Summary A study was made of substance P-like-immunoreactive nerves within the small intestine of platypus, an Australian prototherian mammal. Both immunoreactive nerve cell bodies and fibres were present. No immunoreactive fibres were found in mesenteric nerves or on intramural blood vessels, suggesting that extrinsic sensory neurons containing substance P do not innervate platypus ileum. This was further supported by the result of in vitro experiments. Although applied substance P (10-9 M-3 × 10-8 M) caused contraction of the longitudinal muscle, neither mesenteric nerve stimulation nor application of capsaicin caused contractions. 相似文献

An in vitro human reconstructed epidermis model (SkinEthic) used for screening acute and chronic skin irritation potential
was validated against in vivo data from skin tolerability studies. The irritation potential of sodium lauryl sulfate (SLS),
calcipotriol and trans-retinoic acid was investigated. The in vitro epidermis-like model consists of cultures of keratinocytes
from human foreskin on a polycarbonate filter. The modulation of cell viability, the release and gene expression of proinflammatory
cytokines, interleukins 1α and 8, and morphological changes were evaluated during 3 days as endpoints representative for an
inflammatory reaction. The cumulative irritation potential of the topical products was evaluated in a human clinical study
by visual scoring and biophysical measurement of inflammatory skin reaction after repeated 24 h applications over 3 weeks
under Finn chamber patches. All topical products that were nonirritating in the human study were noncytotoxic and did not
induce cytokine expression in the in vitro acute model (day 1 exposure). All irritating controls exhibited specific cell viability
and cytokine patterns, which were predictive of the in vivo human data. The ranking of mild to moderate skin irritation potential
was based on the lack of cytotoxicity and the presence of cytokine patterns including gene expression specific for each irritant,
using the chronic in vitro model (up to 3 days exposure).
The human reconstructed epidermis model SkinEthic was shown to be a reliable preclinical tool predicting the irritation potential
of topical products. Moreover, it is a useful model in a two-step tiered strategy for screening acute and chronic irritation
potential for the selection of vehicles for new topical drugs.

The accumulation of histone H1 ° (also denoted IP 25) in murine erythroleukemia cells, induced to differentiate with hexamethylene bis-acetamide, was shown to precede by 15–20 h the appearance in the culture of cells irreversibly committed to differentiate. In addition the rates of accumulation of H1 ° and of committed cells vary in a similar manner with the HMBA concentration. Flow microfluorimetric analysis demonstrated that the accumulation of H1 ° did not occur simultaneously in all the cells. This accumulation of histone H1 ° was initiated first in cell in the G2 phase of the cell cycle and subsequently in the cells situated in all the phases of the cell cycle. 相似文献

Cytochrome bd is one of the two quinol oxidases in the respiratory chain of Escherichia coli. The enzyme contains three heme prosthetic groups. The dioxygen binding site is heme d, which is thought to be part of the heme-heme binuclear center along with heme b(595), which is a high-spin heme whose function is not known. Protein sequence alignments [Osborne, J. P., and Gennis, R. B. (1999) Biochim. Biophys Acta 1410, 32--50] of cytochrome bd quinol oxidase sequences from different microorganisms have revealed a highly conserved sequence (GWXXXEXGRQPW; bold letters indicate strictly conserved residues) predicted to be on the periplasmic side of the membrane between transmembrane helices 8 and 9 in subunit I. The functional importance of this region is investigated in the current work by site-directed mutagenesis. Several mutations in this region (W441A, E445A/Q, R448A, Q449A, and W451A) resulted in a catalytically inactive enzyme with abnormal UV--vis spectra. E445A was selected for detailed analysis because of the absence of the absorption bands from heme b(595). Detailed spectroscopic and chemical analyses, indeed, show that one of the three heme prosthetic groups in the enzyme, heme b(595), is specifically perturbed and mostly missing from this mutant. Surprisingly, heme d, while known to interact with heme b(595), appears relatively unperturbed, whereas the low-spin heme b(558) shows some modification. This is the first report of a mutation that specifically affects the binding site of heme b(595). 相似文献
