Distribution and conservation of mobile elements in the genus Drosophila 总被引:13,自引:1,他引:12
Essentially nothing is known of the origin, mode of transmission, and
evolution of mobile elements within the genus Drosophila. To better
understand the evolutionary history of these mobile elements, we examined
the distribution and conservation of homologues to the P, I, gypsy, copia,
and F elements in 34 Drosophila species from three subgenera. Probes
specific for each element were prepared from D. melanogaster and hybridized
to genomic DNA. Filters were washed under conditions of increasing
stringency to estimate the similarity between D. melanogaster sequences and
their homologues in other species. The I element homologues show the most
limited distribution of all elements tested, being restricted to the
melanogaster species group. The P elements are found in many members of the
subgenus Sophophora but, with the notable exception of D. nasuta, are not
found in the other two subgenera. Copia-, gypsy-, and F-element homologues
are widespread in the genus, but their similarity to the D. melanogaster
probe differs markedly between species. The distribution of copia and P
elements and the conservation of the gypsy and P elements is inconsistent
with a model that postulates a single ancient origin for each type of
element followed by mating-dependent transmission. The data can be
explained by horizontal transmission of mobile elements between
reproductively isolated species.

Fusarium graminearum Schwabe (Fusarium head blight, FHB) and Puccinia triticina Eriks (leaf rust) are two major fungal pathogens posing a continuous threat to the wheat crop; consequently, identifying resistance genes from various sources is always of importance to wheat breeders. We identified tightly linked single nucleotide polymorphism (SNP) markers for the FHB resistance quantitative trait locus (QTL) Qfhs.pur-7EL and the leaf rust resistance locus Lr19 using genotyping-by-sequencing (GBS) in a wheat–tall wheatgrass introgression-derived recombinant inbred line (RIL) population. One thousand and seven hundred high-confidence SNPs were used to conduct the linkage and QTL analysis. Qfhs.pur-7EL was mapped to a 2.9 cM region containing four markers within a 43.6 cM segment of wheatgrass chromosome 7el2 that was translocated onto wheat chromosome 7DL. Lr19 from 7el1 was mapped to a 1.21 cM region containing two markers in the same area, in repulsion. Five lines were identified with the resistance-associated SNP alleles for Qfhs.pur-7EL and Lr19 in coupling. Two SNP markers in the Qfhs.pur-7EL region were converted into PCR-based KASP markers. Investigation of the genetic characteristics of the parental lines of this RIL population indicated that they are translocation lines in two different wheat cultivar genetic backgrounds instead of 7E–7D substitution lines in Thatcher wheat background, as previously reported in the literature. 相似文献

ABSTRACT. Troglocorys cava n. g., n. sp. is described from the feces of wild eastern chimpanzee, Pan troglodytes schweinfurthii, in Uganda. This new species has a spherical body with a frontal lobe, a long vestibulum, a cytoproct located at the posterior dorsal side of the body, an ovoid macronucleus, a contractile vacuole near the cytoproct, and a large concavity on the left surface of the body. Buccal ciliature is non‐retractable and consists of three ciliary zones: an adoral zone surrounding the vestibular opening, a dorso‐adoral zone extending transversely at the basis of the frontal lobe, and a vestibular zone longitudinally extending in a gently spiral curve to line the surface of the vestibulum. Two non‐retractable somatic ciliary zones comprise arches over the body surface: a short dorsal ciliary arch extending transversely at the basis of the frontal lobe and a wide C‐shaped left ciliary arch in the left concavity. Because of the presence of three ciliary zones in the non‐retractable buccal ciliature, the present genus might be a member of the family Blepharocorythidae, but the large left concavity and the C‐shaped left ciliary arch are unique, such structures have never been described from other blepharocorythids. 相似文献

A method is described in which selective silver staining of Alzheimer's neurofibrillary changes is combined with staining of cell nuclei, Nissl material, and lipofuscin granules. Formalin fixed, paraffin embedded sections of human autopsy tissue are silver stained according to a method proposed by Gallyas. Lipofuscin is stained by crotonaldehyde fuchsin following performic acid oxidation. Nissl substance is visualized by either Darrow red or gallocyanin-chrome alum staining. Architectonic units showing the specific pathology and the neuronal types prone to develop the neurofibrillary changes can be recognized using this technique. 相似文献

Summary The introduction of molecular biology methodologies to plant improvement programs offers an invaluable opportunity for extensive germplasm characterization. However, the detection of adequate DNA polymorphism in self-pollinating species remains on obstacle. We have optimized a denaturing-gradient-gel electrophoresis (DGGE) system which, when used in combination with random amplified polymorphic DNA (RAPD) analysis, greatly facilitates the detection of reproducible DNA polymorphism among closely related plant lines. We have used this approach to estimate pedigree relationships among a spectrum of plant materials in wheat, barley and oat. Based on analysis with one or two primers, we were able to distinguish soft from hard winter wheat, and 2-rowed from 6-rowed barley. Further analysis with additional primers allowed resolution of polymorpisms even among closely related lines in highly selected populations. We placed 17 cultivars of oat into two distinct clusters that differed significantly from previous oat pedigree assessments. We believe that DGGE-RAPD is a superior method for detecting DNA polymorphism when compared to RFLP, agarose-RAPD, or polyacrylamide-RAPD methods. 相似文献

The worldwide increase in the use of antibiotics as an integral part of poultry and livestock production industry has recently received increasing attention as a contributory factor in the international emergence of antibiotic-resistant bacteria in human beings. To gauge the presence of the aforementioned scenario in the Indian context, a preliminary survey was conducted to assess the use of chlortetracycline (CTC) in 12 commercial layer farms and to quantify and confirm its residue in the egg. Samples of feed and eggs were collected at day 0 (prior to CTC addition), 3rd, 5th and 7th day during treatment and on the 9th and 14th day (2nd and 7th day after withdrawal of CTC) from each of the 12 commercial poultry farms studied. Concentration of CTC in feed was significantly (P<0.01) high on the 3rd, 5th and 7th day. On the 9th day and 14th day CTC concentration in feed was significantly (P<0.01) lower compared to the earlier 3 days studied. A highly significant difference (P<0.01) of the antibiotic residue in egg was observed in all the 5 days with high residual levels of CTC in egg. CTC in feed and its residue in egg were detected even on the 9th and 14th day respectively. 相似文献

Genetics in the post-genomic period is shifting from structural to functional genetics or genomics. Meanwhile, the use of
twins is largely expanding from traditional heritability estimation for disease phenotypes to the study of both diseases and
various molecular phenotypes, such as the regulatory phenotypes in functional genomics concerning gene expression and regulation,
by engaging both classical twin design and marker-based gene mapping techniques in genetic epidemiology. New research designs
have been proposed for making novel uses of twins in studying the molecular basis in the epigenetics of human diseases. Besides,
twins not only serve as ideal samples for disease gene mapping using conventional genetic markers but also represent an excellent
model for associating DNA copy number variations, a structural genetic marker, with human diseases. It is believed that, with
the rapid development in biotechniques and new advances in bioinformatics, the unique samples of twins will make new contributions
to our understanding of the nature and nurture in complex disease development and in human health. This paper aims at summarizing
the new uses of twins in current genetic studies and suggesting novel proposes together with useful design and analytical
