Synopsis Before the decline of the species flock of haplochromine cichlids of Lake Victoria due to the Nile perch upsurge, there were many co-existing haploehromine species such as the taxonomically and ecologically well-studied zooplanktivores of the Mwanza Gulf. In spite of the scarcely separated niches of some of these species, no sign of competition for space or food could be demonstrated. As is argued in this paper, optical differentiation could well be an aspect of adaptive radiation of these zooplanktivores, particularly among the highly sympatric species. Our hypothesis is based on the morphological modifications of retinal structures in nine zooplanktivorous species. Interspecific variation was observed in composition, size and density of the photoreceptors and ganglion cells. The analyses included the intraretinal variation and size dependency of some of the structural parameters. The optical functions deduced from retinal structure indicate distinct interspecific differences in sensitivity thresholds and a slight differentiation in visual resolution. These functions correlate poorly with the photic conditions of the species-specific habitats. The optical properties can, on the other hand, be connected with the more subtle differentiation in food items and feeding behaviour among these species. It is our concluding hypothesis, that the optical differentiation among the haplochromine zooplanktivores primarily served resource partitioning by different modes of visual prey detection rather than niche partitioning by habitat. 相似文献

The binuclear copper active site of Carcinus hemocyanin has been reconstituted by incubating apohemocyanin with Cu(I) in the presence of Br- ions. At constant Cu(I) concentration the kinetics of reconstitution depends on both pH and Br- concentration. The process is faster at pH 6.0 than at pH 7.0 and in both cases the reaction is accelerated by increasing Br- concentration from 0.1 M to 0.4 M. At pH 6.0 a time-dependent inactivation of the O2-binding properties of reconstituted hemocyanin is observed. This effect is attributed to a perturbation in the active site microenvironment caused by unspecifically bound copper. Br- ions show a protective effect probably by chelating excess metal. 相似文献

The human cytomegalovirus-encoded glycoproteins US2 and US11 target newly synthesized major histocompatibility complex class I heavy chains for degradation by mediating their dislocation from the endoplasmic reticulum back into the cytosol, where they are degraded by proteasomes. A functional ubiquitin system is required for US2- and US11-dependent dislocation of the class I heavy chains. It has been assumed that the class I heavy chain itself is ubiquitinated during the dislocation reaction. To test this hypothesis, all lysines within the class I heavy chain were substituted. The lysine-less class I molecules could no longer be dislocated by US2 despite the fact that the interaction between the two proteins was maintained. Interestingly, US11 was still capable of dislocating the lysine-less heavy chains into the cytosol. Ubiquitination does not necessarily require lysine residues but can also occur at the N terminus of a protein. To investigate the potential role of N-terminal ubiquitination in heavy chain dislocation, a lysine-less ubiquitin moiety was fused to the N terminus of the class I molecule. This lysine-less fusion protein was still dislocated in the presence of US11. Ubiquitination could not be detected in vitro, either for the lysine-less heavy chains or for the lysine-less ubiquitin-heavy chain fusion protein. Our data show that although dislocation of the lysineless class I heavy chains requires a functional ubiquitin system, the heavy chain itself does not serve as the ubiquitin acceptor. Background
Francisella tularensis is a highly virulent facultative intracellular bacterium, disseminating in vivo mainly within host mononuclear phagocytes. After entry into macrophages, F. tularensis initially resides in a phagosomal compartment, whose maturation is then arrested. Bacteria escape rapidly into the cytoplasm, where they replicate freely. We recently demonstrated that nucleolin, an eukaryotic protein able to traffic from the nucleus to the cell surface, acted as a surface receptor for F. tularensis LVS on human monocyte-like THP-1 cells.Methodology/Principal Findings
Here, we followed the fate of nucleolin once F. tularensis has been endocytosed. We first confirmed by siRNA silencing experiments that expression of nucleolin protein was essential for binding of LVS on human macrophage-type THP-1 cells. We then showed that nucleolin co-localized with intracellular bacteria in the phagosomal compartment. Strikingly, in that compartment, nucleolin also co-localized with LAMP-1, a late endosomal marker. Co-immunoprecipation assays further demonstrated an interaction of nucleolin with LAMP-1. Co-localization of nucleolin with LVS was no longer detectable at 24 h when bacteria were multiplying in the cytoplasm. In contrast, with an iglC mutant of LVS, which remains trapped into the phagosomal compartment, or with inert particles, nucleolin/bacteria co-localization remained almost constant.Conclusions/Significance
We herein confirm the importance of nucleolin expression for LVS binding and its specificity as nucleolin is not involved in binding of another intracellular pathogen as L. monocytogenes or an inert particle. Association of nucleolin with F. tularensis during infection continues intracellularly after endocytosis of the bacteria. The preservative properties of thyme essential oil (3%) with a known composition were evaluated in two types of final formulations, suitable for use as pharmaceutical or cosmetic vehicles, by means of a standard challenge test proposed by the latest European Pharmacopoeia. The required preservation efficacy criteria were satisfied against the bacterial strains, against the yeast in one of the formulations, but not against the mould strain involved in this study. Interactions between the essential oil compounds and other factors present in the final formulation might have influenced the activity of this essential oil, leading to an incomplete satisfaction of the criteria. 相似文献

The roles of sodium-ATP (NaATP) and ferric-ATP complex (FeATP) on the proliferation and infiltration of lymphocytes have been
studied by evaluation of the hypertrophy and histopathologic examination of spleen and liver, as well of the modifications
in the elemental balance of iron, calcium, magnesium, and phosphorus. The results showed that in the implicated biochemical
processes, calcium and magnesium have a principal role. An in vitro study on a cell model has permited one to evaluate the
effects of deferoxamine, a known iron chelator and inhibitor of human lymphocyte proliferation, on FeATP-modified cellular
calcium fluxes. These findings showing a reduced 45Ca2+ uptake in the presence of deferoxamine appear to indicate that in vivo the chelation of blood-borne iron by ATP might play
