Any given single nucleotide polymorphism (SNP) in a genome may have little or no functional impact. A biologically significant effect may possibly emerge only when a number of key SNP-related genotypes occur together in a single organism. Thus, in analysis of many SNPs in association studies of complex diseases, it may be useful to look at combinations of genotypes. Genes related to signal transmission, e.g., ion channel genes, may be of interest in this respect in the context of bipolar disorder. In the present study, we analysed 803 SNPs in 55 genes related to aspects of signal transmission and calculated all combinations of three genotypes from the 3×803 SNP genotypes for 1355 controls and 607 patients with bipolar disorder. Four clusters of patient-specific combinations were identified. Permutation tests indicated that some of these combinations might be related to bipolar disorder. The WTCCC bipolar dataset were use for replication, 469 of the 803 SNP were present in the WTCCC dataset either directly (n = 132) or by imputation (n = 337) covering 51 of our selected genes. We found three clusters of patient-specific 3×SNP combinations in the WTCCC dataset. Different SNPs were involved in the clusters in the two datasets. The present analyses of the combinations of SNP genotypes support a role for both genetic heterogeneity and interactions in the genetic architecture of bipolar disorder. 相似文献

Changes in mortality and turnover of selected woody plants which dominate and characterise the arid vegetation of the island of Santa Fe, Galapagos, have been studied regularly during the period 1972–2003. After the eradication of feral goats from Santa Fe in 1971, the vegetation started to recover. The previously threatened small tree Scalesia helleri ssp. helleri recovered and some recruitment of the dominant trees Opuntia echios var. barringtonensis and Bursera graveolens took place. The dominant shrub species Cordia lutea, Encelia hispida and Lantana peduncularis increased in number, whereby the shrub layer is becoming denser and the structure of the predominant dry season deciduous steppe forest vegetation is changing. The strong impact of El Niño on the survival and recruitment of Opuntia indicates that recovery of Opuntia-dominated vegetation should be viewed on a time-scale of perhaps 150 years or more, in correspondence with the long life-expectancy of adult Opuntia individual. It is suggested that recruitment and survival of Opuntia may be hindered if future El Niño events are to occur with increased strength, and that this would alter the aspect of the Santa Fe vegetation. 相似文献

The chemistry of Type 2 copper depleted T2D Rhus laccase has been investigated with regard to the binding of peroxide, and the ability of the enzyme to undergo reduction and reoxidation. Although the peroxide affinity is diminished in the T2D enzyme (104 M?1) relative to the holo-enzyme ((? 108 M?1) the actual mode of binding as a Type 3 μ-peroxo complex remains, as indicated by absorption and CD spectral measurements. Anaerobic reductive and reoxidative titrations with hydroquinone and hydrogen peroxide respectively revealed that the Type 3 copper pairwise interaction is disrupted during reduction but can be restored on reoxidation. The concept of separate Type 2 and Type 3 copper redox centers is suggested to be inadequate in view of the loss of functional integrity by the Type 3 site on removal of Type 2 copper. 相似文献

BACKGROUND: The growth-suppressive effect of systemic glucocorticoids in children is well established, however, recovery of growth after withdrawal of short-term treatment in school-age children has not been evaluated. OBJECTIVE: To assess short-term growth after withdrawal of systemic glucocorticoids. Methods: A post-hoc analysis of data from a double-blind lower leg growth trial which compared 5 mg prednisolone once daily in the evening with placebo was performed. The study consisted of run-in, treatment, wash-out and run-out periods of 1 week duration. In 10 children with asthma (mean age 11 years) lower leg growth measured with the knemometer could be studied up to 3 weeks after withdrawal of prednisolone. RESULTS: Mean (SEM) lower leg growth rates during run-in, prednisolone treatment and the first, second and third weeks after withdrawal of prednisolone were 0.48 (0.15), -0.27 (0.20), 0.53 (0.19), 0.72 (0.16) and 0.66 (0.14) mm/week, p < 0.001. Mean growth rates during run-in and the first, second and third weeks after withdrawal of prednisolone did not vary, p = 0.68. CONCLUSION: Recovery of suppressed lower leg growth rates occurs within a week after withdrawal of exogenous glucocorticoids. 相似文献

BACKGROUND: and Aims Submerged plants possess well-developed aerenchyma facilitating intra-plant gas-phase diffusion of O2 to below-ground tissues, which are usually buried in anoxic sediments. However, aquatic habitats differ in terms of O2 fluctuations in the water column and in O2 consumption of the sediment, and aquatic plants differ in aerenchymal volume and resistance to O2 diffusion through the plant and across leaf and root surfaces. The hypothesis that the freshwater isoetid Lobelia dortmanna and the marine seagrass Zostera marina should display pronounced contrasts in intra-plant O2 dynamics because of differences in morphology/anatomy, physiology and growth habitat was tested. METHODS: In order to determine the O2 dynamics and relate this to the anatomy and morphology of the two species, O2 microelectrodes were inserted in the aerenchyma of leaves and roots, the sediment pore-water, and the water column in the field. Manipulation of water column O2 in the laboratory was also carried out. KEY RESULTS: It was found that intra-plant transport of O2 between leaf and root tips takes place more readily in L. dortmanna than in Z. marina due to shorter distances and greater cross-sections of the aerenchyma. The major exchange of O2 across roots of L. dortmanna can be accounted for by small intra-plant resistances to diffusion, larger root than leaf surfaces, and greater radial diffusive resistance of leaves than roots. In contrast, the major O2 exchange across leaves than roots of Z. marina can be accounted for by the opposite anatomical-morphological features. The larger aerenchymal volume and the smaller metabolic rates of L. dortmanna compared to Z. marina imply that turnover of O2 is slower in the aerenchyma of L. dortmanna and O2 fluctuations are more dampened following changes in irradiance. Also, O2 accumulated in the aerenchyma can theoretically support dark respiration for a few hours in L. dortmanna but for only a few minutes in Z. marina. CONCLUSIONS: The build-up of O2 in the pore-water of L. dortmanna sediments during the day as a result of high release of photosynthetic O2 from roots and low O2 consumption of sediments means that sediment, aerenchyma and water are important O2 sources for respiration during the following night, while Z. marina relies on the water column as the sole source of O2 because its sediments are anoxic. These differences between L. dortmanna and Z. marina appear to represent a general difference between the isoetid species mainly inhabiting sediments of low reducing capacity of oligotrophic lakes and the elodeid freshwater species and marine seagrasses mainly inhabiting sediments of higher reducing capacity in more nutrient-rich habitats. 相似文献

Patients with congestive heart failure (CHF) are prone to increased skeletal muscle fatigue. Elevated circulatory concentrations of tumor necrosis factor (TNF)-alpha and monocyte chemoattractant protein-1, which may stimulate matrix metalloproteinase (MMP) activity and, thereby, contribute to skeletal muscle dysfunction, are frequently found in CHF. However, whether skeletal muscle MMP activity is altered in CHF is unknown. Hence, we have used a gelatinase assay to assess the activity of MMP and tissue inhibitors of MMP in single skeletal muscles of rats with CHF 6 wk after induction of myocardial infarction. Sham-operated (Sham) rats were used as controls. We also measured the gene expression and protein contents of MMP-2 and MMP-9 in skeletal muscles of these rats. Plasma MMP activity was nearly seven times higher (P < 0.05) in CHF than in Sham rats. Concomitantly, the MMP activity within single slow- and fast-twitch skeletal muscles of CHF rats increased two- to fourfold compared with Sham animals, whereas tissue inhibitor of MMP activity did not differ (P > 0.05). Preformed MMP-2 and MMP-9 were probably activated in CHF, because neither their gene expression nor protein levels were altered (P > 0.05). Serum concentrations of TNF-alpha and monocyte chemoattractant protein-1 remained unchanged (P > 0.05) between CHF and Sham rats during the 6-wk observation period. We conclude that development of CHF in rats enhances MMP activity, which in turn may distort the normal contractile function of skeletal muscle, thereby contributing to increased skeletal muscle fatigue. 相似文献

FK-506, a widely used immunosuppressant, has caused a few clinical cases with QT prolongation and torsades de pointe at high blood concentration. The proarrhytmogenic potential of FK-506 was investigated in single rat ventricular cells using the whole cell clamp method to record action potentials (APs) and ionic currents. Fluorescence measurements of Ca2+ transients were performed with indo-1 AM using a multiphotonic microscope. FK-506 (25 micromol/l) hyperpolarized the resting membrane potential (RMP; -3 mV) and prolonged APs (AP duration at 90% repolarization increased by 21%) at 0.1 Hz. Prolongation was enhanced by threefold at 3.3 Hz, and early afterdepolarizations (EADs) occurred in 59% of cells. EADs were prevented by stronger intracellular Ca2+ buffering (EGTA: 10 vs. 0.5 mmol/l in the patch pipette) or replacement of extracellular Na+ by Li+, which abolishes Na+/Ca2+ exchange [Na+/Ca2+ exchanger current (INaCa)]. In indo-1-loaded cells, FK-506 generated doublets of Ca(2+) transients associated with increased diastolic Ca2+ in one-half of the cells. FK-506 reversibly decreased the L-type Ca2+ current (ICaL) by 25%, although high-frequency-dependent facilitation of ICaL persisted, and decreased three distinct K+ currents: delayed rectifier K+ current (IK; >80%), transient outward K+ current (<20%), and inward rectifier K+ current (IK1; >40%). A shift in the reversal potential of IK1 (-5 mV) accounted for RMP hyperpolarization. Numerical simulations, reproducing all experimental effects of FK-506, and the use of nifedipine showed that frequency-dependent facilitation of ICaL plays a role in the occurrence of EADs. In conclusion, the effects of FK-506 on the cardiac AP are more complex than previously reported and include inhibitions of IK1 and ICaL. Alterations in Ca2+ release and INaCa may contribute to FK-506-induced AP prolongation and EADs in addition to the permissive role of ICaL facilitation at high rates of stimulation. 相似文献

To investigate early secretory defects in prediabetes, we evaluated beta-Cell function and insulin sensitivity (M value, by euglycemic clamp) in 26 normotolerant first-degree relatives of type 2 diabetic patients (FDR) and 17 age- and weight-matched control subjects. beta-Cell function was assessed by modeling analysis of glucose and C-peptide concentrations measured during 24 h of standardized living conditions. Fasting and total insulin secretion (ISR) were increased in FDR, as was ISR at a reference 5 mM glucose level (ISR5, 107 +/- 6 vs. 87 +/- 6 pmol x min(-1) x m(-2), P < 0.05). ISR5 was inversely related to M in controls (ISR5 = k/M1.23, rho = -0.74, P < 0.005) but not in FDR; when M was accounted for (by calculating a compensation index ISR5 x M1.23), compensation for insulin resistance was impaired in FDR (10.8 +/- 1.0 vs. 13.4 +/- 0.6 units, P < 0.05). Potentiation of ISR, expressing relative transient increases in glucose-stimulated ISR during meals, was impaired in FDR (1.29 +/- 0.08 vs. 1.62 +/- 0.08 during 1st meal, P < 0.02). Moreover, the potentiation time course was related to glucose-dependent insulin-releasing polypeptide (GIP) concentrations in both groups, and the sensitivity of potentiation to GIP derived from this relationship tended to be impaired in FDR. Compensation index, potentiation, and sensitivity to GIP were interrelated parameters (P < 0.05 or less). beta-Cell function parameters were also related to mean 24-h glucose levels (r2 = 0.63, P < 0.0001, multivariate model). In conclusion, although in absolute terms ISR is increased in insulin-resistant FDR, beta-cell function shows a cluster of interrelated abnormalities involving compensation for insulin resistance, potentiation, and sensitivity to GIP, suggesting a beta-cell defect in the amplifying pathway of insulin secretion. 相似文献

Chlorosomes, the light-harvesting apparatus of green bacteria, are a unique antenna system, in which pigments are organized in aggregates rather than associated with proteins. Isolated chlorosomes from the green sulphur bacterium Chlorobium tepidum contain 10 surface-exposed proteins. Treatment of chlorosomes from Chlorobium tepidum with protease caused changes in the spectral properties of bacteriochlorophyll c and digestion of chlorosome proteins. Using SDS-PAGE analysis, immunoblotting and matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF-MS) we have investigated the topology of the 59-residue CsmA protein. Our results show that at the N-terminus, the only amino acid available for protease degradation is the methionine. At the C-terminus, amino acids can be removed by protease treatment to produce a residual protein containing at least the sequence between residues 2 and 38. These results indicate that the N-terminal portion of the CsmA protein, which is predicted to be mainly hydrophobic, is buried in the chlorosome envelope. 相似文献
