Aerobiology of caves is still in its infancy. At present, no clear information has been generated on the limits of acceptance of fungal spores in air which permit classification of the atmosphere of a cave as not dangerous for the conservation of rock-art paintings. We had the unique opportunity to visit and sample different caves in Spain and France, under different managements. We obtained a collection of data related with contamination episodes that permitted the formulation of a tentative index of fungal hazard in show caves. This is supported by the concentration of fungal spores in the cave air, the knowledge of the cave history and management, and a detailed survey of the different halls of the caves. The index classifies the risks into five categories: category 1 identifies a cave without fungal problems, category 2 is an alarm signal for caves, category 3 is a cave threatened by fungi, category 4 is assigned to a cave already affected by fungi, and category 5 is a cave with an irreversible ecological disturbance. This index, a working hypothesis, is launched to promote interest and forum discussion and should be validated by the scientific community after being updated with more surveys and cave analyses carried out under different managements and with different contamination episodes.  相似文献

A 120ps molecular dynamics (MD) trajectory was calculated and analyzed for a putative Pro-36-Gly mutant of the potato carboxypeptidase A (CPA) protein inhibitor (PCIm). The mutant protein's fold shows a large degree of stability, judged from its low alpha-carbon r.m.s. deviation from the X-ray structure of the wild type PCI (PCIw). The N-terminal tail of PCIm differs slightly less from the X-ray structure than it does in PCIw, while the mutant's C-terminal tail (the primary contact site with CPA) and residues 13-17 present deviations as they approach each other. Differences in fluctuation pattern exist between PCIm and PCIw in residues 2-4 (the N-terminal tail), 13-17, 22-23, 28-31 (the secondary contact site with CPA) and 37-38 (the C-terminal tail); the latter region is rigidified in PCIm. Results show that the MD method is able to sense local perturbative effects produced by amino acid substitutions in flexible regions of protein molecules. The simulation suggests that the conformation of the C-terminal tail is less favorable for interaction with the target protein in the mutant than it is in the wild type protein. The Pro-36-Gly mutant is predicted to be a less potent inhibitor.  相似文献

The influence of the bonding form distribution of Fe, Ni, Co and Mn and their potential bioavailability during the anaerobic degradation of maize straw was investigated. Two reactors were operated over 117 days at 37°C and different dosage strategies of mineral were studied in reactor (R2). Control reactor (R1) was metal‐limited over time. mineral supplementation (1 g L?1) once a week reported the highest methane yield (257 mL g?1 VS) with 30% of increment. Ni and Co predominated in their oxidizable bonding forms and Fe mainly existed as residual and oxidizable fractions. The potential bioavailability (Mn ?? Co ≈ Ni ? Fe) of R2 was higher comparing to R1. Metal deprivation in R1 led to depletion of both sequential extraction fractions and total metal concentrations until the end of the process. This study confirmed that the dosage strategy of mineral has a stimulatory effect on methane production from crop maize waste.  相似文献

From the anthracological study of considerable quantities of charcoal recovered from the excavation of the settlement at Los Castillejos de Montefrío, a synthesis was made of the vegetation dynamics during Recent Prehistory of the area, from the middle of the sixth millennium bc to the beginning of the second millennium bc. The vegetation dynamics mark four anthracological phases over this sequence. From the Late Neolithic (end of the fifth millennium bc) the influence of different activities such as livestock raising, burning and agriculture become evident in the development of the natural vegetation, altering species composition and appearance frequency. The last anthracological phase, MF4, spanning the second half of the third millennium bc, saw the regeneration of the tree vegetation as certain former activities ceased, indicating the environmental singularity of this enclave within the context of the southern Iberian Peninsula.  相似文献

Data reported in the literature indicate that lipid movement between intracellular organelles can occur through contacts and close physical association of membranes (Vance, J.E. 1990. J Biol Chem 265: 7248-7256). The advantage of this mechanism is that the direct interaction of membranes provides the translocation event without the involvement of lipid-transport systems. However, pre-requisite for the functioning of this machinery is the presence of protein factors controlling membrane association and fusion. In the present work we have found that liposomes fuse to mitochondria at acidic pH and that the pre-treatment of mitochondria with pronase inhibits the fusogenic activity. Mixing of 14C-phosphatilyserine (PS) labeled liposomes with mitochondria at pH 6.0 results in the translocation of 14C-PS into mitochondria and in its decarboxylation to14 C-phosphatidylethanolamine through the PS decarboxylase activity localized on the outer surface of the inner mitochondrial membrane. Incorporation of 14C-PS is inhibited by the pre-treatment of mitochondria with pronase or with EEDQ, a reagent for the derivatization of the protonated form of carboxylic groups. These results indicate the presence of a protein associated with mitochondria which is able to trigger the fusion of liposomes to the mitochondrial membrane. A partial purification of a mitochondrial fusogenic glycoprotein is described in this work. The activity of the fusogenic protein appears to be dependent on the extent of protonation of the residual carboxylic groups and is influenced by the glucidic moiety, as demonstrated by its interaction with Concanavalin A. The purifed protein is able to promote the recover of the14 C-PS import from liposomes to pronase-treated mitochondria. Therefore, the protein is candidate to be an essential component in the machinery for the mitochondrial import of PS. (Mol Cell Biochem 175: 71–80, 1997)  相似文献
