The geographic occurrence of moggel Labeo umbratus × mud fish Labeo capensis hybrids and the threat of hybridisation to the genetic integrity of L. umbratus were investigated. Genetic evidence from mtDNA cytb and nDNA s7 intron sequence data indicated probable interspecific hybridisation in two impoundments, Hardap Dam (Orange River Basin, Namibia) and Darlington Dam (Sundays River Basin, Eastern Cape, South Africa). Some putative hybrids were morphologically identifiable on account of their meristic and morphometric intermediacy to the parent species. Human activities appear to drive the observed hybridisation, either through dam construction and direct stocking of L. umbratus or via translocation of Labeo capensis and the previously isolated Orange River lineage of L. umbratus into the southern-flowing systems by inter-basin water transfers. It is recommended that further translocation of fish from these areas should be avoided.  相似文献

Purpose

Due to their production intensity, different foods of animal or plant origin play a crucial role in the assessment of the environmental impacts of human nutrition and diets. Based on a representative nutrition survey in Germany from the year 2006, a life cycle assessment (LCA) was conducted to quantify nutrition-related emissions of animal and plant-based foods (excluding beverages), with a special focus on the socio-demographic factor gender.

Materials and methods

For the study, representative data sets concerning German food production and consumption were used. These were complemented by the Danish LCA Food database and other LCA data to analyse the impact of food imports. As regards environmental impact assessment, global warming potential (GWP) was assessed, which included emissions from direct land use change and land use (dLUC, LU), along with three inventory indicators (ammonia emissions, land use, blue water use). The following food groups were analysed from cradle-to-store and their impacts were evaluated and compared with each other: animal-based foods (meat products, milk products, egg products and fish products), plant-based foods (grain products, vegetables, fruits, potato products, margarine/oils, sugar/sweets). The reference year in the study is the year 2006.

Results and discussion

For all indicators, the results show strong variation between the genders. Even if the physiologically different consumption patterns among men and women are adjusted on a weight basis, men show a higher impact in terms of GWP (CO2 eq. +25%), ammonia emissions (+30%) and land use (+24%). In contrast, women demonstrate a higher water demand (+11%). These differences are primarily caused by a higher share of meat and meat products in the usual diet of men (+28%) as well as of fruit and vegetables in the diet of women (+40%). If men were to shift qualitatively to the usual diet of women, then 14.8 Mt CO2 eq. and 60.1 kt ammonia emissions could be saved annually. Within the system boundaries of our study, this would translate into a reduction of 12% of CO2 eq. and 14% of ammonia emissions. With regard to land use, this equals an area of 15,613?km2?year?1 (?11%), whereas the total blue water demand would be increased by 94?Mm3?year?1 (+7%). Limitations within this study are caused by the system boundaries cradle-to-store and are also due to the restricted set of environmental indicators which were analysed. Nonetheless, our results for GWP and land use are in keeping with previous studies. The results concerning ammonia and blue water use are limited when compared with other study results.

Conclusions

The study shows that within one society distinct diet profiles with markedly different environmental impacts are already established. Taking cultural and physiological considerations among the genders into account, these differences could be seen as offering potential opportunities to strengthen sustainable diet profiles. Further research should also consider health impact assessments to ensure that alterations in diet profiles due to environmental constraints do not lead to disadvantageous public health effects. Particular attention should be paid here to potentially undernourished subgroups (such as the elderly, sick people, pregnant women).  相似文献

The separation of sister chromatids in anaphase depends on the dissociation of cohesin from chromosomes. In vertebrates, some cohesin is removed from chromosomes at the onset of anaphase by proteolytic cleavage. In contrast, the bulk of cohesin is removed from chromosomes already in prophase and prometaphase by an unknown mechanism that does not involve cohesin cleavage. We show that Polo-like kinase is required for the cleavage-independent dissociation of cohesin from chromosomes in Xenopus. Cohesin phosphorylation depends on Polo-like kinase and reduces the ability of cohesin to bind to chromatin. These results suggest that Polo-like kinase regulates the dissociation of cohesin from chromosomes early in mitosis.  相似文献

Aims

Aim was to elucidate the specific role of pattern recognition receptors in vascular dysfunction during polymicrobial sepsis (colon ascendens stent peritonitis, CASP).

Methods and Results

Vascular contractility of C57BL/6 (wildtype) mice and mice deficient for Toll-like receptor 2/4/9 (TLR2-D, TLR4-D, TLR9-D) or CD14 (CD14-D) was measured 18 h following CASP. mRNA expression of pro- (Tumor Necrosis Factor-α (TNFα), Interleukin (IL)-1β, IL-6) and anti-inflammatory cytokines (IL-10) and of vascular inducible NO-Synthase (iNOS) was determined using RT-qPCR. Wildtype mice exhibited a significant loss of vascular contractility after CASP. This was aggravated in TLR2-D mice, blunted in TLR4-D animals and abolished in TLR9-D and CD14-D animals. TNF-α expression was significantly up-regulated after CASP in wildtype and TLR2-D animals, but not in mice deficient for TLR4, -9 or CD14. iNOS was significantly up-regulated in TLR2-D animals only. TLR2-D animals showed significantly higher levels of TLR4, -9 and CD14. Application of H154-ODN, a TLR9 antagonist, attenuated CASP-induced cytokine release and vascular dysfunction in wildtype mice.

Conclusions

Within our model, CD14 and TLR9 play a decisive role for the development of vascular dysfunction and thus can be effectively antagonized using H154-ODN. TLR2-D animals are more prone to polymicrobial sepsis, presumably due to up-regulation of TLR4, 9 and CD14.  相似文献

Background

Epidemiological case-control studies have revealed associations between mitochondrial haplogroups and the onset and/or progression of various multifactorial diseases. For instance, mitochondrial haplogroup T was previously shown to be associated with vascular diseases, including coronary artery disease and diabetic retinopathy. In contrast, haplogroup H, the most frequent haplogroup in Europe, is often found to be more prevalent in healthy control subjects than in patient study groups. However, justifications for the assumption that haplogroups are functionally distinct are rare. Therefore, we attempted to compare differences in mitochondrial function between haplogroup H and T cybrids.

Methodology/Principal Findings

Mitochondrial haplogroup H and T cybrids were generated by fusion of HEK293 cells devoid of mitochondrial DNA with isolated thrombocytes of individuals with the respective haplogroups. These cybrid cells were analyzed for oxidative phosphorylation (OXPHOS) enzyme activities, mitochondrial DNA (mtDNA) copy number, growth rate and susceptibility to reactive oxygen species (ROS). We observed that haplogroup T cybrids have higher survival rate when challenged with hydrogen peroxide, indicating a higher capability to cope with oxidative stress.

Conclusions/Significance

The results of this study show that functional differences exist between HEK293 cybrid cells which differ in mitochondrial genomic background.  相似文献

Dengue is the most important arboviral infection of humans. Thrombocytopenia is frequently observed in the course of infection and haemorrhage may occur in severe disease. The degree of thrombocytopenia correlates with the severity of infection, and may contribute to the risk of haemorrhage. As a result of this prophylactic platelet transfusions are sometimes advocated for the prevention of haemorrhage. There is currently no evidence to support this practice, and platelet transfusions are costly and sometimes harmful. We conducted a global survey to assess the different approaches to the use of platelets in dengue. Respondents were all physicians involved with the treatment of patients with dengue. Respondents were asked that their answers reflected what they would do if they were the treating physician. We received responses from 306 physicians from 20 different countries. The heterogeneity of the responses highlights the variation in clinical practice and lack of an evidence base in this area and underscores the importance of prospective clinical trials to address this key question in the clinical management of patients with dengue.  相似文献
