Questions : Woody encroachment in savannas has been associated with changing taxonomic composition and ecosystem function. Interestingly, there is little understanding of how encroachment impacts plant functional diversity and how those changes relate to plant demography, a crucial mediator between taxonomic composition and ecosystem function. Location : Southeastern Brazil. Methods: Using a landscape scale fire suppression experiment in a diverse Brazilian savanna, we quantify how change in species composition over seven years impacted vegetative and reproductive tree functional diversity as determined by new recruits, dead and surviving trees. Results: Over seven years, tree above-ground biomass increased by 15%, while total species richness did not change. Despite minor changes, species composition remained overall similar (82%), with few species contributing significantly to plot dissimilarity over time. There were small changes in vegetative traits, where the community-weighted mean increased in maximum tree height (↑ 2.1%) and specific leaf area (↑ 5.3%), and decreased in wood density (↓ 1.3%) and bark thickness (↓ 9.4%). Changes in reproductive traits were larger than in vegetative traits, with an increase in the prevalence of monoecy (↑ 32.6%), dioecy (↑ 44.2%), large seeds (↑ 20.3%), animal-mediated seed dispersal (↑ 4.9%) and pollination by very small insects (↑ 45.5%), and a decrease in the prevalence of hermaphroditism (↓ 9%), small seeds (6.8%) and pollination by small insects (12.5%). The overall decrease in bark thickness and increase in monoecy and dioecy were mainly driven by characters of the new recruits, while the overall increase in specific leaf area (SLA) and decrease in small seeds appeared largely determined by the loss of trees possessing those traits. Conclusions: Encroachment leads to changes that are likely increasing ecosystem vulnerability to fire and drought. Further, the compositional changes observed appear to drive marked change in reproductive traits, indicating increasing dependence on animals for dispersal and reproduction. Understanding post-hoc encroachment impacts in an era of widespread pervasive encroachment is fundamental to reconciling ecosystem functions such as nutrient cycling and pollination services as there is a loss of species with open ecosystem life-history strategies. Among savannas, there remains an urgent need to understand relationships between woody cover and ecosystem function to determine thresholds in woody cover promoting resilient savanna ecosystems. 相似文献

Leishmania (Viannia) braziliensis control and tissue damage relate to the effector immune response, which in turn affects clinical outcome. Leishmania reactive CD4+ and CD8+ T cells are expanded in long-term healed cutaneous leishmaniasis (hCL) patients but their functional characteristics remain to be determined. This study investigates antigen-specific recall in long-term healed CL caused by L. braziliensis infection. Healed CL subjects were grouped according to the time elapsed since the end of therapy: less than two years and two to five years. Activation phenotype (CD69+ or CD25+) and subpopulations of memory T cell phenotypes [central memory (Tcm): CD45RO+ CCR7+ or effector memory (Tem): CD45RO+ CCR7-] were quantified in ex vivo blood mononuclear cells and after Leishmania antigens stimuli. A reduction in the percentage of activated Leishmania-responder CD4+ and CD8+ T cells in hCL was associated with the time elapsed since clinical cure. Percentage of CD69+ in TCD4+ and TCD8+ cells were negatively correlated with IL-10 levels. Ex vivo analyses showed contracted Tem CD4+ and Tem CD8+ compartments from hCL with long time elapsed since clinical cure, although renewal of these compartments was observed following in vitro exposure to leishmanial stimuli. Our results show that healed L. braziliensis infected patients exhibit a recall response to Leishmania antigens with evident expansion of effector memory T cells. Regulated leishmanial-specific response seems to emerge only about two years after initial contact with the parasite antigens. 相似文献

Canatoxin, a urease isoform from Canavalia ensiformis seeds, shows insecticidal activity against different insect species. Its toxicity relies on an internal 10 kDa peptide (pepcanatox), released by hydrolysis of Canatoxin by cathepsins in the digestive system of susceptible insects. In the present work, based on the N-terminal sequence of pepcanatox, we have designed primers to amplify by PCR a 270-bp fragment corresponding to pepcanatox using JBURE-II cDNA (one of the urease isoforms cloned from C. ensiformis, with high identity to JBURE-I, the classical urease) as a template. This amplicon named jaburetox-2 was cloned into pET 101 vector to obtain heterologous expression in Escherichia coli of the recombinant protein in C-terminal fusion with V-5 epitope and 6-His tag. Jaburetox-2Ec was purified on Nickel-NTA resin and bioassayed in insect models. Dysdercus peruvianus larvae were fed on cotton seed meal diets containing 0.01% (w/w) Jaburetox-2Ec and, after 11 days, all individuals were dead. Jaburetox-2Ec was also tested against Spodoptera frugiperda larvae and caused 100% mortality. In contrast, high doses of Jaburetox-2Ec were innocuous when injected or ingested by mice and neonate rats. Modeling of Jaburetox-2Ec, in comparison with other peptide structures, revealed a prominent beta-hairpin motif consistent with an insecticidal activity based on either neurotoxicity or cell permeation. 相似文献

The antileishmanial efficacy of the reference drug N-methylglucamine antimoniate (Glucantime) was evaluated in groups of rhesus monkeys with acute and chronic Leishmania (Viannia) braziliensis cutaneous infection. The therapeutic responses in experimental animals to either a low dose (5 mg/kg body wt/day for 28 days) or a routine dose (20 mg/kg/day for 28 days) of pentavalent antimony were similar to those reported in the human disease. Primates were cured of their lesions after treatment, but with cryptic parasitism and/or relapse. The rhesus model of L. (V.) braziliensis cutaneous leishmaniasis therefore provides an additional resource for preclinical trials with newer drugs. 相似文献

The control region (CR) is the largest noncoding segment of the mitochondrial DNA and includes the major regulatory elements for its replication and expression. In addition, the high level of intraspecific genetic variability found in the CR favors its use in phylogeographical and population genetic studies of a variety of organisms. However, most of the work on the structure of the CR has focused on vertebrates and insects, and little is known about the evolution of the CR in other taxa. In this study, we sequenced the entire CR of several individuals of 2 crab species: Ucides cordatus (Ocypodidae) and Cardisoma guanhumi (Gecarcinidae). There were neither large conserved regions in the CR of either species nor any similarity among species at the nucleotide level. However, the spatial pattern of genetic variability on the CR was similar among species. In addition, interesting similarities were found in the formation of stable secondary structures and in the position of regulatory elements. These results indicate that the evolution of CR in crustaceans is a remarkably dynamic process, with most homology among species being found at the secondary level. 相似文献
