Patterns of reproduction were investigated in some microalgal species of Chlorophyceae (Botryosphaerella sudetica, Neochloris aquatica, Neochloris vigensis, Bracteacoccus minor). Under continuous light, the microalgae reproduced asexually producing autospores. However, appropriate manipulation of external conditions led to a change in the reproduction pattern towards production of zoospores or gametes. Production of zoospores and gametes was inhibited by light; motile cells emerged when microalgae were cultivated in darkness. The period of dark treatment necessary for zoosporogenesis or gametogenesis differed substantially among species that were tested. Sexual reproduction was observed in Neochloris vigensis and Bracteacoccus minor, whose generative life cycle had not been previously reported. The morphology of motile cells, the mode of sexual reproduction, and the efficiency of both the production of motile stages and mating events, were investigated. In order to gain detailed insights into patterns of reproduction, Botryosphaerella sudetica was selected for investigation under different light treatments. Non-actinic red light applied in the early phase of dark cultivation (up to 2 h) suppressed both zoosporogenesis and gametogenesis. However, after a 3-h dark pre-treatment, red light treatment had no effect on zoosporogenesis or gametogenesis. In contrast, non-actinic blue light did not block zoosporogenesis or gametogenesis, regardless of the time of treatment. The possible role of a red-light photoreceptor in zoosporogenesis and gametogenesis is discussed.  相似文献

ABSTRACT

The Uroglena-like morphotype represents a prototype of a colonial naked chrysophyte, comprising plastid-bearing cells that are arranged as the surface monolayer of the spherical colony. So far, insufficient molecular characterization appears to be the most significant brake on the modern taxonomic revision of this ecologically and morphologically coherent group of organisms. The general aim of this work was to conduct a modern taxonomic revision of Uroglena-like flagellates by using combined molecular, morphological and ultrastructural methodology, complemented by exploring type localities of Uroglena volvox and Uroglenopsis americana in Europe and North America, respectively. On the basis of phylogenetic analysis of concatenated nuclear SSU rDNA and plastid rbcL sequences we show that Uroglena-like colonial flagellates form three genetically and morphologically distinct lineages within the Ochromonadales (Chrysophyceae), distinguished here as Uroglena, Uroglenopsis and Urostipulosphaera gen. nov. The taxonomic status of the other chrysophyte genera with spherical colonies is discussed in light of our findings.  相似文献

Botulinum neurotoxins (BoNTs) are the most lethal biotoxins known to mankind and are responsible for the neuroparalytic disease botulism. Current treatments for botulinum poisoning are all protein based and thus have a limited window of treatment opportunity. Inhibition of the BoNT light chain protease (LC) has emerged as a therapeutic strategy for the treatment of botulism as it may provide an effective post exposure remedy. Using a combination of crystallographic and modeling studies a series of hydroxamates derived from 1-adamantylacetohydroxamic acid (3a) were prepared. From this group of compounds, an improved potency of about 17-fold was observed for two derivatives. Detailed mechanistic studies on these structures revealed a competitive inhibition model, with a Ki = 27 nM, which makes these compounds some of the most potent small molecule, non-peptidic BoNT/A LC inhibitors reported to date.  相似文献

Larva of the fruit beetle Dicronocephalus wallichi bourgoini Pouillaude 1914 is described and illustrated. The following unique morphological characters within Cetoniinae are discussed: shape of claw, spindle-shaped last antennomera, general body shape. The presence of metathoracic egg-bursters in the first instar larva has been found. Its significance for distinguishing the first instar larvae of Cetoniinae is discussed. The nesting behaviours of D. wallichi bourgoini and D. adamsi(Pascoe 1863) are described. The course of the life cycle of both species under laboratory condition is presented and discussed.  相似文献

The alga Parachlorella kessleri, strain CCALA 255, grown under optimal conditions, is characterized by storage of energy in the form of starch rather than lipids. If grown in the complete medium, the cultures grew rapidly, producing large amounts of biomass in a relatively short time. The cells, however, contained negligible lipid reserves (1–10% of DW). Treatments inducing hyperproduction of storage lipids in P. kessleri biomass were described. The cultures were grown in the absence or fivefold decreased concentration of either nitrogen or phosphorus or sulfur. Limitation by all elements using fivefold or 10‐fold diluted mineral medium was also tested. Limitation with any macroelement (nitrogen, sulfur, or phosphorus) led to an increase in the amount of lipids; nitrogen limitation was the most effective. Diluted nutrient media (5‐ or 10‐fold) were identified as the best method to stimulate lipid overproduction (60% of DW). The strategy for lipid overproduction consists of the fast growth of P. kessleri culture grown in the complete medium to produce sufficient biomass (DW more than 10 g/L) followed by the dilution of nutrient medium to stop growth and cell division by limitation of all elements, leading to induction of lipid production and accumulation up to 60% DW. Cultivation conditions necessary for maximizing lipid content in P. kessleri biomass generated in a scale‐up solar open thin‐layer photobioreactor were described. Biotechnol. A series of new monophosphates of 1-[2-(phosphonomethoxy)alkyl]thymines, such as PMPTp, 3-MeO-PMPTp, HPMPTp, and FPMPTp, were synthesized and tested for their ability to inhibit human thymidine phosphorylase. Kinetic measurements of enzyme activity were performed using thymidine and inorganic phosphate as the substrates. The data show that some monophosphates provide a considerable increase of the multisubstrate inhibitory effect. The highest inhibitory potency was found with (R)-FPMPTp 4c (K i dT = 4.09 ± 0.47 μM, K i(Pi) = 2.13 ± 0.29 μM) and (R) 3-MeO-PMPTp 4d (K i dT = 5.78 ± 0.71 μM, K i(Pi) = 2.71 ± 0.37 μM).  相似文献

Abstract

Two subspecies of Cistus incanus L. occurring in the southern Balkans were studied: C. incanus L. ssp. incanus and C. incanus ssp. creticus (L.) Heywood. After studying the morphological differences, the communities dominated by these two subspecies were sampled according to the central European method. The localities of the relevés show the distribution pattern of the two subspecies. The typical subspecies can be found in the area of distribution of thermophilous deciduous forests of Carpinion orientalis, whereas the subspecies creticus grows in the area of the evergreen broad‐leaved forests of Quercetea ilicis. The communities were classified as Diantho–Cistetum incani Micevski et Matevski ex ?arni, Matevski et ?ilc ass. nova hoc loco and Calicotomo villosae–Cistetum cretici Oberdorfer 1954. Analyses of traits (life span, life form, proportion of certain families, bioindicator values and chorotypes) reveal that the harshest site conditions prevail within Diantho–Cistetum incani, which appears as an initial stage of the vegetation succession. Calicotome–Cistetum cretici is a subsequent stage of succession, since the extreme degradation of vegetation on these sites results in ephemeral communities dominated by Poa bulbosa assigned to Romulion.  相似文献
