Sex chromosomes of the Japanese frog Rana rugosa are heteromorphic in the
male (XX/XY) or in the female (ZZ/ZW) in two geographic forms, whereas they
are still homomorphic in both sexes in two other forms (Hiroshima and
Isehara types). To make clear the origin and differentiation mechanisms of
the heteromorphic sex chromosomes, we isolated a sex-linked gene, ADP/ATP
translocase, and constructed a phylogenetic tree of the genes derived from
the sex chromosomes. The tree shows that the Hiroshima gene diverges first,
and the rest form two clusters: one includes the Y and Z genes and the
other includes the X, W, and Isehara genes. The Hiroshima gene shares more
sequence similarity with the Y and Z genes than with the X, W, and Isehara
genes. This suggests that the Y and Z sex chromosomes originate from the
Hiroshima type, whereas the X and W chromosomes originate from the
Isehara-type sex chromosome. Thus, we infer that hybridization between two
ancestral forms, with the Hiroshima-type sex chromosome in one and the
Isehara-type sex chromosome in the other, was the primary event causing
differentiation of the heteromorphic sex chromosomes.

Pyrolysis–gas chromatography (Py–GC) combined with on-line methylation was applied to a correlation analysis between the distributions of fatty acid components in the lipids of zooplankter individuals and those of ingested algae using principal component analysis (PCA). Py–GC in the presence of organic alkali, tetramethylammonium hydroxide (TMAH), was used to estimate the apparent distributions of fatty acid components contained in a single individual zooplankter weighing several tens of micrograms and a small sample size of ingested algae samples in the order of 10 μg. The observed fatty acid compositions were used as a database for the PCA in order to discriminate the zooplankton and ingested algae samples. The result obtained indicated that the fatty acid compositions of zooplankton individuals used in this work were significantly reflected in those of their ingested food in spite of some contribution from isomerization and/or elongation of fatty acid components during digestion of the ingested algae phytoplankton in living zooplankters. 相似文献

The RH gene family in vertebrates consists of four major genes (RH, RHAG, RHBG, and RHCG). They are thought to have emerged in the common ancestor of vertebrates after two rounds of whole genome duplication (2R-WGD). To analyze the detailed phylogenetic relationships within the RH gene family, we determined three types of cDNA sequence that belong to the RH gene family in lamprey (Lethenteron reissneri) and designated them as RHBG-like, RHCG-like1, and RHCG-like2. Phylogenetic analyses clearly showed that RHCG-like1 and RHCG-like2 genes, which were probably duplicated in the lamprey lineage, are orthologs of gnathostome RHCG. In contrast, the clear phylogenetic position of the RHBG-like gene could not be obtained. Probably some convergent events for cyclostome RHBG-like genes prevented the accurate identification of their phylogenetic positions. 相似文献

Abstract: We previously reported that an endogenous activating substance different from bovine serum albumin, phospholipids and heparin, exists in the extract from bovine pineal glands and that this substance interacts with tryptophan-5-monooxygenase under reducing conditions with sulfhydryl reagents, to stimulate monooxygenase activity. The present paper reports that the activating substance is of peptide nature; that it is sensitive to trypsin-digestion; and that it does not change the apparent K m 's for substrates, L-tryptophan and oxygen, and coenzyme, reduced biopterin or DMPH4 : but that it increases the V max 1.5- to 2.3-fold. These results suggest that an activating protein, present in some particles of the cell structure, activates tryptophan-5-monooxygenase under the regulation of a sulfhydryl compound. The apparent K m 's for reduced biopterin and DMPH4 were 77.2μM and 294 μM, respectively. The apparent K m 's for L-tryptophan and oxygen with reduced biopterin were 15.0 μM and 4.7%, respectively: with DMPH4 , they were 11.0 μM and 8.5%, respectively. Significant inhibition of both L-tryptophan and oxygen was observed with reduced biopterin, but not with DMPH4 (at the tested concentrations of up to 0.5 MM and 20%, respectively). 相似文献

The present study characterized the receptor‐dependent regulation of dendrite formation of noradrenaline (NA) and dopamine (DA) in cultured neurons obtained from embryonic day 16 rat cerebral cortex. Morphological diversity of cortical dendrites was analyzed on various features: dendrite initiation, dendrite outgrowth, and dendrite branching. Using a combination of immunocytochemical markers of dendrites and GABAergic neurons, we focused on the dendrite morphology of non‐GABAergic neurons. Our results showed that (1) NA inhibited the dendrite branching, (2) β adrenergic receptor (β‐AR) agonist inhibited the dendrite initiation, while promoted the dendrite outgrowth, (3) β1‐AR and β2‐AR were present in all the cultured neurons, and both agonists inhibited the dendrite initiation, while only β1‐AR agonist induced the dendrite branching; (4) DA inhibited the dendrite outgrowth, (5) D1 receptor agonist inhibited the dendrite initiation, while promoted the dendrite branching. In conclusion, this study compared the effects of NA, DA and their receptors and showed that NA and DA regulate different features on the dendrite formation of non‐GABAergic cortical neurons, depending on the receptors. © 2012 Wiley Periodicals, Inc. The two major chemoreceptors of Escherichia coli, Tsr and Tar, mediate opposite responses to the same changes in cytoplasmic pH (pH(i)). We set out to identify residues involved in pH(i) sensing to gain insight into the general mechanisms of signaling employed by the chemoreceptors. Characterization of various chimeras of Tsr and Tar localized the pH(i)-sensing region to Arg(259)-His(267) of Tar and Gly(261)-Asp(269) of Tsr. This region of Tar contains three charged residues (Arg(259)-Ser(261), Asp(263), and His(267)) that have counterparts of opposite charge in Tsr (Gly(261)-Glu(262), Arg(265), and Asp(269)). The replacement of all of the three charged residues in Tar or Arg(259)-Ser(260) alone by the corresponding residues of Tsr reversed the polarity of pH(i) response, whereas the replacement of Asp(263) or His(267) did not change the polarity but altered the time course of pH(i) response. These results suggest that the electrostatic properties of a short cytoplasmic region within the linker region that connects the second transmembrane helix to the first methylation helix is critical for switching the signaling state of the chemoreceptors during pH sensing. Tropical rainforests in South‐East Asia have been affected by climatic fluctuations during past glacial eras. To examine how the accompanying changes in land areas and temperature have affected the genetic properties of rainforest trees in the region, we investigated the phylogeographic patterns of a widespread dipterocarp species, Shorea leprosula. Two types of DNA markers were used: expressed sequence tag‐based simple sequence repeats and chloroplast DNA (cpDNA) sequence variations. Both sets of markers revealed clear genetic differentiation between populations in Borneo and those in the Malay Peninsula and Sumatra (Malay/Sumatra). However, in the south‐western part of Borneo, genetic admixture of the lineages was observed in the two marker types. Coalescent simulation based on cpDNA sequence variation suggested that the two lineages arose 0.28–0.09 million years before present and that following their divergence migration from Malay/Sumatra to Borneo strongly exceeded migration in the opposite direction. We conclude that the genetic structure of S. leprosula was largely formed during the middle Pleistocene and was subsequently modified by eastward migration across the subaerially exposed Sunda Shelf. 相似文献
