1. We assessed the role of cyanobacterial–bacterial consortia (Gloeotrichia echinulata phycospheres) for net changes in inorganic carbon, primary production (PP) and secondary production in Lake Erken (Sweden). 2. At the time of sampling, large colonies of G. echinulata formed a massive bloom with abundances ranging from 102 colonies L?1 in the pelagic zone to 5000 colonies L?1 in shallow bays. These colonies and their surrounding phycospheres contributed between 17 and 92% of total PP, and phycosphere‐associated bacteria contributed between 8.5 and 82% of total bacterial secondary production. PP followed a diurnal cycle, whereas bacterial production showed no such pattern. Over a 24 h period, carbon dioxide measurements showed that the phycospheres were net autotrophic in the top layer of the water column, whereas they were net heterotrophic below 2 m depth. 3. Sequencing and phylogenetic analysis of 16S rRNA genes of attached bacteria revealed a diverse bacterial community that included populations affiliated with Proteobacteria, Bacteriodetes, Acidobacteria, Fusobacteria, Firmicutes, Verrucomicrobia, and other Cyanobacteria. 4. Compared with their planktonic counterparts, bacteria associated with cyanobacterial phycospheres had lower affinity for arginine, used as a model compound to assess uptake of organic compounds. 5. Extrapolation of our data to the water column of lake Erken suggests that microorganisms that were not associated with cyanobacteria dominated CO2 production at the ecosystem scale during our experiments, as CO2 fixation balanced CO2 production in the cyanobacterial phycospheres.  相似文献

Males of the colour polymorphic Australian painted dragon lizard Ctenophorus pictus occur in red or yellow head colouration. In a previous experiment, we showed that red is associated with a higher probability of winning staged contests for resources (females or space) and that manipulation of male colouration by painting males in the opposing morph changed the dynamics of staged interactions by prolonging them 30‐fold. Thus, colour is linked to behavioural differences between males and is involved in information transfer between competing males. This inherent red dominance could result in yellow males being marginalized to poorer quality territories in terms of access to females, food, perch sites or shade. With an experiment in the wild, we test to what extent this prediction is upheld, and how colour manipulation affects morph‐specific success in territory acquisition when male body size, territory quality and emergence time from hibernation are controlled through manipulation or randomization. There was no significant effect of colour category per se, although on average red males remained closer to the release sites (our proxy for territory acquisition ability) than yellow males and artificially altered morphs moved the furthest away. There was a significant interaction effect between colour category and experimental release position, which may be linked to differences in how exposed (or not) these positions were and morph‐specific ability to cope with such exposure (e.g. ‘boldness’). Our data show that territory acquisition success is not merely a function of competitive ability but a composite outcome of a suite of factors, including signal perception.  相似文献

We studied the interactive effects of elevated concentrations of CO2 and O3 on radial growth and wood properties of four trembling aspen (Populus tremuloides Michx.) clones and paper birch (Betula papyrifera Marsh.) saplings. The material for the study was collected from the Aspen FACE (free‐air CO2 enrichment) experiment in Rhinelander (WI, USA). Trees had been exposed to four treatments [control, elevated CO2 (560 ppm), elevated O3 (1.5 times ambient) and combined CO2 + O3] during growing seasons 1998–2008. Most treatment responses were observed in the early phase of experiment. Our results show that the CO2‐ and O3‐exposed aspen trees displayed a differential balance between efficiency and safety of water transport. Under elevated CO2, radial growth was enhanced and the trees had fewer but hydraulically more efficient larger diameter vessels. In contrast, elevated O3 decreased radial growth and the diameters of vessels and fibres. Clone‐specific decrease in wood density and cell wall thickness was observed under elevated CO2. In birch, the treatments had no major impacts on wood anatomy or wood density. Our study indicates that short‐term impact studies conducted with young seedlings may not give a realistic view of long‐term ecosystem responses.  相似文献

Abstract. The activity of the green alga Scenedesmus obliquus was studied in simplified nutrient solutions (20 mol m−3 NaNO3, 20 mol m−3 NH4C1, 20 mol m−3 NH4NO3, and 20 mol m−3 NaCl, respectively) at 25 °C. The experiments were performed under welldefined incident photon density fluxes ranging from 10 to 200 μmol m2 s−1, Light-dependent changes in pH and alkalinity (A) were followed by means of a potentiometric method using a glass electrode. In the experiments, carbon dioxide with known partial pressure was bubbled through the algal suspension, and during dark periods ul intervals of 1 h, the solution was allowed to equilibrate with the gas phase. This technique was applied to calculate equilibrium values of pH and alkalinity at regular intervals during a 12-h period. Results obtained in NaNO3, solution show a linear increase in A with time, at each level of illumination studied. After an initial drop, A also increases in NH4NO3, solution in a similar way to that in NaNO3 solution. The change in A with time was also found to increase linearly with the photon density flux studied and no saturation level could be defined. In experiments in NaCl solution, no changes in A were registered while measurements in NH4Cl solution showed a decrease in A with time.  相似文献

1. It is often assumed that lakes highly influenced by terrestrial organic matter (TOM) have low zooplankton food quality because of elemental and/or biochemical deficiencies of the major particulate organic carbon pools. We used the biochemical [polyunsaturated fatty acids, especially eicosapentaenoic acid (EPA) – 20:5ω3] and elemental (C : P ratio) composition of particulate matter (PM) as qualitative measures of potential zooplankton food in two categories of lakes of similar primary productivity, but with contrasting TOM influence (clear water versus humic lakes). 2. C : P ratios (atomic ratio) in PM were similar between lake categories and were above 400. The concentration (μg L−1) and relative content (μg mg C−1) of EPA, as well as the particulate organic carbon concentration, were higher in the humic lakes than in the clear‐water lakes. 3. Our results show high fatty acid quality of PM in the humic lakes. The differences in the biochemical quality of the potential zooplankton food between lake categories can be attributed to the differences in their phytoplankton communities. 4. High biochemical quality of the food can result in high efficiency of energy transfer in the food chain and stimulate production at higher trophic levels, assuming that zooplankton are able to ingest and digest the resource available.  相似文献

In embryos of the white mutant axolotl, prospective pigment cells are unable to migrate from the neural crest (NC) due to a deficiency in the subepidermal extracellular matrix (ECM). This raises the question of the molecular nature of this functional defect. Some PGs can inhibit cell migration on ECM molecules in vitro, and an excess of this class of molecules in the migratory pathways of neural crest cells might cause the restricted migration of prospective pigment cells seen in the white mutant embryo. In the present study, we use several monoclonal antibodies against epitopes on keratan sulphate (KS) and chondroitin sulphate (CS) and LM immunofluorescence to examine the distribution of these glycosaminoglycans at initial (stage 30) and advanced (stage 35) stages of neural crest cell migration. Most KS epitopes are more widely distributed in the white mutant than in the wild type embryo, whereas CS epitopes show very similar distributions in mutant and wild type embryos. This is confirmed quantitatively by immunoblotting: certain KS epitopes are more abundant in the white mutant. TEM immunogold staining reveals that KS as well as CS are present both in the basal lamina and in the interstitial ECM in both types of embryos. It remains to be investigated whether the abundance of certain KS epitopes in the white mutant embryo might contribute to the deficiency in supporting pigment cell migration shown by its ECM.  相似文献

Fractions of acetone extracts from heartwood, sapwood and wood from the reaction zone in roots of Norway spruce, attacked by Fomes annosus, were tested concerning their effect on the activity of cellulase, polygalacturonase, aryl-β-glucosidase and laccase, obtained from culture filtrates of the same fungus. A specific inhibition of polygalacturonase was recorded with the resinous fractions. The hydrolysing enzymes were more inhibited by the lignans of the reaction zone in the presence of fungal laccase than in the absence of laccase. The pH dependence of the inhibiting effect of phenols varied among the enzymes. The four enzymes were inhibited by partly different lignan fractions, and synergistic effects between minor compounds were indicated. The phenolic fractions inhibited weakly the biosynthesis of the enzymes. It was obvious that results from enzyme inhibition studies in vitro do not reflect the complicated mutual interrelationships between production and activity of the extracellular fungal enzymes and the chemical constituents of the host in vivo.  相似文献
