Common bean (Phaseolus vulgaris L.) varies in growth habit from aggressive climbing types to bush beans. Growth habit is determined by a combination of factors
including determinate versus indeterminate growth, total plant height, degree of branching and internode length. Together
these factors make up climbing ability. The objective of this research was to determine the quantitative trait loci (QTL)
controlling climbing ability in a F5:8 recombinant inbred line population derived from an inter-gene pool cross of an aggressive indeterminate climbing bean with
type IV growth habit (G2333) by an indeterminate bush bean of type IIb growth habit (G19839). The population was planted in
four randomized complete block design experiments across environments that varied in altitude (from 1,000 to 1,750 masl) and
soil fertility (low versus high phosphorus). QTL were identified for plant height, internode length and number of branches
per plant on a genetic map covering all common bean linkage groups with a total length of 1,175 cM. In addition a scale was
developed to evaluate overall climbing ability and was also used to identify QTL. A total of 7 QTL were found for plant height,
9 for climbing ability, 6 for internode length and 1 for branch number. The largest number and most significant QTL were found
on the lower half of linkage group B04 suggesting a major pleiotropic locus for growth habit traits at this location of the
genome that is distinct from previously characterized genes which control plant morphology of the crop. 相似文献

The host encoded cellular prion protein (PrPC) is an N-linked glycoprotein tethered to the cell membrane by a glycophosphatidylinositol (GPI) anchor. Under certain conditions, PrPC can undergo conversion into a conformationally-altered isoform (PrPSc) widely believed to be the pathogenic agent of transmissible spongiform encephalopathies (TSEs). Understanding the tissue-specific expression of PrPC is crucial considering that cells expressing high levels of PrPC bear a risk for conversion and accumulation of PrPSc. In the present study, fifteen bovine somatic tissues were analyzed for PrPC expression by quantitative western blot and immunohistochemistry. Quantitative western blot analysis revealed highest expression of PrPC in cerebellum, obex and spinal cord. Intermediate levels were detected in thymus, intestine, nerve, heart and spleen, and lower levels in lung, muscle, kidney, lymph node, skin, pancreas and liver. Immunohistochemical analysis detected intense cellular-specific PrPC staining in neurons, thymocytes and lymphocytes. PrPC was also detected in the enteric wall, pancreatic islets of langerhans, myocardium, pulmonary alveolar sacs, renal glomeruli and dermal epithelial cells. This study demonstrated the quantitatively varied, wide-spread, tissue- and cell-specific expression pattern of PrPC in bovine somatic tissues. The aim of this study was to determine whether season of birth and length of nursing affected the duration of postpartum anestrus in Creole female goats maintained on a constant plane of nutrition in subtropical Mexico. Three experiments were conducted in the Laguna region in the State of Coahuila, Mexico (26 degrees N). In the first experiment, 34 goats gave birth in January; in the second, 31 females gave birth in May; and in the third, 22 goats kidded in October. At parturition, females were allocated to 1 of 3 groups based on body weight and date of parturition: kids were weaned at 2, 30 or 90 d according to their group. After weaning, females were milked manually once a day until the end of the study. All animals were kept in a shed and were fed alfalfa ad libitum and given 200 g of concentrate daily. Starting 1 wk after parturition, estrous behavior was detected twice daily using an apron-bearing male, and blood samples were obtained twice weekly to determine ovarian activity from the plasma progesterone levels. A strong effect of month of parturition was found on the duration of postpartum anestrus (P < 0.0001), which was longer in females kidding in January (about 200 d) than in those kidding in May (about 100 d) or October (about 50 d). A tendency for an interaction between season of parturition and length of nursing was observed in the length of anovulation (P < 0.07): for parturition in October, anestrus was longer when kids were weaned after 90 d than after 2 or 30 d (P < 0.01). Season of parturition also affected dates of reinitiation of ovulatory and estrous activity (P < 0.001). Proportions of normal, short and long cycles and of associations between estrous and ovulations were not influenced by season of parturition or the age of weaning. These data demonstrate that in subtropical latitudes, season of parturition can dramatically influence the duration of postpartum anestrus independently of the availability of food. 相似文献

Thermal tolerance of Photosystem II (PSII) highly influences plant distribution worldwide because it allows for photosynthesis during periods of high temperatures and water stress, which are common in most terrestrial ecosystems and particularly in dry and semi-arid ones. However, there is a lack of information about how this tolerance influences invasiveness of exotic species in ecosystems with seasonal drought. To address this question for Mediterranean-type ecosystems (MTE) of the Iberian Peninsula, we carried out an experiment with fifteen phylogenetically related species (8 invasive and 7 native, Pinus pinaster Ait., Pinus radiata D. Don, Schinus molle Linn., Elaeagnus angustifolia L., Eucalyptus globulus Labill., Acacia melanoxylon R. Br., Gleditsia triacanthos L., Pistacia terebinthus L., Rhamnus alaternus L., Anagyris foetida L., Colutea arborescens L., Oenothera biennis L., Epilobium hirsutum L., Achillea filipendulina Lam. and Achillea millefolium L). Seedlings were grown and maximal photochemical efficiency of PSII (Fv/Fm) was measured at two water availabilities (well-watered and with water stress). PSII thermal tolerance measurements were related to specific leaf area (SLA), which varied significantly across the study species, and to the mean potential evapotranspiration (PET) of the month with the lowest precipitation in the native areas of both groups and in the invaded area of the Iberian Peninsula. Additionally, PSII thermal tolerance measurements under water stress were phylogenetically explored. Invasive and native species neither differed in SLA nor in their thermal tolerance under well-watered conditions. For well-watered plants, SLA was significantly and positively related to PSII thermal tolerance when all species were explored together regardless of their invasive nature. However, this relationship did not persist under water stress and invasive species had higher plastic responses than Mediterranean natives resulting in higher leaf temperatures. Higher PSII thermal tolerance could explain invasiveness because it allows for longer periods of carbon acquisition under water stress. In fact, PSII thermal tolerance was positively related to the PET of the invaded and native areas of the Iberian Peninsula. PSII thermal tolerance was not related to PET at the native range of the invasive species, suggesting that successful invasive species were plastic enough to cope with novel dry conditions of the Iberian Peninsula. Moreover, our phylogenetic results indicate that future scenarios of increased aridity in MTE associated to climate change will filter invasion success by taxonomic identity. Myeloid-derived suppressor cells are increased in the peripheral blood of advanced-stage cancer patients; however, no studies have shown a correlation of these immunosuppressive cells with clinical outcomes in melanoma patients. We characterized the frequency and suppressive function of multiple subsets of myeloid-derived suppressor cells in the peripheral blood of 34 patients with Stage IV melanoma, 20 patients with Stage I melanoma, and 15 healthy donors. The frequency of CD14+ MDSCs (Lin? CD11b+ HLA-DR? CD14+ CD33+) and CD14? MDSCs (Lin? CD11b+ HLA-DR? CD14? CD33+) was increased in the peripheral blood of Stage IV melanoma patients relative to healthy donors. The frequency of CD14+ and CD14? MDSCs correlated with each other and with the increased frequency of regulatory T cells, but not with classically defined monocytes. CD14? MDSCs isolated from the peripheral blood of Stage IV melanoma patients suppressed T cell activation more than those isolated from healthy donors, and the frequency of these cells correlated with disease progression and decreased overall survival. Our study provides the first evidence that the frequency of CD14? MDSCs negatively correlates with clinical outcomes in advanced-stage melanoma patients. Phosphoinositides control many different processes required for normal cellular function. Myotubularins are a family of Phosphatidylinositol 3-phosphate (PtdIns3P) phosphatases identified by the positional cloning of the MTM1 gene in patients suffering from X-linked myotubular myopathy and the MTMR2 gene in patients suffering from the demyelinating neuropathy Charcot-Marie-Tooth disease type 4B. MTM1 is a phosphatidylinositol phosphatase with reported specificity toward PtdIns3P, while the related proteins MTMR2 and MTMR3 hydrolyze both PtdIns3P and PtdIns(3,5)P2. We have investigated MTM1 and MTMR6 and find that they use PtdIns(3,5)P2 in addition to PtdIns3P as a substrate in vitro. The product of PtdIns(3,5)P2 hydrolysis, PtdIns5P, causes MTM1 to form a heptameric ring that is 12.5 nm in diameter, and it is a specific allosteric activator of MTM1, MTMR3, and MTMR6. A disease-causing mutation at arginine 69 of MTM1 falling within a putative pleckstrin homology domain reduces the ability of the enzyme to respond to PtdIns5P. We propose that the myotubularin family of enzymes utilize both PtdIns3P and PtdIns(3,5)P2 as substrates, and that PtdIns5P functions in a positive feedback loop controlling their activity.
