Host plant specificity was examined in symbiosis between Rhizobium strains isolated from legume-tree root nodules and herbaceous or woody legumes from which they were isolated. Strain GRH2 isolated from Acacia cyanophylla formed effective nodules on Acacia, Prosopis and Medicago sativa as well. Nitrogenase activity, measured as acetylene reduction, of strain GRH2 in symbiosis with Prosopis chilensis was the highest (P 0.05) among the tropical legumes studied and was similar to those found for other associations involving herbaceous legumes. Relative efficiency of nitrogenase varied from 0.3 to near 1 during the light time of the photoperiod. However no hydrogen uptake activity was detected by the amperometric method used. Rhizobium strains GRH3, GRH5 and GRH9 isolated from A. melanoxylon, P. chilensis and Sophora microphylla, respectively, also showed a very low host-range specificity. All isolates were infective and effective on at least one of the herbaceous legumes tested. These data demonstrate the lack of specificity of Rhizobium strains isolated from nitrogen-fixing tree root nodules and that these strains can form effective nodules on herbaceous legumes. 相似文献

In 7-day chick embryo dorsal root ganglia and epidermis cocultures, nerve fibers avoid the epidermis. Previous studies have indicated that glycoproteic factors, secreted by epidermis, could be involved in this phenomenon. Treatment of epidermis by beta-D-xyloside, a specific proteoglycan synthesis inhibitor, abolishes the avoidance reaction. The same result is obtained when anti-chondroitin sulfate antibodies are added to the culture medium. Using HPLC and 35SO4 labeling combined with chondroitinase and hyaluronidase treatment, it has been demonstrated that chondroitin sulfate is present in the epidermal conditioned medium. This suggests that a chondroitin sulfate proteoglycan secreted by the epidermis is implicated in the neurite avoidance reaction and that epidermis could therefore control its own "noninnervation". In vivo, inhibitory influences by local extracellular components may control the guidance of growth cones during nerve pattern formation. 相似文献

Charcoal rot (Macrophomina phaseolina) is a major disease of beans (Phaseolus vulgaris L.) in Mexico. The use of germplasm combining high‐yield stability with resistance to drought and charcoal rot could reduce damage from this disease. In this study, we compared the Eberhart and Russell method and the Additive Main Effect and Multiplicative Interaction (AMMI) model plus biplot analysis for measuring grain yield (GY) and charcoal rot resistance (CHRR) stabilities in 98 F8 : 10 recombinant inbred lines (RILs) derived from a cross between bean adapted to the tropics (BAT) 477 (resistant) × Pinto UI‐114 (susceptible). Experiments were conducted from 2007 to 2009 in Isla, Cotaxtla, Río Bravo and Díaz Ordaz, México, under irrigated or terminal drought conditions. anova detected significant differences (P ≤ 0.05) in GY and CHRR among environments, genotypes and genotype × environment interactions (GEI). Most RILs showed good responses to unfavourable environments based on GY (48) and CHRR (40). AMMI anova s for both traits showed that all sources of variation in the model accounted for approximately 49% of the total squared sum. For the first principal component (PC1), we found 13 RILs that were stable for GY, and for the second (PC2), we found 9 that were stable for GI. For CHRR, we detected 14 stable RILs (PC1) and eight (PC2). Biplot analysis showed the largest vectors for Díaz Ordaz (irrigated and drought, 2008), where the highest and most variable GYs were detected. The shortest vectors were found in Isla (drought, 2007) and Río Bravo (irrigated and drought, 2008), where the lowest and least variable GY were found. We found differential responses of RILs to locations, years and soil humidity conditions as well as significant GEI based on GY and CHRR. Reprimo (RPRM), a downstream effector of p53-induced cell cycle arrest at G2/M, has been proposed as a putative tumor suppressor gene (TSG) and as a potential biomarker for non-invasive detection of gastric cancer (GC). The aim of this study was to evaluate the epigenetic silencing of RPRM gene by promoter methylation and its tumor suppressor function in GC cell lines. Furthermore, clinical significance of RPRM protein product and its association with p53/p73 tumor suppressor protein family was explored. Epigenetic silencing of RPRM gene by promoter methylation was evaluated in four GC cell lines. Protein expression of RPRM was evaluated in 20 tumor and non-tumor matched cases. The clinical significance of RPRM association with p53/p73 tumor suppressor protein family was assessed in 114 GC cases. Tumor suppressor function was examined through functional assays. RPRM gene expression was negatively correlated with promoter methylation (Spearman rank r = -1; p = 0.042). RPRM overexpression inhibited colony formation and anchorage-independent growth. In clinical samples, RPRM gene protein expression was detected in 75% (15/20) of non-tumor adjacent mucosa, but only in 25% (5/20) of gastric tumor tissues (p = 0.001). Clinicopathological correlations of loss of RPRM expression were significantly associated with invasive stage of GC (stage I to II-IV, p = 0.02) and a positive association between RPRM and p73 gene protein product expression was found (p<0.0001 and kappa value = 0.363). In conclusion, epigenetic silencing of RPRM gene by promoter methylation is associated with loss of RPRM expression. Functional assays suggest that RPRM behaves as a TSG. Loss of expression of RPRM gene protein product is associated with the invasive stage of GC. Positive association between RPRM and p73 expression suggest that other members of the p53 gene family may participate in the regulation of RPRM expression. 相似文献

The Orce skull fragment from southern Spain, dated at 1.6 Myr, has been a subject of heated controversy since it was first discovered in 1982. If it is hominid, as its discoverers contend, it is by far the oldest fossil hominid yet found in western Europe and implies that human populations settled this region much earlier than was previously realized. Numerous stone artifacts found at the Orce sites provide evidence that hominids were indeed present there in the Lower Pleistocene. Some paleontologists maintain that the 8 cm diameter occipital fragment is from a horse, not a hominid. Two independent investigations of the residual proteins in the skull were undertaken, one at the University of Granada in Spain, the other at the University of California, San Francisco. Two immunological methods of comparable sensitivity were employed for detection and species attribution of protein extracted from fossil bone: the Granada team used an enzyme-linked-immunosorbent assay (ELISA), and the UCSF team used a radioimmunoassay (RIA). Both teams obtained reactions characteristic of human albumin in the Orce skull and horse albumin in some of the horse fossils. Long term non-progressor patients (LTNPs) are characterized by the natural control of HIV-1 infection. This control is related to host genetic, immunological and virological factors. In this work, phylogenetic analysis of the proviral nucleotide sequences in env gene from a Spanish HIV-1 LTNPs cohort identified a cluster of 6 HIV-1 controllers infected with closely-related viruses. The patients of the cluster showed common clinical and epidemiological features: drug user practices, infection in the same city (Madrid, Spain) and at the same time (late 70’s-early 80’s). All cluster patients displayed distinct host alleles associated with HIV control. Analysis of the virus envelope nucleotide sequences showed ancestral characteristic, lack of evolution and presence of rare amino-acids. Biological characterization of recombinant viruses with the envelope proteins from the cluster viruses showed very low replicative capacity in TZMbl and U87-CD4/CCR5 cells. The lack of clinical progression in the viral cluster patients with distinct combinations of protective host genotypes, but infected by low replicating viruses, indicate the important role of the virus in the non-progressor phenotype in these patients. 相似文献

The PreS1 protein is present on the outermost part of the hepatitis B virus (HBV) surface and has been shown to have a pivotal function in viral infectivity and assembly. The development of reagents with high affinity and specificity for PreS1 is of great significance for early diagnosis and treatment of HBV infection. A phage display library of dodecapeptide was screened for interactions with purified PreS1 protein. Alignment of the positive phage clones revealed a putative consensus PreS1 binding motif of HXnHXmHP/R. Moreover, a peptide named P7 (KHMHWHPPALNT) was highly enriched and occurred with a surprisingly high frequency of 72%. A thermodynamic study revealed that P7 has a higher binding affinity to PreS1 than the other peptides. Furthermore, P7 was able to abrogate the binding of HBV virions to the PreS1 antibody, suggesting that P7 covers key functional sites on the native PreS1 protein. This newly isolated peptide may, therefore, be a new therapeutic candidate for the treatment of HBV. The consensus motif could be modified to deliver imaging, diagnostic, and therapeutic agents to tissues affected by HBV. 相似文献
