The complement system is an essential part of the innate immune system by acting as a first line of defense which is stabilized by properdin, the sole known positive regulator of the alternative complement pathway. Dysregulation of complement can promote a diversity of human inflammatory diseases which are treated by complement inhibitors. Here, we generated a novel blocking monoclonal antibody (mAb) against properdin and devised a new diagnostic assay for this important complement regulator. Mouse mAb 1340 specifically detected native properdin from human samples with high avidity. MAb 1340 inhibited specifically the alternative complement mediated cell lysis within a concentration range of 1–10 µg/mL. Thus, in vitro anti-properdin mAb 1340 was up to fifteen times more efficient in blocking the complement system as compared to anti-C5 or anti-Ba antibodies. Computer-assisted modelling suggested a three-dimensional binding epitope in a properdin-C3(H2O)-clusterin complex to be responsible for the inhibition. Recovery of properdin in a newly established sandwich ELISA using mAb 1340 was determined at 80–125% for blood sample dilutions above 1∶50. Reproducibility assays showed a variation below 25% at dilutions less than 1∶1,000. Systemic properdin concentrations of healthy controls and patients with age-related macular degeneration or rheumatic diseases were all in the range of 13–30 µg/mL and did not reveal significant differences. These initial results encourage further investigation into the functional role of properdin in the development, progression and treatment of diseases related to the alternative complement pathway. Thus, mAb 1340 represents a potent properdin inhibitor suitable for further research to understand the exact mechanisms how properdin activates the complement C3-convertase and to determine quantitative levels of properdin in biological samples.  相似文献

We suggest that interactions with strangers at work influence the likelihood of depressive disorders, as they serve as an environmental stressor, which are a necessary condition for the onset of depression according to diathesis-stress models of depression. We examined a large dataset (N = 76,563 in K = 196 occupations) from the German pension insurance program and the Occupational Information Network dataset on occupational characteristics. We used a multilevel framework with individuals and occupations as levels of analysis. We found that occupational environments influence employees’ risks of depression. In line with the quotation that ‘hell is other people’ frequent conflictual contacts were related to greater likelihoods of depression in both males and females (OR = 1.14, p<.05). However, interactions with the public were related to greater likelihoods of depression for males but lower likelihoods of depression for females (ORintercation = 1.21, p<.01). We theorize that some occupations may involve interpersonal experiences with negative emotional tones that make functional coping difficult and increase the risk of depression. In other occupations, these experiences have neutral tones and allow for functional coping strategies. Functional strategies are more often found in women than in men.  相似文献

Alterations in bone remodeling are a major public health issue, as therapeutic options for widespread bone disorders such as osteoporosis and tumor-induced osteolysis are still limited. Therefore, a detailed understanding of the regulatory mechanism governing bone cell differentiation in health and disease are of utmost clinical importance. Here we report a novel function of carcinoembryonic antigen-related cell adhesion molecule 1 (CEACAM1), a member of the immunoglobulin superfamily involved in inflammation and tumorigenesis, in the physiologic regulation of bone remodeling. Assessing the expression of all members of the murine Ceacam family in bone tissue and marrow, we found CEACAM1 and CEACAM10 to be differentially expressed in both bone-forming osteoblasts and bone-resorbing osteoclasts. While Ceacam10-deficient mice displayed no alteration in structural bone parameters, static histomorphometry demonstrated a reduced trabecular bone mass in mice lacking CEACAM1. Furthermore, cellular and dynamic histomorphometry revealed an increased osteoclast formation in Ceacam1-deficient mice, while osteoblast parameters and the bone formation rate remained unchanged. In line with these findings, we detected accelerated osteoclastogenesis in Ceacam1-deficient bone marrow cells, while osteoblast differentiation, as determined by mineralization and alkaline phosphatase assays, was not affected. Therefore, our results provide in vivo and in vitro evidence for a physiologic role of CEACAM1 in the regulation of osteoclastogenesis.  相似文献

The Massim, a cultural region that includes the southeastern tip of mainland Papua New Guinea (PNG) and nearby PNG offshore islands, is renowned for a trading network called Kula, in which different valuable items circulate in different directions among some of the islands. Although the Massim has been a focus of anthropological investigation since the pioneering work of Malinowski in 1922, the genetic background of its inhabitants remains relatively unexplored. To characterize the Massim genomically, we generated genome-wide SNP data from 192 individuals from 15 groups spanning the entire region. Analyzing these together with comparative data, we found that all Massim individuals have variable Papuan-related (indigenous) and Austronesian-related (arriving ∼3,000 years ago) ancestries. Individuals from Rossel Island in southern Massim, speaking an isolate Papuan language, have the highest amount of a distinct Papuan ancestry. We also investigated the recent contact via sharing of identical by descent (IBD) genomic segments and found that Austronesian-related IBD tracts are widely distributed geographically, but Papuan-related tracts are shared exclusively between the PNG mainland and Massim, and between the Bismarck and Solomon Archipelagoes. Moreover, the Kula-practicing groups of the Massim show higher IBD sharing among themselves than do groups that do not participate in Kula. This higher sharing predates the formation of Kula, suggesting that extensive contact between these groups since the Austronesian settlement may have facilitated the formation of Kula. Our study provides the first comprehensive genome-wide assessment of Massim inhabitants and new insights into the fascinating Kula system.  相似文献

Melanin in the skin can be divided into eumelanin and pheomelanin subtypes. Simultaneous quantification of these subtypes could clarify their relation to skin type and skin cancer development. We describe a novel, sensitive liquid chromatography–tandem mass spectrometry method to quantify two eumelanin markers, pyrrole‐2,3,5‐tricarboxylic acid (PTCA) and pyrrole‐2,3‐dicarboxylic acid (PDCA), and two pheomelanin markers, thiazole‐4,5‐dicarboxylic acid (TDCA) and thiazole‐2,4,5 tricarboxylic acid (TTCA), performed in a single run using the same biopsy. Volunteers with either Fitzpatrick skin type (FST) I/II or III/IV (n = 30) each provided a 4‐mm punch biopsy from the buttock. Upon analysis, the FST I + II group had significantly less of all four melanin biomarkers (PTCA, 0.75 ng/mm2; PDCA, 0.08 ng/mm2; TTCA, 0.24 ng/mm2; and TDCA, 0.10 ng/mm2) versus the FST III + IV group (PTCA, 4.89 ng/mm2; PDCA, 0.22 ng/mm2; TTCA, 2.61 ng/mm2; and TDCA, 0.72 ng/mm2), p ≤ 0.003. We find that this new LC‐MS/MS method is sensitive enough to quantify eumelanin and pheomelanin markers even in the lightest skin types.  相似文献

255 instances of “brow raise” filmed in three cultures in unstaged social interactions were analyzed on different levels using the “Facial Action Coding System” (FACS, Ekman & Friesen 1978). Contraction and slackening of the M. frontalis, pars medialis et lateralis are involved, and show temporal constancy in all three cultures, creating a pattern with a typical movement configuration. This configuration is discussed as a universal prerequisite for stimulus generation perception. The total time of contraction varies with contextual features, and brow raise in openings is longer than during interactions. An analysis of co-occurrence of other facial movements showed universal patterns occurring in all three cultures. Brow raise is most often accompanied by a smile. The antithesis of this “eyebrow flash”, both in neuromuscular and semantic aspect, is brought about by the action of the M. corrugator supercilii, lowering the brows and pulling them together. In addition, a set of functional patterns could be identified in all three cultures, ranging from a factual “yes” to a “yes to social contact”. Thus, the eyebrow flash can be interpreted as a “social marking-tool” which emphasizes the meaning of other facial cues, head movements and even verbal statements.  相似文献

Zusammenfassung In Regenwäldern Südostliberias nahe Zwedru (Tchien) 6° 12 N / 8° 11 W, Grand Gedeh County, wurde 1981 – 1984 neunmal, durchweg in der Trockenzeit, ein offensichtlich unbekannter Bülbül beobachtet. Ein Exemplar wurde gesammelt. Die Art wird alsPhyllastrephus leucolepis spec. nov. beschrieben. Das Verhalten, mit geöffneten Flügeln durch die Zweige zu hüpfen, legt nahe, daß das arteigene, auffällige Flügelmuster, zwei helle Binden, als optisches Signal dient. Habitat und Baumartenzusammensetzung im Übergangsbereich zwischen immergrünem und halbimmergrünem tropischem Regenwald werden beschrieben. Die Möglichkeit der Artgleichheit mitPhyllastrephus icterinus wird diskutiert, aber auf Grund von Färbungs- und Verhaltensunterschieden sowie mehrerer Beobachtungsorte als unwahrscheinlich betrachtet.
A new species of bulbul from West Africa (Aves, Pycnonotidae)
Summary In the rain forests of Southeast-Liberia near Zwedru, Tchienregion, 6° 12 N / 8° 11 W, Grand Gedeh County, between 1981 and 1984, always in the dry season, an obviously unknown Bulbul was sighted. One specimen was collected. The species is described asPhyllastrephus leucolepis spec. nov. The wing length of the holotype was fresh 74/75 mm; dry 71/72 mm; Culmen 17 mm, Tarsus 23 mm. The colour of the head, mantle, tail and underside corresponds more or less toPhyllastrephus icterinus. The wing pattern is uniquely characteristic. Over all the feathers of the Alula, the median and greater primary and secondary coverts, there is a light section in the distal parts, which is bordered by dark feathertips. Through the outer parts of all the primaries and secondaries there is an additional broad light wing bar, darktipped at the end of the feathers. The coloration of the outerwebs of the primaries and secondaries inleucolepis is browner, less olivegreen than inicterinus. The coloration of the innerwebs is darkbrown, inicterinus nearly black. Wings and tail are more rusty coloured. The behaviour of jumping with open wings through the twigs shows obviously that the wing pattern is used as an optical signal in the dark forest.Habitat and tree species composition in the transitional zone between evergreen and semideciduous tropical rain forest are described. The possibility of synonymity toPhyllastrephus icterinus is discussed, but due to differences in coloration and behaviour as well as the fact that several observations were made in different places, it is considered unlikely.
