Chronic rotavirus infection of an infant with severe combined immunodeficiency (SCID) was studied by virological examinations in association with long-term observation of his symptoms and immune status. During eleven months of hospitalization, the patient was suffering from incurable severe diarrhea with persisting excretion of rotaviruses detected by electron microscopy and the reversed-passive hemagglutination (R-PHA) test and had transient hepatitis symptom despite multiple administrations of human gammaglobulin and high calorie fluids. The detected viruses were morphologically recognized as rotavirus with double capsid structure. Polyacrylamide gel electrophoretic (PAGE) analysis of their genomic RNAs showed the long electropherotype of group A virus with abnormal migration profiles changing considerably from the early to the late phase of illness: (1) The 11th segment became undetectable; (2) the molecular weight of the 6th segment slightly increased; (3) seven to fourteen extra segments appeared; and (4) PAGE patterns of viral genomic RNAs changed every three or four months. These findings suggest that chronic infection with rotavirus accompanied the generation of extra viral genomic segments and their unusual assortments in an immunodeficient host.  相似文献

Isolated single smooth muscle cells from the fundus of a guinea-pig stomach were permeabilized by use of streptolysin-O (0.5 U/ml). Most of the permeabilized cells responded to 0.6 microM Ca2+, but not to 0.2 microM Ca2+, with a resulting maximal cell shortening to approximately 71% of the resting cell length. These cells were relaxed again by washing with the Ca2+-free solution (2.5 nM free Ca2+) for 3-5 min. Addition of 10 microM acetylcholine (ACh) resulted in both a marked decrease in the concentration of Ca2+ required to trigger a threshold response and an increase in the maximal cell shortening, indicating that the cells retained the muscarinic receptor function. When the cell treated with a protein kinase C (PKC) inhibitor, K-252b (1 microM), for 3 min was exposed to 10 microM ACh in the presence of K-252b, the cell shortened within 2 min with a maximal cell shortening. When the cell shortening was induced by 10 microM ACh plus 1 microM Ca2+ in the presence of K-252b (1 microM) or more selective PKC inhibitors, such as calphostin C (1 microM) or PKC pseudosubstrate peptide (100 microM), the extension of the shortened cells, by washing with the Ca2+-free solution, was significantly inhibited. In contrast, K-252b (1 microM) did not inhibit the relaxation of Ca2+-induced shortened cells. These results suggest that the receptor-mediated activation of PKC in the process of ACh-induced cell shortening plays a role in the subsequent relaxation of the shortened cells.  相似文献

Growth rates of individual leaves attached to sunflower (Helianthus annuus) plants were measured experimentally under different levels of environmental productivity, modified by irradiance and nutrient conditions. The unfolding rate and final area of an individual leaf increased with increasing environmental productivity. The final area of an individual leaf also varied according to differences in leaf order. The declining pattern of relative leaf area growth rate (RLGR) varied with environmental productivity; leaves in a productive environment had a longer period of high sustained initial RLGR than leaves in a less productive environment. However, maximum RLGR was hardly influenced by leaf order or environmental factors such as irradiance and mineral nutrition. A group of the medaka,Oryzias latipes (Cyprinodontidae, orange-red variety, 25 males and 25 females), was kept in an aquarium, which was placed outdoors under natural conditions from December 1984 to January 1986. Locomotor activity at three layers (upper, middle, and lower layers) was recorded with a phototransistor system in each season. In summer, the fish showed typical diurnal activity at all three layers and the activity was greater than in other seasons. However, in autumn and winter, the fish became less active and showed relatively high activity at night at the upper or middle layer and diurnal activity at the lower layer. Nocturnal activity seemed to appear when the water temperature was decreased and the photoperiod was shortened. A free-running activity rhythm was also recorded under continuous darkness (DD) in each season; however, the fish showed clear free-running activity rhythms under DD only in summer.  相似文献

The purpose of the present study was to examine the distribution of subcutaneous fat and to derive several equations to predict body density (BD) using an amplitude modulation type of ultrasonoscope (A-mode ultrasonoscope, FUKUDA FT-100). Subjects were 188 male physical education major college students ranging in age from 18 to 24 years. Fifty subjects who were randomly selected out of the 188 subjects were measured for BD by the underwater weighing technique and were used to derive the equation for estimating the BD. Four points (scapular, triceps, suprailiac, and thigh) of subcutaneous fat which had been commonly selected, height, and weight were measured. The four measurements of fat for the 188 subjects indicated rather small means and small standard deviations respectively. Furthermore, histograms of those measurements tended to show a significant skewness for low values and deviated from the normal probability curve (p less than 0.01). Regarding the means, they were almost all the same except for suprailiac measurements. Suprailiac measurements showed more large values and were distributed rather more widely than the other measurements. Derivations of the multiple regression equations from anthropometric measurements were made using the Wherry-Doolittle test selection method (Clarke & Clarke, 1972). Four measurements (triceps, suprailiac, height, and weight) were selected by the Wherry-Doolittle method.  相似文献
