Association mapping of salt tolerance in barley (Hordeum vulgare L.)   总被引:1,自引:0,他引:1  
A spring barley collection of 192 genotypes from a wide geographical range was used to identify quantitative trait loci (QTLs) for salt tolerance traits by means of an association mapping approach using a thousand SNP marker set. Linkage disequilibrium (LD) decay was found with marker distances spanning 2–8 cM depending on the methods used to account for population structure and genetic relatedness between genotypes. The association panel showed large variation for traits that were highly heritable under salt stress, including biomass production, chlorophyll content, plant height, tiller number, leaf senescence and shoot Na+, shoot Cl? and shoot, root Na+/K+ contents. The significant correlations between these traits and salt tolerance (defined as the biomass produced under salt stress relative to the biomass produced under control conditions) indicate that these traits contribute to (components of) salt tolerance. Association mapping was performed using several methods to account for population structure and minimize false-positive associations. This resulted in the identification of a number of genomic regions that strongly influenced salt tolerance and ion homeostasis, with a major QTL controlling salt tolerance on chromosome 6H, and a strong QTL for ion contents on chromosome 4H.  相似文献

Background

Acetylcholine, the primary parasympathetic neurotransmitter in the airways, plays an important role in bronchoconstriction and mucus production. Recently, it has been shown that acetylcholine, by acting on muscarinic receptors, is also involved in airway inflammation and remodelling. The mechanism(s) by which muscarinic receptors regulate inflammatory responses are, however, still unknown.

Methods

The present study was aimed at characterizing the effect of muscarinic receptor stimulation on cytokine secretion by human airway smooth muscle cells (hASMc) and to dissect the intracellular signalling mechanisms involved. hASMc expressing functional muscarinic M2 and M3 receptors were stimulated with the muscarinic receptor agonist methacholine, alone, and in combination with cigarette smoke extract (CSE), TNF-α, PDGF-AB or IL-1β.

Results

Muscarinic receptor stimulation induced modest IL-8 secretion by itself, yet augmented IL-8 secretion in combination with CSE, TNF-α or PDGF-AB, but not with IL-1β. Pretreatment with GF109203X, a protein kinase C (PKC) inhibitor, completely normalized the effect of methacholine on CSE-induced IL-8 secretion, whereas PMA, a PKC activator, mimicked the effects of methacholine, inducing IL-8 secretion and augmenting the effects of CSE. Similar inhibition was observed using inhibitors of IκB-kinase-2 (SC514) and MEK1/2 (U0126), both downstream effectors of PKC. Accordingly, western blot analysis revealed that methacholine augmented the degradation of IκBα and the phosphorylation of ERK1/2 in combination with CSE, but not with IL-1β in hASMc.

Conclusions

We conclude that muscarinic receptors facilitate CSE-induced IL-8 secretion by hASMc via PKC dependent activation of IκBα and ERK1/2. This mechanism could be of importance for COPD patients using anticholinergics.  相似文献

Livestock manure contributes considerably to global emissions of ammonia (NH3) and greenhouse gases (GHG), especially methane (CH4) and nitrous oxide (N2O). Various measures have been developed to mitigate these emissions, but most of these focus on one specific gas and/or emission source. Here, we present a meta‐analysis and integrated assessment of the effects of mitigation measures on NH3, CH4 and (direct and indirect) N2O emissions from the whole manure management chain. We analysed the effects of mitigation technologies on NH3, CH4 and N2O emissions from individual sources statistically using results of 126 published studies. Whole‐chain effects on NH3 and GHG emissions were assessed through scenario analysis. Significant NH3 reduction efficiencies were observed for (i) housing via lowering the dietary crude protein (CP) content (24–65%, compared to the reference situation), for (ii) external slurry storages via acidification (83%) and covers of straw (78%) or artificial films (98%), for (iii) solid manure storages via compaction and covering (61%, compared to composting), and for (iv) manure application through band spreading (55%, compared to surface application), incorporation (70%) and injection (80%). Acidification decreased CH4 emissions from stored slurry by 87%. Significant increases in N2O emissions were found for straw‐covered slurry storages (by two orders of magnitude) and manure injection (by 26–199%). These side‐effects of straw covers and slurry injection on N2O emission were relatively small when considering the total GHG emissions from the manure chain. Lowering the CP content of feed and acidifying slurry are strategies that consistently reduce NH3 and GHG emissions in the whole chain. Other strategies may reduce emissions of a specific gas or emissions source, by which there is a risk of unwanted trade‐offs in the manure management chain. Proper farm‐scale combinations of mitigation measures are important to minimize impacts of livestock production on global emissions of NH3 and GHG.  相似文献

The construction of three dams induced large changes in the tide range in the Eastern Scheldt, a tidal inlet in the southwestern part of the Netherlands. In 1986 the mean high tide level was reduced by almost one metre. This excluded the greater part of the medium-high marshes almost completely from tidal flooding.The absence of tidal flooding in the summer of 1986, combined with a net precipitation deficit, increased the bulk density of the sediment irreversibly from 400 to 530 kg m–3 in the top 5 cm. The subsidence of the backmarshes varied from 1 to 8 cm.The loss of moisture allowed oxygen to diffuse into the initial reducing sediment. This altered the geochemistry of the sediment significantly. The redoxcline was lowered from a mean depth of about 15 cm in 1985 to 20–30 cm in the summer of 1986. Evidence of pyrite oxidation within a narrow depth interval of 15–30 cm was obtained from the change in the composition of pore waters. The rapid increase in redox potentials (up to 600 mV) and total dissolved iron (up to 5 mM 1–1) and SO inf4 sup2– (up to 65 mM 1–1) and the decrease in pH (up to 4.5–2.5) all suggest a rapid oxidation of pyrite. Acidic conditions were found only in sediments with low contents of calcium carbonate and high contents of pyrite. The importance of seasonal changes in redox processes on the partial decalcification of the salt marsh sediments is discussed.The established of a new tide range was reflected in the pH and Eh of the sediment. In December 1987 the depth profiles of pH and Eh were again close to those observed in 1985.  相似文献

Denitrification in the top and sub soil of grassland on peat soils   总被引:2,自引:0,他引:2  
Denitrification is an important process in the nitrogen (N) balance of intensively managed grassland, especially on poorly drained peat soils. Aim of this study was to quantify the N loss through denitrification in the top and sub soil of grassland on peat soils. Sampling took place at 2 sites with both control (0 N) and N fertilised (+ N) treatments. Main difference between the sites was the ground water level. Denitrification was measured on a weekly basis for 2 years with a soil core incubation technique using acetylene (C2H2) inhibition. Soil cores were taken from the top soil (0–20 cm depth) and the sub soil (20–40 cm depth) and incubated in containers for 24 hours. The denitrification rate was calculated from the nitrous oxide production between 4 and 24 hours of incubation. Denitrification capacities of the soils and the soil layers were also determined.The top soil was the major layer for denitrification with losses ranging from 9 to 26 kg N ha–1 yr–1 from the O N treatment. Losses from the top soil of the + N treatment ranged from 13 to 49 kg N ha–1 yr–1. The sub soil contributed, on average, 20% of the total denitrification losses from the 0–40 layer. Losses from the 0–40 cm layer were 2 times higher on the + N treatment than on the O N treatment and totalled up to 70 kg N ha–1 yr–1. Significant correlation coefficients were found between denitrification activity on the one hand, and ground water level, water filled pore space and nitrate content on the other, in the top soil but not in the sub soil. The denitrification capacity experiment showed that the availability of easily decomposable organic carbon was an important limiting factor for the denitrification activity in the sub soil of these peat soils.  相似文献

Background

In this study the one and six months effects of the computer-tailored YouRAction (targeting individual level determinants) and YouRAction+e (targeting in addition perceived environmental determinants) on compliance with the moderate-to-vigorous physical activity (MVPA) guideline and weight status are examined. In addition the use and appreciation of both interventions are studied.

Methods

A three-armed cluster randomized trial was conducted in 2009–2010 with measurements at baseline, one and six months post intervention. School classes were assigned to one of the study arms (YouRaction, YouRAction+e and Generic Information (GI) control group). MVPA was derived from self-reports at baseline, one and six months post intervention. Body Mass Index and waist circumference were measured at baseline and six months post intervention in a random sub-sample of the population. Use of the interventions was measured by webserver logs and appreciation by self-reports. Multilevel regression analyses were conducted to study the effects of the intervention against the GI control group. ANOVA''s and chi-square tests were used to describe differences in use and appreciation between study arms.

Results

There were no statistically significant intervention effects on compliance with the MVPA guideline, overweight or WC. Access to the full intervention was significantly lower for YouRAction (24.0%) and YouRAction+e (21.7%) compared to the GI (54.4%).

Conclusion

This study could not demonstrate that the YouRAction and YouRAction+e interventions were effective in promoting MVPA or improve anthropometric outcomes among adolescents, compared to generic information. Insufficient use and exposure to the intervention content may be an explanation for the lack of effects.

Hydrodynamic forces and sediment discharges determine the sedimentary environment and surface morphology of the intertidal area in the Westerschelde estuary in the S.W. Netherlands. Sandflats (clay content <8%) are found in the central part, mudflats (>8% clay) and especially salt marshes (>15% clay) occur in sheltered places.The semi-terrestrial soil of the intertidal area is a complex environment where hydrosphere, biosphere, lithosphere, and atmosphere interact. Surface morphology and the semi-diurnal tide may significantly modify this interaction pattern. Organic matter and constituents of the clay-size fraction are the most reactive compounds in the soil.The pathway and rate of organic matter decomposition determines the nature of many biogeochemical processes. Sulfur transformations are dominant processes in the sulfidic grey-black coloured horizons underneath the generally thin yellow-brown surface layer. The sulfur geochemistry also affects the sorption-behaviour of metals. Oxygen intrusion by diffusion, burrowing fauna and plant roots cause a local and sometimes temporary oxidation of reduced inorganic compounds. Thus, in the surface layers, and in and around tubes and roots at greater depth in the soil, a dynamic cycling occurs of redox active compounds, which affects the sorption mechanism ofe.g. metals-ions.  相似文献
