Turnover times for water passing through several Sulfolobus acidocaldarius-containing springs were determined by measuring the dilution rates of small amounts of sodium chloride that were added to the springs. Chloride was diluted out exponentially, while concentrations of the bacteria remained constant. Additionally, temperature, pH, and chemical composition of the springs also remained constant during the time that the chloride was being diluted. The springs are thus steady-state systems, and since the rates of bacterial growth must be at least equal to the chloride dilution rates, minimal doubling times for the bacterial populations can be calculated. Half-times for chloride dilution, equivalent to bacterial doubling times, were on the order of 10 to 20 h for springs ranging in volume from about 20 to 2,000 liters, but approximately 30 days for two larger springs of about 1 million liters. Formaldehyde-fixed cells of a serologically distinguishable strain of S. acidocaldarius were also added as markers to four of the smaller springs, and the dilution rates of these bacteria were compared with the chloride dilution rates. The rates agreed reasonably well, thus verifying the growth rates obtained from the chloride dilution rates. In three springs, exponential growth was studied by draining the springs and allowing them to refill with bacteria-free water. Exponential doubling times were on the order of a few hours, much more rapid than steady-state doubling times. The methods used in this work may have wider utility in aquatic environments.  相似文献

A technique was developed for measuring (14)CO(2) uptake by chemolithotrophic bacteria directly in pyritic materials associated with coal and coal refuse. There was good correlation between (14)CO(2) uptake, as determined by this technique, and the most probable number of iron-oxidizing bacteria. Maximal (14)CO(2) uptake occurred in coal refuse material 2 to 3 years old, and only slight incorporation was demonstrated in fresh material or material 40 years old. Samples taken from the surface of the coal refuse pile always demonstrated maximal (14)CO(2) uptake, and in most samples, only slight activity was demonstrated at depths below 8 to 10 cm. Optimal uptake of (14)CO(2) by natural samples occurred at 20 to 30 C and at a moisture content of between 23 and 35%. In addition to chemolithotrophic bacteria, heterotrophic fungi and yeasts were also routinely isolated in high numbers from acidic coal refuse. In contrast, acidophilic, heterotrophic bacteria were either absent or present in low numbers in such acidic samples.  相似文献

Chromosomes and sex determination of 9 species of Haemaphysalis assigned to 4 subgenera are described. H. (tAlloceraea) kitaokai possesses an XX∶XO sex chromosome system with 18 autosomes plus XX in females; 18 plus X in males. H. (Kaiseriana) hystricis has 18 +XX and 18 + XY in females and males, respectively, in most specimens, but a supernumerary chromosome is present in some individuals. A supernumerary chromosome was also observed in 1 male H. (Aborphysalis) formosensis. These two species are the second and third species of ticks reported to have supernumerary chromosomes. H. formosensis, H. (Kaiseriana) bispinosa, H. (Haemaphysalis) campanulata, H. (H.) flava, H. (H.) megaspinosa, H. (H.) japonica, and H. (H.) pentalagi possess 20 autosomes plus 2 sex chromosomes in females and 20+1 sex chromosomes in males. Phylogenetic relationships within the genus Haemaphysalis are briefly discussed.  相似文献

The final phase of spermiogenesis in the hard tick Dermacentor andersoni occurs in the seminal receptacle of engorged females. This phase is characterized mainly by rupture of the outer sheath of advanced spermatids at their pointed tips, and subsequent elongation as the internal column of these cells evaginates. The sheath invaginates at the opposite end to form the narrow posterior half of the sperm cell. Rupture of the endospermatophoric wall releases the large mass of contained spermatozoa, which begin migration up one or both oviducts. During the migration to the ovarian lumen, spermatozoa exhibit a propensity for active penetration of epithelial cells lining the oviducts. This phenomenon is also observed with spermatozoa in the ovarian lumen. Penetration of epithelial cells by spermatozoa is interpreted to be a response by the latter in seeking eggs for penetration and fertilization.  相似文献

Phenol oxidase activity of activated A1 and A2 (A3) electrophoretic components from 19 lozenge mutant and three lozenge double mutant strains was compared to that of wild type flies. Melanin production by the activated A components with tyrosine as substrate was compared to activity in the same acrylamide gels with dopa as substrate. Melanin production decreased, first in the A1 band and then in the A2 (A3) band, with increased morphological expression of the mutant genes. No melanin bands were obtained with either substrate in five of the more severely affected mutants. A possible correlation between phenol oxidase activity and quinone production necessary for normal development of eyes, female accessory sex organs, and claws is discussed.Supported by PHS grant AM-08331-05.  相似文献

A species of Zygogonium forms extensive dark purple mats in Yellowstone National Park in acidic habitats adjacent to thermal areas. These mats range up to 6 cm in thickness and up to 3000 m2 in areal extent. Temperatures in the mats varied from 20–31 C and pH varied from 2.4–3.1. These mats form on soil in areas where a moist surface is created by the presence of small acidic springs or seeps. The effect of light, temperature, and pH on photosynthesis was studied in the field by use of 14CO2. Photosynthesis increased in rate up to full sunlight; light inhibition was not observed. Temperature optimum for photo-synthesis was 25 C. A broad pH optimum was found between 1.0 and 5.0. The Zygogonium mats have a high water holding capacity and create a moist habitat in which Euglena and Chlamydomonas develop. The mats also serve as repositories for the eggs of the brine fly Ephydra bruesi and both larvae and adults of this fly probably consume Zygogonium filaments as their main food source.  相似文献

The isolation of a new thermophilic bacterium, Thermus aquaticus gen. n. and sp. n., is described. Successful enrichment requires incubation at 70 to 75 C, and the use of nutrient media relatively dilute with respect to the organic components. Strains of T. aquaticus have been isolated from a variety of thermal springs in Yellowstone National Park and from a thermal spring in California. The organism has also been isolated from man-made thermal habitats, such as hot tap water, in geographical locations quite distant from thermal springs. Isolates of T. aquaticus are gram-negative nonsporulating nonmotile rods which frequently form long filaments at supraoptimal temperatures or in the stationary phase. All isolates form a yellow cellular pigment, probably a carotenoid. A characteristic structure formed by all isolates is a large sphere, considerably larger than a spheroplast. These large spheres, as well as lysozyme-induced spheroplasts, are resistant to osmotic lysis. Deoxyribonucleic acid base compositions of four strains were determined by CsCl density gradient ultracentrifugation and found to be between 65.4 and 67.4 moles per cent guanine plus cytosine. The growth of all isolates tested is inhibited by fairly low concentrations of cycloserine, streptomycin, penicillin, novobiocin, tetracycline, and chloramphenicol. Nutritional studies on one strain showed that it did not require vitamins or amino acids, although growth was considerably faster in enriched than in synthetic medium. Several sugars and organic acids served as carbon sources, and either NH(4) (+) or glutamate could serve as nitrogen source. The organism is an obligate aerobe and has a pH optimum of 7.5 to 7.8. The optimum temperature for growth is 70 C, the maximum 79 C, and the minimum about 40 C. The generation time at the optimum is about 50 min. The possible relationships of this new genus to the myxobacteria, flexibacteria, and flavobacteria are discussed.  相似文献
