In the present note we have investigated the cytochemical localization of acetylcholinesterase (AChE) in the chick ciliary ganglion (CG) after post-ganglionic axotomy obtained by ablation of the eyeball. Preliminary results show at quite early stages after axotomy a remarkable reduction of cytoplasmic AChE, the residual one being localized in the rough endoplasmic reticulum. On the contrary synaptic areas, in particular those concerning the calyciform synapses, still show a marked AchE activity, similarly to what observed in physiological conditions. The decrease of cytoplasmic AChe in axotomized CG does suggest the possibility that such AChE undergoes to a topographical rearrangement moving towards the synaptic areas of ganglionic neurons.  相似文献

Abstract Potassium-limited cultures of Candida utilis grown with ammonium chloride as the sole nitrogen source attained a higher dry weight than similar cultures grown with sodium nitrate as the sole nitrogen source. This increase depended on the extracellular ammonia concentration and was not due to accumulation of storage polymers. We conclude that in this yeast ammonium ions can fulfill to some extent the physiological role of potassium ions and that the intracellular concentration of ammonium ions is predominantly determined by the ammonia concentration in the culture medium.  相似文献

We investigated the expression of the tumor necrosis factor (TNF) gene during development of mouse embryos, and observed its transient expression on Days 9 and 10 of gestation. We also detected a 25-kDa protein showing immunological cross-reactivity with mouse TNF antibody in an extract of 10-day embryos. These results suggest that TNF plays a role in mammalian ontogenesis.  相似文献

Biotin synthetase (BS) catalyses the biotransformation of dethiobiotin (DTB) to biotin. Here we report the cloning, characterization and expression of the gene encoding BS of Bacillus sphaericus. A recombinant plasmid pSB01, containing an 8.2-kb DNA fragment from B. sphaericus, was isolated by phenotypic complementation of an Escherichia coli bioB strain. Nucleotide sequence analysis of this fragment and N-terminal sequence determination of the recombinant protein product revealed that the bioB gene of B. sphaericus consists of a 996-bp open reading frame which is closely associated with at least one other gene. E. coli cells transformed with a bioB expression vector performed efficient bioconversion of DTB to biotin under defined culture conditions. Biotin production from transformed Bacillus subtilis and B. sphaericus recombinant strains was also demonstrated. Comparison of the amino acid sequences of BS from E. coli and B. sphaericus revealed extensive similarity.  相似文献

Altitudinal zonation of evergreen, deciduous and coniferous forests on Mt. Emei (3099 m asl, 29°34.5' N, 103°21.5' E), Sichuan, China was studied to understand the transition of vegetation zonation from tropical to temperate mountains in humid Asia. On the basis of quantitative data on floristic composition and community structure sampled at ten plots selected in different altitudes on the eastern slope of the mountain, forest zonation and the inter-relationships among different life-forms of trees in each zonal forest community were studied quantitatively. Three forest zones were identified physiognomically along the altitudinal gradient, viz. (i) the evergreen broad-leaved forest zone (660–1500 m asl), (ii) the mixed forest zone (1500–2500 m asl), and (iii) the coniferous forest zone (2500–3099 m asl). Great compositional changes were observed along elevation, and the zonal forest communities were characterized by their dominants and floristic composition. Maximum tree height decreased from 33 m at lower middle altitude (965 m asl) to 13 m near the summit (2945 m asl). There was no apparent deciduous forest zone along the altitudinal gradient, but true mixed forests of three life-forms (evergreen, deciduous, and coniferous) were formed around 2000–2500 m asl. Patches of deciduous forest were found in a lower part of the mixed forest zone, particularly on scree slopes, between 1450 m and 1900 m asl. These patches were dominated by the Tertiary relic deciduous trees, such as Davidia involucrata, Tetracentron sinense, and Cercidiphyllum japonicum var. sinense. High species diversity in the mixed forest zone resulted from the overlapping of different life-forms at middle altitudes, which is partly due to wider variety of temperature-altitude correlations. A comparison of the altitudinal zonation with the other east Asian mountain vegetation clarified that Mt. Emei is located exactly at the ecotone between tropical and temperate zonation types in eastern Asia.  相似文献

Human mesenchymal stem cells (hMSCs) are able to both self-replicate and differentiate into a variety of cell types. Fibroblast growth factor-2 (FGF-2) stimulates the growth of hMSCs in vitro, but its mechanisms have not been clarified yet. In this study, we investigated whether cellular senescence was involved in the stimulation of hMSCs growth by FGF-2 and the expression levels of transforming growth factor-beta1 and -beta2 (TGF-betas). Because hMSCs were induced cellular senescence due to long-term culture, FGF-2 decreased the percentage of senescent cells and suppressed G1 cell growth arrest through the suppression of p21(Cip1), p53, and p16(INK4a) mRNA expression levels. Furthermore, the levels of TGF-betas mRNA expression in hMSCs were increased by long-term culture, but FGF-2 suppressed the increase of TGF-beta2 mRNA expression due to long-term culture. These results suggest that FGF-2 suppresses the hMSCs cellular senescence dependent on the length of culture through down-regulation of TGF-beta2 expression.  相似文献
