The method of D-dimer quantification in the human blood plasma has been developed using monoclonal antibodies 111-3b and II-4d. The method has been verified on the blood plasma of the patients with ischemic heart disease with and without stenocardia and with hypertension. The results showed that at ischemic heart disease with and without stenocardia and at hypertension the quantities of D-dimer in the blood plasma were generally less than the highest normal level 500 ng/ml (64.3%, 76.2% and 95%, correspondingly). The semiquantitative measurements of soluble fibrin levels in blood plasmas of the patients with ischemic heart disease and hypertension have been performed. It has been shown that the quantity of soluble fibrin at these diseases range greatly from < 0.03 mg/ml to 0.15 mg/ml. There was no correlation between the quantities of D-dimer and soluble fibrin in blood plasmas of the patients. Electrophoresis in PAAG with SDS showed that the soluble fibrin at these diseases had the mo- lecular mass of the fibrin (ogen). Thus the soluble fibrin in blood plasmas analysed consisted mainly of fibrin desAA oligomers (may be with fibrinogen incorporation) which are not stabilized by the factor XIIIa. 相似文献

The main function of the chaperone GroEL is to prevent nonspecific association of nonnative protein chains and provide their correct folding. In the present work, the renaturation kinetics of three globular proteins (human alpha-lactalbumin, bovine carbonic anhydrase, and yeast phosphoglycerate kinase) in the presence of different molar excess of GroEL (up to 10-fold) was studied. It was shown that the formation of the native structure during the refolding of these proteins is retarded with an increase in GroEL molar excess due to the interaction of kinetic protein intermediates with the chaperone. Mg(2+)-ATP and Mg(2+)-ADP weaken this interaction and decrease the retarding effect of GroEL on the protein refolding kinetics. The theoretical modeling of protein folding in the presence of GroEL showed that the experimentally observed linear increase in the protein refolding half-time with increasing molar excess of GroEL must occur only when the protein adopts its native structure outside of GroEL (i.e. in the free state), while the refolding of the protein in the complex with GroEL is inhibited. The dissociation constants of GroEL complexed with the kinetic intermediates of the proteins studied were evaluated, and a simple mechanism of the functioning of GroEL as a molecular chaperone was proposed. 相似文献

The nucleotide sequence of the variant of human immunodeficiency virus of type 1 (HIV-1), mostly widespread on the territory of the Novosibirsk region, was determined. The analysis of the nucleotide sequence confirmed that this variant belonged to HIV-1 of subtype A. The HIV-1 recombinant variant of subtype envB/envA with the recombination area within the second conservative region C2 of gene env, so far unknown, was detected and characterized. In HIV-1 the area at the beginning of gene env (5'-env) was found to belong to subtype B and the sequence at the end of gene env (3'-env), to subtype A. The analysis of the amino acid sequence of the third variable region of gene env demonstrated that the viruses under study belonged to macrophagotropic "slow/low" variants, characterized by low replication speed. The analysis of nucleotide sequences of the isolated variants of HIV-1 revealed their close genetic relationship with HIV-1 isolates circulating on the territory of Ukraine. 相似文献

The main idea of the authors' paper is to propose the most reasonable way of actively introducing the digital principle into the traditional roentgenological section of radiation diagnosis. For this, a luminophore digital radiography system has been chosen. The authors of the paper give a full-scale assessment and appropriate recommendations for its use. The paper essentially discusses the entire complex of matters that permit assessment whether its sound use is possible in regional and municipal health care systems. This is both a section devoted to a dose load, by making a comparative assessment of luminophore radiography and "the green system" and a study of different clinical diseases (456 cases). In their study, the authors have applied an original principle in the formation of an image obtained and some other approaches in order to make a comprehensive assessment of this method. In the authors' opinion, luminophore radiography has a variety of advantages. Firstly, this technique can be simultaneously applied to several nondigital apparatuses, including those available in the ward and it shows a rather diagnostic effectiveness and economic profitability, yields a qualitative image of varying density tissues upon single exposure, and has some other capacities of the CR system as a digital technique. All this things considered, the authors consider that luminophore radiography may be one of the main ways of introducing a digital technique into the conventional roentgenological section of radiation diagnosis at the level of regional and municipal heath care systems. 相似文献

Mutant strains of Penicillium roquefortii VKM F-141 and P. Fellutanum VKM F-1073 were obtained by mutagenesis induced by ultraviolet irradiation, N-methyl-N-nitronitrosoguanidine and bromouracil. By the rates of alkaloid production, the mutant strains can be divided into three groups: 1) unable to synthesize alkaloids; 2) with a high rate of biosynthesis; 3) with changed alkaloid composition. A comparative analysis of the individual and joint influence of bovine prolactin (BPRL) and bovine somatotropin (BST) on DNA synthesis in cultured bovine granulosa cells from follicles of 3-5 mm in diameter was conducted, and a possibility for regulation of BPRL binding to receptors on the cells by BST was examined. Despite a unidirectional growth-promoting action of BPRL and BST on granulosa cells, their joint mitogenic effect on the cells was not additive at low concentrations of BPRL. The addition of BST to the culture medium did not alter the biphasic character of dependence of DNA synthesis in the cells on BPRL concentration, but increased the maximum effective concentration of the latter. When culturing granulosa cells with BST, the absence of its influence on the level of BPRL specific binding to the cells was shown. This fact suggests that BST modulating action on the mitogenic effect of BPRL is not a result of a change in the number of free receptors for PRL on granulosa cells. The results obtained are discussed in relation to the notion of similarity of receptor functional properties and intracellular signal ways for PRL and ST. 相似文献

The spectrum and frequency of mutations of exon 7 of the gene for phenylalanine hydroxylase (PAH) were studied in 34 phenylketonuria (PKU) patients living in Novosibirsk oblast. The five most prevalent mutations constituted 17.64% of defective alleles: R243Q (1.47%), R252W (1.47%), R261Q (5.88%), E280K (1.47%), and P281L (7.35%). Yolk globules in developing oocytes of Tilapia mosambique are formed by two processes: 1) biosynthetical activity of oocyte organoides; 2) vitellogenin migration by micropinocytosis and its further transformation. Undoubtedly, yolk globules of endogenic and exogenic origin are fused. The primary yolk globules are spherical, and the secondary ones are lobular. The latter originate by incorporating the former. The fast growth of the late vitellogenic stage oocytes occurs as a result of active migration of primary yolk globules into the central part of the oocyte and as their association with the secondary yolk globules. The effect of melatonin (0.1 microM) on the contractile response to electrical field stimulation (EFS) of the juvenile rat tail artery segment was studied. The spontaneous decrease in reactivity of the segment observed in the course of our experiments was accompanied with a melatonin-evoked increase in the reactivity which was proportional to this decrease and was not connected with a sensitization of the segment to the substance. Perfusion of the segment with an acidic solution leads to a more pronounced inhibition of the response as well as to a greater melatonin-evoked protentiation of the response. Noradrenaline-evoked response was not augmented by melatonin. The results suggest that the potentiating effect of melatonin on the EFS-evoked response of the juvenile rat tail artery depends on degree of the change in the artery reactivity and was not due to change in sensitivity of postjunctional membrane to noradrenaline. 相似文献
