Detachment Activity but Not Cytotoxicity Induced in a T-Cell Clone Following Antigen Presentation in the Presence of Thymic Epithelial Cells

The selective induction of effector functions of a T-cell clone (DB14), specific to pigeon cytochrome c 43–58 (p 43–58) and restricted to I-A^b, was analyzed using a professional antigen-presenting cell, B hybridoma (Th 2.58), and various non-professional antigen-presenting cells (APC), L cells transfected with I-A^b (I-A^b L cells), a medullary thymic epithelial cell line (m-TEC) and a cortical thymic epithelial cell line (c-TEC). The m-TEC and c-TEC expressed I-A^b upon induction with interferon γ (IFN-γ). When stimulated with p 43–58 in the presence of I-A^b L cells as well as Th 2.58 cells, the DB14 cells showed marked proliferation and, after 18 hr of culturing, exhibited significant cytotoxicity against the APC. By contrast, in the presence of m, c-TEC, the DB14 cells showed neither proliferation nor cytotoxicity against these TEC but exhibited considerable detachment activity towards them. Furthermore, DB14 cells became expressed activation markers (CD69 or CD44) following stimulation with p 43–58 plus m-TEC or c-TEC. The addition of rIL-2 to the culture of DB14 cells, p 43–58 and m-TEC or c-TEC, restored the proliferative responses. However, it was shown that anergy was not involved in the negligible proliferative responses of DB14 cells after stimulation with p 43–58 plus m, c-TEC. The present findings indicate that differences in APC functions are present among non-professional APC and suggest that the selective induction of T-cell functions can be achieved using the appropriate non-professional APC. The characteristic activation of T cells by TEC may be related to their functional roles in situ.     

Prior induction of heme oxygenase-1 with glutathione depletor ameliorates the renal ischemia and reperfusion injury in the rat

Heme oxygenase (HO)-1 catalyzes the rate-limiting step in heme degradation releasing iron, carbon monoxide, and biliverdin. Induction of HO-1 occurs as an adaptive and protective response to oxidative stress. Ischemia and reperfusion (IR) injury seems to be mainly caused by the oxidative stress. In this study, we have examined whether prior induction of HO-1 with buthionine sulfoximine (BSO), a glutathione (GSH) depletor, affects the subsequent renal IR injury. BSO (2 mmol/kg body weight) was administered intraperitoneally into rats, the levels of HO-1 protein increased within 4 h after the injection. When BSO was administered into rats at 5 h prior to the renal 45 min of ischemia, the renal IR injury was assessed by determining the levels of blood urea nitrogen and serum creatinine, markers for renal injury, after 24 h of reperfusion. The renal injury was significantly improved as compared to the rats treated with IR alone. Administration of zinc-protoporphyrin IX, an inhibitor of HO activity, reduced the efficacy of BSO pretreatment on the renal IR injury. Our findings suggest that the prior induction of HO-1 ameliorates the subsequent renal IR injury.     

Expression of Activin Receptor-like Kinase 7 in Adipose Tissues

The tissue distribution of activin receptor-like kinase 7 (Alk7) expression, the signaling ability of Alk7 variants, and Alk7 expression in response to β₃-adrenergic receptor activation were examined. Expression levels of Alk7 varied greatly among tissues but were highest in white adipose tissue and brown adipose tissue. In addition to full-length Alk7 (Alk7-v1), Alk7-v3, an Alk7 variant, was expressed in adipose tissues, brain, and ovary. Nodal transmits signals via Alk7 in cooperation with its coreceptor, Cripto. Evaluation of the ability of Alk7 variants to confer Nodal signaling using luciferase-based reporter assays showed that Alk7-v3 does not transmit Nodal-Cripto-mediated signals. Expression of Alk7 was down-regulated in brown but not in white adipose tissue treated with CL316,243, a β₃-adrenergic receptor agonist. These results suggest involvement of Alk7 in modulation of metabolism in the adipose tissues in response to β₃-adrenergic receptor activation.     

Wild host plants of four spider mite species (Acari: Tetranychidae) infesting fruit crops in Okinawa

To determine the wild host plants of four major spider mite species infesting fruit crops grown in the subtropical Asian region, we collected and identified mites from non-crop plants throughout the islands of Okinawa, southwestern Japan. Although the two mango pests, Oligonychus coffeae and O. biharensis, are polyphagous, they did not share any wild host species in the field, indicating that their source plants are completely separate in Okinawa. Several major wild hosts were determined for Eutetranychus africanus, a pest of papaya and citrus. Its host range partially overlapped with that of O. biharensis. The citrus red mite, Panonychus citri, was very rare on non-crop plants in Okinawa, suggesting that it maintains its population mainly on the citrus trees in the area. These results are of great significance when considering vegetation control as part of the integrated management of these pest mites. During our survey, two non-pest species, Panonychus caglei (new to Japan) and Oligonychus gotohi (new to Okinawa), were also found.
