For the purpose of efficiently utilizing the renewable solar energy, it is of vital importance to understand the key factors that contribute to the performance merits for photocatalysis applications. In this work, we find that anatase titania nanostructures with high efficiency in photoelectrochemical cell (PEC) do not necessarily retain the same good performance when used in direct heterogeneous reaction (DHR). Investigation is carried out to elucidate how the electronic properties of the different nanostructures are correlated with the PEC and DHR efficiencies. Good PEC cell performance is identified to be related to topotactically formed samples with intimately connected particles that facilitate easy charge transfer. Additional benefit for PEC cell is found to be achieved from the vectorial conduction pathway in the pseudo one dimensional structure. On the other hand, high activity of DHR photocatalysis is attributed mainly to the exposed high reactivity crystal facets. The presence of anatase TiO2 {010} facets is identified to enhance electron‐hole separation and create specific surface states that facilitate interactions across the semiconductor/electrolyte interfaces.  相似文献

Three species of Plectus Bastian, 1865 viz., P. aquatilis and P. pusillus from Kerman province and P. velox from Alborz province, Iran are described and illustrated. Partial sequences of 18S region of ribosomal DNA gene were amplified for P. aquatilis and P. pusillus. The Blast results of population of P. aquatilis from Iran showed 8–10 nucleotides differences with populations of the same species (AF036602; GQ892827; AY284700) reported from the UK, Belgium and The Netherlands, respectively. Whereas Iranian population of P. pusillus showed 14–16 nucleotides differences with P. cf. pusillus (AY284705; AY284704) reported from The Netherlands. Molecular analysis revealed close relationship of the Iranian plectids with P. cf. parvus (AY284699) reported from The Netherlands. Phylogenetic relationships with other related species in the genus Plectus and closely related genera that are available in the GenBank are given.  相似文献

Urinary miRNAs are discussed as potential biomarkers for bladder cancer. The majority of miRNAs, however, are downregulated, making it difficult to utilize reduced miRNA signals as reliable diagnostic tools. Because the downregulation of miRNAs is frequently associated with hypermethylation of the respective regulative sequences, we studied whether DNA hypermethylation might serve as an improved diagnostic tool compared to measuring downregulated miRNAs. miRNA expression arrays and individual qPCR were used to identify and confirm miRNAs that were downregulated in malignant urothelial cells (RT4, 5637 and J82) when compared to primary, non-malignant urothelial cells (HUEPC). DNA methylation was determined by customized PCR-arrays subsequent to methylation-sensitive DNA-restriction and by mass spectrometry. miRNA expression and DNA methylation were determined in untreated cells and in cultures treated with the demethylating agent 5-Aza-2′-deoxycytidine. miR-200b, miR-152 and miR-10a displayed differential expression and methylation among untreated cancer cell lines. In addition, reduced miRNA expression of miR-200b, miR-152, and miR-10a was associated with increased DNA methylation in malignant cells versus HUEPC. Finally, the demethylation approach revealed a causal relationship between both parameters for miR-152 in 5637 and also suggests a causal connection of both parameters for miR-200b in J82 and miR-10a in 5637. In conclusion, our studies in multiple bladder cancer cell lines and primary non-malignant urothelial cells suggest that hypermethylation of miR-152, miR-10a and miR-200b regulative DNA sequences might serve as epigenetic bladder cancer biomarkers.  相似文献

Abstract

Gobies (family Gobiidae) were in the past and now important components of marine ecosystems as an essential part of the food chain. However, the early fossil record of this group is relatively meager, with only scarce skeletal remains. The oldest known representative of the genus Gobius has been recently described from the Early Miocene of Czech Republic as Gobius jarosi P?ikryl & Reichenbacher, 2018. Here we present a detailed study of a well-preserved goby skeleton of the same age from the Harta locality (Poland). This specimen is assumed as belonging to Gobius jarosi based on its almost complete morphological and meristic identity with the type material from the Vá?any nad Litavou locality. Some aspects of the paleogeography and paleoecology of the Early Miocene fish assemblage from Harta with special reference to the Carpathian Basin are also discussed in this paper.  相似文献

Latent Membrane Protein 2A (LMP2A) is an Epstein–Barr virus-encoded protein that is important for the maintenance of latent infection. Its activity affects cellular differentiation, migration, proliferation and B cell survival. LMP2A resembles a constitutively activated B cell antigen receptor and exploits host kinases to activate a set of downstream signaling pathways. In the current study we demonstrate the interaction of LMP2A with intersectin 1 (ITSN1), a key endocytic adaptor protein. This interaction occurs via both the N- and C-tails of LMP2A and is mediated by the SH3 domains of ITSN1. Additionally, we identified the Shb adaptor and the Syk kinase as novel binding ligands of ITSN1. The Shb adaptor interacts simultaneously with the phosphorylated tyrosines of LMP2A and the SH3 domains of ITSN1 and mediates indirect interaction of ITSN1 to LMP2A. Syk kinase promotes phosphorylation of both ITSN1 and Shb adaptors in LMP2A-expressing cells. In contrast to ITSN1, Shb phosphorylation depends additionally on Lyn kinase activity.Considering that Shb and ITSN1 are implicated in various receptor tyrosine kinase signaling, our results indicate that LMP2A can affect a number of signaling pathways by regulating the phosphorylation of the ITSN1 and Shb adaptors.  相似文献

N-terminal targeting signals (presequences) direct proteins across the TOM complex in the outer mitochondrial membrane and the TIM23 complex in the inner mitochondrial membrane. Presequences provide directionality to the transport process and regulate the transport machineries during translocation. However, surprisingly little is known about how presequence receptors interact with the signals and what role these interactions play during preprotein transport. Here, we identify signal-binding sites of presequence receptors through photo-affinity labeling. Using engineered presequence probes, photo cross-linking sites on mitochondrial proteins were mapped mass spectrometrically, thereby defining a presequence-binding domain of Tim50, a core subunit of the TIM23 complex that is essential for mitochondrial protein import. Our results establish Tim50 as the primary presequence receptor at the inner membrane and show that targeting signals and Tim50 regulate the Tim23 channel in an antagonistic manner.  相似文献

Spatial and stochastic models are often straightforward to simulate but difficult to analyze mathematically. Most of the mathematical methods available for nonlinear stochastic and spatial models are based on heuristic rather than mathematically justified assumptions, so that, e.g., the choice of the moment closure can be considered more of an art than a science. In this paper, we build on recent developments in specific branch of probability theory, Markov evolutions in the space of locally finite configurations, to develop a mathematically rigorous and practical framework that we expect to be widely applicable for theoretical ecology. In particular, we show how spatial moment equations of all orders can be systematically derived from the underlying individual-based assumptions. Further, as a new mathematical development, we go beyond mean-field theory by discussing how spatial moment equations can be perturbatively expanded around the mean-field model. While we have suggested such a perturbation expansion in our previous research, the present paper gives a rigorous mathematical justification. In addition to bringing mathematical rigor, the application of the mathematically well-established framework of Markov evolutions allows one to derive perturbation expansions in a transparent and systematic manner, which we hope will facilitate the application of the methods in theoretical ecology.  相似文献

Molecular structures of native and a pair of modified small interfering RNA-RNA duplexes containing carbocyclic [6?'-(R)-OH/7?'-(S)-methyl]- and [6?'-(S)-OH/7?'-(S)-methyl]-carba-LNA-thymine nucleotides, which are two diastereomeric analogs of the native T nucleotide, incorporated at position 13 in the antisense (AS) strand of siRNA, have been simulated using molecular mechanics/dynamics techniques. The main aim of the project has been to find a plausible structural explanation of why modification of siRNA at T(13) position by the [6?'(R)-O-(p-Toluoyl)-7?'(S)-methyl]-carba-LNA-Thymine [IC(50) of 3.32 ± 0.17 nM] is ca 24 times more active as an RNA silencing agent against the target HIV-1 TAR RNA than the [6?'(S)-O-(p-Toluoyl)-7?'(S)-methyl]-counterpart [IC(50) of 79.8 ± 17 nM] [1]. The simulations reveal that introduction of both C6?'(R)-OH and C6?'(S)-OH stereoisomers does not lead even to local perturbation of the siRNA-RNA duplex structures compared to the native, and the only significant difference between 6?'(S)- and 6?'(R)-diastereomers found is the exposure of the 6?'-OH group of the 6?'(R)-diastereoisomer toward the edge of the duplex while the 6?'-hydroxyl group of the 6?'(S)-diastereoisomer is somewhat buried in the minor groove of the duplex. This rules out a hypothesis about any possible local distortion by the nature of chemical modification of the siRNA-target the RNA duplex, which might have influenced the formation of the effective RNA silencing complex (RISC) and puts some weight on the hypothesis about the 6?'-hydroxy group being directly involved with most probably Ago protein, since it is known from exhaustive X-ray studies [2, 3] that the core residues are indeed involved with hydrogen bonding with the internucleotidyl phosphates. Further systematic investigation is in progress to map the position-dependent functional and nonfunctional interactions of the modified [6?'(R or S)-O-(p-Toluoyl)-7?'(S)-methyl]-carba-LNA-T with the Ago2 protein of the RISC.  相似文献
