Two COS mixtures and a low molecular weight chitosan (LMWC) were tested for potential cytotoxicity and genotoxicity upon human lymphocytes. Genotoxicity was evaluated in vitro by cytokinesis-blocked micronucleus and alkaline comet assays, while cytotoxicity was assessed by flow cytometry analysis. Our results suggest that COS do not exhibit any genotoxicity upon human lymphocytes, independently of MW or concentration. However, above 0.07 mg/mL COS induced strong cytotoxic effects. According to the concentration used, such cytotoxicity will induce cell death, essentially by necrosis (>0.10 mg/mL) and/or apoptosis (<0.10 mg/mL). The level of necrosis/apoptosis induced by high COS concentrations, suggests a promising use as apoptosis inducers in specific cancer situations. 相似文献

The importance of the tongue during feeding, and the limited information on the tongue of most aquatic mammals led us to investigate its morphological aspects in sexually immature and mature Sotalia guianensis. Six tongues were measured and photo-documented after their removal from the oral cavity. The samples were divided into rostral, middle, and caudal regions, and examined using light microscopy and scanning electron microscopy (S.E.M.). Sotalia guianensis tongue presented lateral grooves from the apex to the middle portion, while the anterolateral region presented marginal papillae. Histological characteristics revealed the presence of a keratinized stratified epithelium, salivary glands in the middle and caudal portions of the tongue, and filiform papillae in the caudal region. S.E.M. images revealed the presence of filiform papillae and ducts of salivary glands in the middle and caudal portions of the tongue. We can conclude that the characteristics found in this study may reflect an adaptation to changes in diet after weaning. 相似文献

The aims of this work were to quantify (i) the effect of the source:sink ratio on stem water potential (SWP) and (ii) the phenotypic plasticity of SWP and its relationship to oil yield components in olive. Trees with a 3-fold variation in the source:sink ratio (crown volume/fruit number per tree) were monitored in 2007-2008 and 2008-2009 in a fully irrigated orchard in Mendoza, Argentina. The combination of rainfall, irrigation, and evaporative demand led to a steady SWP largely above -1.65 MPa in 2007-2008 and a marked seasonal decline from -1.13 MPa to -2.04 MPa in trees with a medium and low source:sink ratio in 2008-2009. Plasticity was quantified as the slope of the norm of reaction for each trait. Across seasons, trees with a high source:sink ratio had a higher SWP than their counterparts with a medium and low source:sink ratio. Plasticity of SWP was highest in olives with a low source:sink ratio (slope=1.28) and lowest for trees with a high source:sink ratio (slope=0.76). The average SWP for each source:sink ratio and season was unrelated to both the source:sink ratio and yield components. On the other hand, the plasticity of SWP was positively associated with fruit number and negatively associated with the source:sink ratio, fruit weight, and fruit oil weight. The plasticity of the SWP was unrelated to SWP per se. It is concluded that understanding the effect of the source:sink ratio on plant water relations would benefit from a dual perspective considering the trait per se and its plasticity. Phosphoinositides control many different processes required for normal cellular function. Myotubularins are a family of Phosphatidylinositol 3-phosphate (PtdIns3P) phosphatases identified by the positional cloning of the MTM1 gene in patients suffering from X-linked myotubular myopathy and the MTMR2 gene in patients suffering from the demyelinating neuropathy Charcot-Marie-Tooth disease type 4B. MTM1 is a phosphatidylinositol phosphatase with reported specificity toward PtdIns3P, while the related proteins MTMR2 and MTMR3 hydrolyze both PtdIns3P and PtdIns(3,5)P2. We have investigated MTM1 and MTMR6 and find that they use PtdIns(3,5)P2 in addition to PtdIns3P as a substrate in vitro. The product of PtdIns(3,5)P2 hydrolysis, PtdIns5P, causes MTM1 to form a heptameric ring that is 12.5 nm in diameter, and it is a specific allosteric activator of MTM1, MTMR3, and MTMR6. A disease-causing mutation at arginine 69 of MTM1 falling within a putative pleckstrin homology domain reduces the ability of the enzyme to respond to PtdIns5P. We propose that the myotubularin family of enzymes utilize both PtdIns3P and PtdIns(3,5)P2 as substrates, and that PtdIns5P functions in a positive feedback loop controlling their activity. The adaptation to utilise lactose as primary carbon and energy source is a characteristic for Streptococcus thermophilus. These organisms, however only utilise the glucose moiety of lactose while the galactose moiety is excreted into the growth medium. In this study we evaluated the diversity of sugar utilisation and the conservation of the gal-lac gene cluster in a collection of 18 S. thermophilus strains isolated from a variety of sources. For this purpose analysis was performed on DNA from these isolates and the results were compared with those obtained with a strain from which the complete genome sequence has been determined. The sequence, organisation and flanking regions of the S. thermophilus gal-lac gene cluster were found to be highly conserved among all strains. The vast majority of the S. thermophilus strains were able to utilize only glucose, lactose, and sucrose as carbon sources, some strains could also utilize fructose and two of these were able to grow on galactose. Molecular characterisation of these naturally occurring Gal+ strains revealed up-mutations in the galKTE promoter that were absent in all other strains. These data support the hypothesis that the loss of the ability to ferment galactose can be attributed to the low activity of the galKTE promoter, probably as a consequence of the adaptation to milk in which the lactose levels are in excess. 相似文献

Ferocactus histrix is a barrel cactus that is widespread in Mexico. A population located in Llanos de Ojuelos, a semiarid zone representative of many disturbed regions in north‐central Mexico, was studied. Over a period of 10 years (1997 to 2007), the average number of individuals decreased from 21.95 to 3.53 plants per 300 m2. A change in population size structure was also registered over this period of time. In 2008, a plot selected on the basis of plant abundance was established within the population and a genetic analysis was conducted with ISTR and ISSR markers. This analysis revealed low levels of genetic diversity (expected heterozygosity (HE) = 0.073, Shannon index (I) = 0.113 and HE = 0.178, I = 0.271, respectively) compared with those of most studied cacti species. The genetic diversity between the different life stages was also evaluated, and a gradual decrease in levels of genetic variation was observed from adults to juveniles and seedlings (HE = 0.130, I = 0.192 to HE = 0.103, I = 0.157). These differences, however, were not significant. Loci fixation and a decrease in the frequency of rare alleles were observed in seedling and juvenile classes. The decline in genetic variation may be associated with recent bottlenecks experienced by the population of F. histrix. If the sizes of local populations of F. histrix continue to decrease, genetic variation will be gradually lost, and the risk of extinction will increase. 相似文献

Peru is the top exporter of fishmeal and fish oil (FMFO) worldwide and is responsible for half and a third of global production, respectively. Landings of “anchoveta” (Engraulis ringens) are used nearly exclusively for FMFO production, despite a proactive national food policy aimed at favoring the direct human consumption of this inexpensive species. It may be surprising that in a country where malnutrition and caloric deficit constitute major issues, a low-priced and highly nutritious fish such as anchovy does not have stronger domestic demand as a food fish. Here, we review and assess eight potential politico-socio-economic processes that can explain this situation. The main explanation are dietary habits, the preference for broiler and the higher profit from anchovy sold as feed fish compared to its use as a food fish due to historically high FMFO prices, boosted by an increasing demand for aquaculture in a context of finite forage and trash fish resources. In addition, the recent introduction of an individual quota system has shifted bargaining power from processors to fishers, thereby increasing competition for the raw material. This competition results in an increase in anchovy prices offered by the feed fish industry due to its onshore processing overcapacity, which is detrimental to the food fish industry. In the end, although the dominant use of anchovy for fish feed is largely explained by integrating these market mechanisms and other minor ones, this use raises other issues, such as rent redistribution through public policies, employment, equitability and utility (low social costs), and resource management (threats to ecosystems or global change).
