Summary Transepithelial Li+ influx was studied in the isolated epithelium from abdominal skin ofRana catesbeiana. With Na+-Ringer's as inside medium and Li+-Ringer's as outside medium, the Li+ influx across the epithelium was 15.6 A/cm2. This influx was considerably reduced by removal of either Na+ or K+ from the inside bath or by the addition of ouabain or amiloride. Epithelial K+ or Na+ concentration was respectively lower in epithelia bathed in K+-free Ringer's or Na+-free Ringer's. In conditions of negligible Na+ transport, a 20mm Li+ gradient (outin) produced across the short-circuited epithelium a Li+ influx of 11.8 A/cm2 and a mean short-circuit current of 10.2 A/cm2. The same Li+ gradient in the opposite direction produced a Li+ outflux of only 1.9 A/cm2. With equal Li+ concentration (10.3 and 20.6mm) on both sides of the epithelium, plus Na+ in the inside solution only, a stable Li+-dependent short-circuit current was observed. Net Li+ movement (outin) was also indirectly determined in the presence of an opposing Li+ gradient. Although Li+ does not substitute for Na+ as an activator of the (Na++K+)-ATPase from frog skin epithelium, Li+ influx appears to be related to Na+–K+ pump activity. It is proposed that the permeability of the outer barrier to Na+ and Li+ is regulated by the electrical gradient produced by electrogenic Na+–K+ pumps located in the membrane of the deeper epithelial cells. 相似文献

A cDNA clone, pMA1949, detects two mRNA species in wheat seedling tissue that are late embryogenesis-abundant (LEA) and dehydration stress-inducible. Sequence analysis of the pMA1949 clone shows it to be a 991 bp partial cDNA encoding a polypeptide of 317 amino acids with homology to two group 3 LEA proteins, carrot (DC8) and a soybean protein encoded by pGmPM2 cDNA. Molecular analysis of the deduced protein reveals a 33 kDa acidic and extremely hydrophilic protein with potential amphiphilic -helical regions. In addition, the protein contains eleven similar, contiguous repeats of 11 amino acids, which are separated by 118 amino acids from two additional and unique repeats of 36 residues each at the carboxyl end of the protein. Comparisons of sequences of reported group 3 LEA proteins revealed that there are two types, separable by sequence similarity of the 11 amino acid repeating motifs and by the presence or absence of a certain amino acid stretch at the carboxyl terminus. In this work we have combined biochemical and electrophysiological approaches to explore the modulation of rat ventricular transient outward K(+) current (I(to)) by calmodulin kinase II (CaMKII). Intracellular application of CaMKII inhibitors KN93, calmidazolium, and autocamtide-2-related inhibitory peptide II (ARIP-II) accelerated the inactivation of I(to), even at low [Ca(2+)]. In the same conditions, CaMKII coimmunoprecipitated with Kv4.3 channels, suggesting that phosphorylation of Kv4.3 channels modulate inactivation of I(to). Because channels underlying I(to) are heteromultimers of Kv4.2 and Kv4.3, we have explored the effect of CaMKII on human embryonic kidney (HEK) cells transfected with either of those Kvalpha-subunits. Whereas Kv4.3 inactivated faster upon inhibition of CaMKII, Kv4.2 inactivation was insensitive to CaMKII inhibitors. However, Kv4.2 inactivation became slower when high Ca(2+) was used in the pipette or when intracellular [Ca(2+)] ([Ca(2+)](i)) was transiently increased. This effect was inhibited by KN93, and Western blot analysis demonstrated Ca(2+)-dependent phosphorylation of Kv4.2 channels. On the contrary, CaMKII coimmunoprecipitated with Kv4.3 channels without a previous Ca(2+) increase, and the association was inhibited by KN93. These results suggest that both channels underlying I(to) are substrates of CaMKII, although with different sensitivities; Kv4.2 remain unphosphorylated unless [Ca(2+)](i) increases, whereas Kv4.3 are phosphorylated at rest. In addition to the functional impact that phosphorylation of Kv4 channels could cause on the shape of action potential, association of CaMKII with Kv4.3 provides a new role of Kv4.3 subunits as molecular scaffolds for concentrating CaMKII in the membrane, allowing Ca(2+)-dependent modulation by this enzyme of the associated Kv4.2 channels. 相似文献

Two experiments were conducted to evaluate the effects of the administration of 125 mg rbST at the time of breeding on fertile and subfertile superovulated ewes – in terms of the ovulation rate, embryo yield and embryonic development. In addition, the effects of the treatment on the plasma progesterone concentrations and interferon-tau (IFNT) expression in the embryos were evaluated in the fertile animals. In the first experiment, carried out on fertile superovulated ewes, the treatment significantly increased the ovulation rate (p < 0.05) and shifted the embryo distribution towards more advanced stages of development (p < 0.01). Plasma progesterone concentrations increased faster in the treated ewes, being significantly different (p < 0.05) between groups at day 6 post-breeding. In the second experiment, embryonic development was more advanced in fertile, but not in subfertile superovulated ewes. The expression of IFNT was significantly (p < 0.01) decreased in blastocyst embryos obtained from rbST treated animals, compared with non-treated ewes. It is concluded that the administration of rbST to subfertile ewes at the time of breeding does not stimulate embryonic development as it does when administered to fertile animals. 相似文献

No-choice cage tests were used to study the toxicity of imidacloprid-treated spheres to Caribbean fruit fly, Anastrepha suspensa (Loew), and its associated parasitoid, Diachasmimorpha longicaudata (Ashmead), in the laboratory. Three imidacloprid sphere treatments (2, 4, and 8% active ingredient [AI] Provado 1.6 F) and an untreated control sphere (no toxicant) were evaluated against A. suspensa. Throughout the observation period (2-72 h), all concentrations of imidacloprid-treated spheres killed significantly more A. suspensa compared with control spheres. After 4 h of exposure to imidacloprid-treated spheres, significantly more A. suspensa were killed on spheres treated with 8% compared with 2% (AI). At 48 and 72 h, there were no significant differences in the mean number of A. suspensa killed at 2, 4, and 8% (AI), potentially indicating that a period of 24 h was sufficient for flies to ingest a lethal dose of the pesticide. Overall, significantly more A. suspensa males were killed after 72 h of exposure to imidacloprid-treated spheres compared with females. For D. longicaudata, only two imidacloprid sphere treatments, 2 and 4% (AI), and an untreated sphere (control) were evaluated for mortality in cage tests. There were no significant differences in mortality of D. longicaudata between the 2 and 4% (AI) imidacloprid-treated spheres. Both rates killed significantly more D. longicaudata compared with the control. However, after 24, 48, and 72 h of exposure to imidacloprid-treated spheres, significantly more D. longicaudata were killed in cages containing 4% compared with 2% (AI) and untreated control spheres. The study demonstrates the potential use of imidacloprid-treated spheres for control of A. suspensa in areas where it may be difficult to apply broad-spectrum insecticides. 相似文献

BackgroundKnowing the prevalence of schistosomiasis is key to informing programmes to control and eliminate the disease as a public health problem. It is also important to understand the impact of infection on child growth and development in order to allocate appropriate resources and effort to the control of the disease.MethodsWe conducted a survey to estimate the prevalence of schistosomiasis among school aged children in villages along the Albert-Nile shore line in the district of Pakwach, North Western Uganda. A total of 914 children aged between 10–15 years were screened for Schistosoma mansoni using the POC-CCA and Kato Katz (KK) techniques. The infection intensities were assessed by POC-CCA and KK as well as CAA tests. The KK intensities were also correlated with POC-CCA and with CAA intensity. Anthropometric measurements were also taken and multivariate analysis was carried out to investigate their association with infection status.ResultsThe prevalence of schistosomiasis using the POC-CCA diagnostic test was estimated at 85% (95% CI: 83–87), being highest amongst children living closer to the Albert-Nile shoreline. Visual scoring of the POC-CCA results was more sensitive than the Kato Katz test and was positively correlated with the quantified infection intensities by the CAA test. The majority of the children were underweight (BMI<18.5), and most notably, boys had significantly lower height for age (stunting) than girls in the same age range (p < 0.0001), but this was not directly associated with S. mansoni infection.ConclusionHigh prevalence of S. mansoni infection in the region calls for more frequent mass drug administration with praziquantel. We observed high levels of stunting which was not associated with schistosomiasis. There is a need for improved nutrition among the children in the area. 相似文献

Background
Pulmonary hypertension is characterized by increased pulmonary artery pressure and carries an increased mortality. Population-based studies into pulmonary hypertension are scarce and little is known about its prevalence in the general population. We aimed to describe the distribution of echocardiographically-assessed pulmonary artery systolic pressure (ePASP) in the general population, to estimate the prevalence of pulmonary hypertension, and to identify associated factors.Methods
Participants (n = 3381, mean age 76.4 years, 59% women) from the Rotterdam Study, a population-based cohort, underwent echocardiography. Echocardiographic pulmonary hypertension was defined as ePASP>40 mmHg.Results
Mean ePASP was 26.3 mmHg (SD 7.0). Prevalence of echocardiographic pulmonary hypertension was 2.6% (95%CI: 2.0; 3.2). Prevalence was higher in older participants compared to younger ones (8.3% in those over 85 years versus 0.8% in those between 65 and 70), and in those with underlying disorders versus those without (5.9% in subjects with COPD versus 2.3%; 9.2% in those with left ventricular systolic dysfunction versus 2.3%; 23.1% in stages 3 or 4 left ventricular diastolic dysfunction versus 1.9% in normal or stage 1). Factors independently associated with higher ePASP were older age, higher BMI, left ventricular diastolic dysfunction, COPD and systemic hypertension.Conclusion
In this large population-based study, we show that pulmonary hypertension as measured by echocardiography has a low prevalence in the overall general population in the Netherlands, but estimates may be higher in specific subgroups, especially in those with underlying diseases. Increased pulmonary arterial pressure is likely to gain importance in the near future due to population aging and the accompanying prevalences of underlying disorders. 相似文献660.

Recognition of DNA double-strand breaks during non-homologous end joining is carried out by the Ku70-Ku80 protein, a 150 kDa heterodimer that recruits the DNA repair kinase DNA-dependent protein kinase catalytic subunit (DNA-PKcs) to the lesion. The atomic structure of a truncated Ku70-Ku80 was determined; however, the subunit-specific carboxy-terminal domain of Ku80--essential for binding to DNA-PKcs--was determined only in isolation, and the C-terminal domain of Ku70 was not resolved in its DNA-bound conformation. Both regions are conserved and mediate protein-protein interactions specific to mammals. Here, we reconstruct the three-dimensional structure of the human full-length Ku70-Ku80 dimer at 25 A resolution, alone and in complex with DNA, by using single-particle electron microscopy. We map the C-terminal regions of both subunits, and their conformational changes after DNA and DNA-PKcs binding to define a molecular model of the functions of these domains during DNA repair in the context of full-length Ku70-Ku80 protein. 相似文献
