The chemotactic potential of SXWS peptides and the components of the extracellular domain of cytokine receptors were investigated in Tetrahymena as a functional index of substitution with different amino acids in the position 'X' of the tetrapeptide. Data obtained demonstrate that position X plays a special determining role in the ligand, SEWS and STWS possess extremely strong chemoattractant ability, and aromatic amino acids result in chemorepellent ligands. Diverse effects of structurally related molecules, for example, SNWS-SDWS, demonstrate a highly sensitive discrimination potential in the applied model system. Physicochemical characteristics (hydropathy, residue size, and solvent-exposed area) of the amino acids were correlated with the chemotactic activity. Data obtained by computer-assisted conformation analysis of SXWS peptides and the highly overlapping chemotactic effects of the investigated SXWS peptides as well as the presence of the amino acids in the 'X' position indicate that member 'X' of the SXWS sequence performs a special role in interactions with the chemotaxis receptors in the membrane.  相似文献

Anthracyclines are potent cytostatic drugs, the correct dosage being critical to avoid possible cardiac side effects. ABCC1 [ATP-binding cassette, sub-family C, member 1; also denoted as MRP1 (multidrug resistance-associated protein 1)] is expressed in the heart and takes part in the detoxification and protection of cells from the toxic effects of xenobiotics, including anthracyclines. Our objective was to search for associations between LV (left ventricular) function and single-nucleotide polymorphisms of the ABCC1 gene in children receiving anthracycline chemotherapy. Data of 235 paediatric patients with acute lymphoblastic leukaemia was analysed. Patients were followed-up by echocardiography (median follow-up 6.3 years). Nine polymorphisms in the ABCC1 gene were genotyped. The ABCC1 rs3743527TT genotype and rs3743527TT–rs246221TC/TT genotype combination were associated with lower LVFS (left ventricular fractional shortening) after chemotherapy. The results suggest that genetic variants in the ABCC1 gene influence anthracycline-induced LV dysfunction.  相似文献

Lake Velencei is a shallow soda lake with extensive reed coverage. In this study, the bacterial communities of reed (Phragmites australis (Cav.) Trin. ex Steudel) rhizomes from healthy and declining stands were compared. Inner and outer rhizome surfaces were sampled. Samples were plated and isolated in September 1998 and June 1999. Phenotypic data of 371 bacterial strains were used for cluster analysis. Identification of phena was based on partial 16S rDNA sequence analysis of representative strains. Healthy reed stand rhizomes in fall 1998 were dominantly colonised by facultatively fermentative organisms, like Erwinia billingiae, Aeromonas sobria, Pantoea agglomerans, and Pseudomonas azotoformans. In the June 1999 sample, mainly Kocuria rosea and various Bacillus spp. dominated. In declining stands of September 1998, a saprotrophic community was found: Acinetobacter spp., Aeromonas hydrophila, Curtobacterium luteum, Agrobacterium vitis, and two further groups representing presumably new taxa. In June 1999, reed rhizomes were colonised by Kocuria rosea, but Dietzia maris and Bacillus cohnii could be isolated as well. Healthy and declining reed stand rhizomes can be distinguished based on the culturable bacterial community. No obligately plant pathogenic bacteria were found, however the possibility of a local, opportunistic bacterial invasion can not be ruled out (e.g. Curtobacterium). The presence of potentially beneficial bacterial species was demonstrated in the healthy reed rhizome rhizosphere (e.g. Pseudomonas azotoformans, Pantoea agglomerans).

Currently available detectors for spores of Bacillus anthracis, the causative agent of anthrax, are inadequate for frontline use and general monitoring. There is a critical need for simple, rugged, and inexpensive detectors capable of accurate and direct identification of B. anthracis spores. Necessary components in such detectors are stable ligands that bind tightly and specifically to target spores. By screening a phage display peptide library, we identified a family of peptides, with the consensus sequence TYPXPXR, that bind selectively to B. anthracis spores. We extended this work by identifying a peptide variant, ATYPLPIR, with enhanced ability to bind to B. anthracis spores and an additional peptide, SLLPGLP, that preferentially binds to spores of species phylogenetically similar to, but distinct from, B. anthracis. These two peptides were used in tandem in simple assays to rapidly and unambiguously identify B. anthracis spores. We envision that these peptides can be used as sensors in economical and portable B. anthracis spore detectors that are essentially free of false-positive signals due to other environmental Bacillus spores.  相似文献

Three new iridoid glycosides, named involucratosides A – C ( 1  –  3 ), were isolated from the H2O subextract of crude MeOH extract prepared from the aerial parts of Asperula involucrata along with a known iridoid glycoside (adoxoside), three flavone glycosides (apigenin 7‐Oβ‐glucopyranoside, luteolin 7‐Oβ‐glucopyranoside, apigenin 7‐O‐rutinoside) as well as two phenolic acid derivatives (chlorogenic acid and ferulic acid 4‐Oβ‐glucopyranoside). Their chemical structures were established by UV, IR, 1D‐ (1H, 13C and JMOD) and 2D‐ (COSY, HSQC, HMBC and NOESY) NMR experiments and HR‐ESI‐MS. In addition, the crude extract, subextracts and isolates were evaluated for their xanthine oxidase inhibitory and antioxidant activities in in vitro tests. This is the first report on the chemical composition and bioactivities of Ainvolucrata.  相似文献

Analysis of the monosaccharide composition of purified cell walls of unicellular and filamentous ascomycetous fungi shows three patterns: (1) the mannose glucose type (for most hemiascomycetous yeasts) (2) the mannose glucose galactose type (for several members of all three main ascomycetous clades) and (3) the mannose glucose galactose rhamnose type (for members of the Euascomycetes and the Protomyces/ Schizosaccharomyces group).In order to estimate the usefulness of the carbohydrate patterns for phylogenetic analysis we compared them with a phylogenetic tree based on 18SrRNA-gene sequences using the Neighbor-Joining Method. In contrast with the situation for basidiomycetous fungi, the Ascomycota show no fixed cell wall type for the three classes. Based on cell wall carbohydrates, sequence data and molecular characters the Hemiascomycetes appear as the first branch within the Ascomycota. A second clade, comprising the genera Schizosaccharomyces, Pneumocystis, Taphrina, Protomyces, Neolecta and Saitoella, appears as a sister group of the Euascomycetes. We discuss the erection of a new class for this group of ascomycetous fungi for which we propose the name Protomycetes.  相似文献

Germ cell testicular cancers are well-curable neoplasms, because total remission can be achieved in about 80% of the cases. However, 15-20% of the patients die due to drug resistance (DR). A number of mechanisms of the multidrug resistance phenotype are known, including MDR/P-glycoprotein (P-gp) and the so-called multidrug resistance associated protein (MRP). Lung Resistance Protein (LRP) is an ATP dependent membrane transporter protein associated with MDR. In our present work we studied the expression of LRP in testicular cancers. LRP expression was determined by immunohistochemistry (IH), Western blot (WB) and RT-PCR techniques. Clinical resistance was defined in accordance with the clinical oncologic rules. In 29 (41%) of 70 primary testicular tumours and in 22 (63%) of 35 cases, elevated LRP levels were established by IH and WB, respectively. In the latter 63%, the LRP mRNA levels were elevated as well. Six cases of the 15 seminomas and 23 cases of the nonseminomatous germ cell tumours (NSGCT) proved to be positive. No relationship was demonstrated between LRP expression and the stage of the disease. Despite the LRP positivity of 6 tumour samples, all of the seminomas proved sensitive. Of the 39 sensitive NSGCT, 27 cases were LRP-negative, whereas 11 tumour samples of 16 patients belonging to the resistant group proved LRP-positive (p=0.04). The authors concluded that the expression of LRP is responsible for clinical drug resistance in non-seminomatous testicular cancer patients.  相似文献
