Double-strand break repair is executed by two major repair pathways: non-homologous end joining (NHEJ) and homologous recombination (HR). Whereas NHEJ contributes to the repair of ionizing radiation (IR)-induced double strand breaks (DSBs) throughout the cell cycle, HR acts predominantly during the S and G2 phases of the cell cycle. The rare-cutting restriction endonuclease, I-SceI, is in common use to study the repair of site-specific chromosomal DSBs in vertebrate cells. To facilitate analysis of I-SceI-induced DSB repair, we have developed a stably expressed I-SceI fusion protein that enables precise temporal control of I-SceI activation, and correspondingly tight control of the timing of onset of site-specific chromosome breakage. I-SceI-induced HR showed a strong, positive linear correlation with the percentage of cells in S phase, and was negatively correlated with the G1 fraction. Acute depletion of BRCA1, a key regulator of HR, disrupted the relationship between S phase fraction and I-SceI-induced HR, consistent with the hypothesis that BRCA1 regulates HR during S phase.  相似文献

Blood-brain barrier (BBB) failure occurs in many neurological diseases and is caused in part by activation of proinflammatory factors including matrix metalloproteinases. Counterbalancing, "BBB protective" cascades have recently been described, including NO-mediated interleukin 6 release by glia. Interleukin 6 has been shown to trigger production of matrix metalloproteinase inhibitors such as alpha2-macroglobulin (alpha2M). We hypothesized that BBB failure may result in increased alpha(2)M release by perivascular astrocytes. This was initially tested in patients undergoing iatrogenic BBB disruption by hyperosmotic mannitol for intra-arterial chemotherapy of brain tumors. Serum samples revealed significantly increased levels of alpha2M at 4 h after BBB disruption by hyperosmotic mannitol. In parallel in vitro experiments, we observed a similar increase of alpha2M release by astrocytes under conditions mimicking BBB failure and perivascular edema. For both experiments, protein analysis was initially performed by bidimensional gel electrophoresis and mass spectrometry followed by Western blotting immunodetection. We conclude that, in addition to proinflammatory changes, BBB failure may also trigger protective release of alpha2M by perivascular astrocytes as well as peripheral source.  相似文献

Histone H2AX phosphorylation on a C-terminal serine residue to form “γ-H2AX” is a critical early event in the chromatin response to chromosomal DNA double strand breaks in eukaryotes. In mammalian cells, γ-H2AX is formed when H2AX is phosphorylated on serine 139 by ATM or by other DNA damage response kinases. H2AX prevents genomic instability and tumorigenesis, and supports class-switch recombination at immunoglobulin heavy chain loci in mammals. We showed previously that H2AX controls double strand break repair by homologous recombination (HR) between sister chromatids. the HR functions of H2AX are mediated by interaction of γ-H2AX with the chromatin-associated adaptor protein MDC1. H2AX is potentially subject to additional post-translational modifications associated with the DNA damage response and with other chromatin functions. To test this idea, we used mass spectroscopy to identify H2AX residues additional to serine 139 that are post-translationally modified following exposure of cells to ionizing radiation (IR) and identified several new IR-responsive residues of H2AX. We determined the impact of IR-responsive H2AX residues on cellular resistance to IR and on H2AX-dependent HR, and also analyzed the contribution to HR of other known or potential post-translationally modified residues of H2AX. The results suggest that the HR and IR-resistance functions of H2AX are controlled in large part by specific MDC1-interacting residues of H2AX, but that additional H2AX residues modulate these core functions of H2AX.Key words: H2AX, homologous recombination, ionizing radiation, double strand break repair, histone, histone code, post-translational modification, chromatin, DNA repair  相似文献

The variation of nutrients over decadal timescales south of the polar front in the Southern Ocean is poorly known because of a lack of continuous observational data in this area. We examined data from long-term continuous hydrographic monitoring of 43 years (1965–2008) in the Indian sector of the Southern Ocean, via the resupply of Antarctic stations under the Japanese Antarctic Research Expedition and Australian Antarctic Research Expedition. We found significant increasing trends in phosphate and nitrate, and a decreasing trend in apparent oxygen utilization (AOU) in intermediate water (neutral density = 27.8–28.1 kgm−3) south of the polar front. The rates of phosphate and nitrate increase are 0.004 µmol yr−1 and 0.02 µmol yr−1, respectively. The rate of decline of AOU was 0.32 µmol yr−1. One reason for this phosphate and nitrate increase and AOU decline is reduced horizontal advection of North Atlantic Deep Water, which is characterized by low nutrients and high AOU. The relationship between climate change and nutrient variability remains obscure, emphasizing the importance of long-term monitoring.  相似文献

The infection rate of eugregarine parasites, Cephaloidophora pacifica, within the digestive tract of larval Antarctic krill, Euphausia superba, was examined using samples collected from the Indian sector of the Southern Ocean. Immature and mature eugregarine gamont stages were found at all larval stages older than Calyptopis I stage. Eugregarine infection in 14.0% (N = 108) of the first feeding stage (Calyptopis I) suggested that krill larvae are at risk from being infected during physiological transition from non-feeding to feeding stages. Eugregarine prevalence and intensity of infection at the three calyptopis stages increased with stage/krill length. Statistical analysis showed that the intensity of C. pacifica infection also increased with host density. Thus, krill density is probably a key determinant of the intensity of infection. We found gamont stage eugregarines in the host hind-gut, blocking the passage of food. Eugregarine infections in larval krill may have a negative impact on digestion and absorption in the host digestive tract.  相似文献

Simian virus 40 (SV40)-transformed Syrian hamster kidney cells produce infectious SV40 virus particles after treatments which damage DNA, such as UV irradiation or mitomycin C treatment. We have found that the induction of SV40 by DNA-damaging agents is greatly stimulated when a typical tumor promoter, 12-O-tetradecanoylphorbol 13-acetate (TPA), is present in the medium. Phorbol, which has a molecular structure similar to TPA but does not have any tumor-promoting activity, showed no such stimulatory effect on SV40 induction. This apparent synergistic effect of DNA-damaging agents and tumor promoter (TPA) was more pronounced when a tumor initiator, benzo [a]pyrene or 2-acetamido-fluorene, was combined with TPA. The effect of TPA on UV-triggered SV40 induction was greatly influenced by the timing of TPA addition to the culture medium, which was most efficient when addition of TPA was 5 to 20 h before UV irradiation. The effect of TPA, however, was not observed in SV40 rescue from hamster cells by cell fusion with permissive monkey (C7) cells.  相似文献

1. Radiotherapy has attracted increasing interest in recent years. It is known that ionizing radiation induces oxygen radical injury, whereas oxidative stress by the radiation can cause cellular responses to defense cellular injury. In this study, the metabolism of antioxidants in response to ionizing radiation to the brain was studied in the brain using experimental rabbits.2. Ionizing radiation to the hemicerebrum caused an increase in the levels of glutathione (GSH) and the activity of a GSH synthesizing enzyme, -glutamylcysteine synthetase (-GCS), and Cu,Zn-superoxide dismutase (Cu,Zn-SOD). Ionizing radiation also induced DNA-damage estimated by the formation of 8-hydroxydeoxyguanosine. These changes were dependent on the radiation dose.3. Previous intrathecal-administration of buthionine sulfoximine (100 M), a specific inhibitor of -GCS, increased DNA damage by radiation in the radiated hemicerebrum. That of S-methyl GSH, on the other hand, resulted in a significant reduction of DNA damage by radiation.4. These results suggest that synthesis of GSH and Cu,Zn-SOD is responsive to ionizing radiation and this induction of antioxidants may play a role in reducing tissue damage in radiotherapy.  相似文献

Summary 1. We studied the effects of BQ-123, a selective ETA receptor antagonist, on125I-endothelin-1 (125I-ET-1) binding to cell surface receptors in surgically excised human meningiomas and on ET-1-induced DNA synthesis in cultured human meningioma cellsin vitro, using a quantitative receptor autoradiographic technique with radioluminography and3H-thymidine incorporation, respectively.2. All of the human meningiomas expressed high-affinity binding sites for125I-ET-1, regardless of differences in histological subtypes (K d=2.6±0.2 nM,B max=374±93 fmol/mg; mean ± SE;n=9).3. BQ-123 competed for125I-ET-1 binding to sections of meningiomas with IC50s of 3.2±0.9×10–7 M, and 10–4 M BQ-123 displaced 80% of the binding.4. ET-1 significantly stimulated DNA synthesis in cultured human meningioma cells, up to 170% of the basal level in the presence of 10–9 M ET-1. BQ-123 inhibited ET-1 (10–9 M)-induced DNA synthesis in meningioma cells, in a dose-dependent manner, and 10–5 M BQ-123 reduced it to 120% of the basal level.5. The number of meningioma cells determined after 4 days in culture was dose dependently increased in the presence of ET-1 (10–9 and 10–7 M). The growth rate of meningioma cells, incubated with 10–9 M ET-1, was reduced by 50% in the presence of 10–7 M BQ-123.6. Our data suggest that (a) human meningioma cells express a large number of ETA endothelin receptors, with a small proportion of non-ETA receptors linked to proliferation of the cells, and (b) ET receptor antagonists, including BQ-123, might prove to be effective treatment for patients with meningioma.  相似文献

Two monoclonal anti-fibronectin antibodies that inhibit fibronectin-mediated cell adhesion have been established and characterized. One antibody, FN12-8, inhibited attachment of rat kidney fibroblasts on the fibronectin-coated substrate in a concentration-dependent manner, attaining a maximal inhibition of greater than 85% at 850 micrograms/ml. Another antibody, FN30-8, caused about 70% inhibition at a concentration as low as 0.85 microgram/ml, although further increase of the antibody concentration did not significantly augment the inhibitory effect. Immunoblot analysis with defined proteolytic fragments revealed that both antibodies are directed to the cell-binding domain of fibronectin. The epitopes for these antibodies were further narrowed down using recombinant cell-binding fragments expressed in Escherichia coli. FN12-8 recognized the 11.5-kDa cell-binding fragment previously characterized by Pierschbacher et al. (1981, Cell 26, 259-267), suggesting that FN12-8 blocks the Arg-Gly-Asp (RGD) cell adhesion signal. FN30-8 could not bind this fragment but did recognize a longer cell-binding fragment containing additional greater than 111 amino acid residues upstream of the 11.5-kDa fragment. Since the RGD-dependent cell adhesion seems to require another signal located at a region 50-160 residues upstream of the 11.5-kDa fragment for full activity, FN30-8 may exert its inhibitory effect by blocking the latter signal.  相似文献

Photosynthetic oxygen production by phytoplankton and community respiration in the Indian sector of the Antarctic Ocean were estimated from changes in oxygen concentrations in light and dark bottles. Gross production varied between 0.1 and 5.1 µmol O2 l-1 day-1. In the same water, community respiration (the sum of oxygen consumption by heterotrophs and phytoplankton) was 0.4-3.6 µmol O2 l-1 day-1, which accounted for 47-343% of the gross production. Algal and heterotrophic respirations were distinguished using some assumptions. These estimates showed that heterotrophic respiration accounted for most of the community respiration (70-91% depending upon the assumptions), indicating that heterotrophic respiration plays an important role in the mineralization of phytoplankton production in the surveyed sea area. Gross production rate correlated with chlorophyll a concentration, showing that the photosynthetic production rate of oxygen depends on the abundance of phytoplankton. Moreover, there was a significant relationship between gross production and community respiration rates. These regression equations suggested that negative net production occurred under the usually low concentration of chlorophyll observed in the Indian sector of the Antarctic Ocean. Hence, the net exchange of carbon dioxide due to biological processes through the sea surface seemed to be not as large as expected in the Antarctic Ocean, although the number of data were limited at this stage.  相似文献
