RP or ion concentration in nerve cells. Removal of Na2+ from this solution causes hyperpolarization of the membrane which disappears when active transport of sodium ions through the membrane is suppressed. Sodium enrichment and potassium impoverishment of the neurons are observed in potassium-free solutions at 4°C. Reaccumulation of K+ and exclusion of Na+ from the solutions of 21°C depends on the concentration of Ca2+ in the medium. The ionic composition of the neurons is not restored upon removal of Ca2+ from the solution. Upon increasing the amount of Ca2+, movement of ions against the concentration gradients is intensified. Thus, it may be concluded that Ca2+ ions on the one hand participate in the maintenance of normal passive permeability of ions through the membrane, and on the other accelerate active transport of K+ and Na+ against the concentration gradients. A dependence of activity of histidase from rat liver and skin on the agents affecting the activity of the adenylate cyclase systeme was studied in vitro. Under conditions optimal for the activity of liver phosphorylase protein kinase the skin extract histidase was activated 2-3-fold. This is indicative of a possibility of regulation of the skin histidase activity via the adenylate cyclase system by modification of enzyme by phosphorylation-dephosphorylation, which is performed by 3':5'-AMP-dependent protein kinase. Theophylline at concentrations of 10(-4) M and 10(-3) M activates partially purified histidase (both liver and skin forms), probably in the course of direct interaction with the enzyme.  相似文献
