Terrestrial ecosystems in northern high latitudes exchange large amounts of methane (CH4) with the atmosphere. Climate warming could have a great impact on CH4 exchange, in particular in regions where degradation of permafrost is induced. In order to improve the understanding of the present and future methane dynamics in permafrost regions, we studied CH4 fluxes of typical landscape structures in a small catchment in the forest tundra ecotone in northern Siberia. Gas fluxes were measured using a closed‐chamber technique from August to November 2003 and from August 2006 to July 2007 on tree‐covered mineral soils with and without permafrost, on a frozen bog plateau, and on a thermokarst pond. For areal integration of the CH4 fluxes, we combined field observations and classification of functional landscape structures based on a high‐resolution Quickbird satellite image. All mineral soils were net sinks of atmospheric CH4. The magnitude of annual CH4 uptake was higher for soils without permafrost (1.19 kg CH4 ha−1 yr−1) than for soils with permafrost (0.37 kg CH4 ha−1 yr−1). In well‐drained soils, significant CH4 uptake occurred even after the onset of ground frost. Bog plateaux, which stored large amounts of frozen organic carbon, were also a net sink of atmospheric CH4 (0.38 kg CH4 ha−1 yr−1). Thermokarst ponds, which developed from permafrost collapse in bog plateaux, were hot spots of CH4 emission (approximately 200 kg CH4 ha−1 yr−1). Despite the low area coverage of thermokarst ponds (only 2.1% of the total catchment area), emissions from these sites resulted in a mean catchment CH4 emission of 3.8 kg CH4 ha−1 yr−1. Export of dissolved CH4 with stream water was insignificant. The results suggest that mineral soils and bog plateaux in this region will respond differently to increasing temperatures and associated permafrost degradation. Net uptake of atmospheric CH4 in mineral soils is expected to gradually increase with increasing active layer depth and soil drainage. Changes in bog plateaux will probably be much more rapid and drastic. Permafrost collapse in frozen bog plateaux would result in high CH4 emissions that act as positive feedback to climate warming.  相似文献

Polypterus faraou sp. nov. (Cladistia, Polypteridae) from the Late Miocene of Toros-Menalla (western Djourab, Chad) is described on the basis of a subcomplete articulated skeleton preserved in three dimensions. This is the first time such a complete fossil polypterid skeleton has been described. It is the only verifiable fossil record for the genus Polypterus . P. faraou closely resembles P. bichir and P. endlicheri , extant fish found in the Chad–Chari system. Intrarelationships among the polypterids are not yet resolved: however, P. faraou shows a primitive shape of the body and a primitive shape of the opening of the lateral line on the scales, similar to that of three living species ( P. bichir , P. endlicheri and P. ansorgii ).  © 2006 The Linnean Society of London, Zoological Journal of the Linnean Society , 2006, 146 , 227–237.  相似文献

Fusarium oxysporum is a soil-borne pathogen that infects plants through the roots and uses the vascular system for host ingress. Specialized for this route of infection, F. oxysporum is able to adapt to the scarce nutrient environment in the xylem vessels. Here we report the cloning of the F. oxysporum global nitrogen regulator, Fnr1 , and show that it is one of the determinants for fungal fitness during in planta growth. The Fnr1 gene has a single conserved GATA-type zinc finger domain and is 96% and 48% identical to AREA-GF from Gibberella fujikuroi , and NIT2 from Neurospora crassa , respectively. Fnr1 cDNA, expressed under a constitutive promoter, was able to complement functionally an N. crassa nit-2 RIP mutant, restoring the ability of the mutant to utilize nitrate. Fnr1 disruption mutants showed high tolerance to chlorate and reduced ability to utilize several secondary nitrogen sources such as amino acids, hypoxanthine and uric acid, whereas growth on favourable nitrogen sources was not affected. Fnr1 disruption also abolished in vitro expression of nutrition genes , normally induced during the early phase of infection. In an infection assay on tomato seedlings, infection rate of disruption mutants was significantly delayed in comparison with the parental strain. Our results indicate that FNR1 mediates adaptation to nitrogen-poor conditions in planta through the regulation of secondary nitrogen acquisition, and as such acts as a determinant for fungal fitness during infection.  相似文献

Parasitic infections may change the equilibrium between the costs and benefits of an animal for maintaining its status in a social group. Consequently, parasites may influence the social status of an animal in a group. The present study investigated whether acute infection with Isospora spp. has any effect on the social relationships (e.g. dominance hierarchy) of male house sparrows and how the infection influences their behaviour, immune status, and body condition. Furthermore, the study allowed us to examine how important the ‘badge of dominance’ is with respect to maintaining social status even when the actual condition is changing as a result of infection. The results obtained showed that an acute infection leads to changes in the dominance hierarchy of a social group and that body mass losses of birds depend on the achieved hierarchy status. A positive correlation between the badge size and male aggressiveness was only found during acute infection. In addition, we also found a relationship between cell‐mediated immune response and male aggressiveness during acute infection. This suggests that male badge size is not sufficient to maintain a given dominance position. On the other hand, badge size, a signal developed during the moult, appears to remain an informative and ‘honest’ signal several months later, reflecting the energy reserves of a bird faced with a demanding stressful situation such as acute infection. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 718–726.  相似文献

ABSTRACT

Here we present an acoustic approach for reliable sexing in four whistling duck species from the genus Dendrocygna and compare it with molecular and cloacal inspection techniques. In the four examined species, the White-faced Whistling Duck D. viduata, Fulvous Whistling Duck D. bicolor, Cuban Whistling Duck D. arborea and Red-billed Whistling Duck D. autumnalis, sexes are indistinguishable by appearance. However all the four species show strong sexual differences in the structure of their species-specific loud whistles. For 59 examined birds, an acoustic-based sexing showed 100% accordance to the DNA PCR analysis, while the cloacal inspection showed only 89.8% accuracy. The results demonstrate that acoustic sexing represents a feasible alternative to the two traditional methods as a noninvasive tool for the distant sexing of the four whistling duck species both in captivity and in the wild.  相似文献

ABSTRACT. A new method of fractionation and purification of different life cycle stages of microsporidia Nosema grylli , parasitizing the fat body of cricket Gryllus bimaculatus , by centrifugation in Percoll density gradient is elaborated. The whole procedure can be summarized as: 1) infected fat body preparation, 2) homogenization in buffer and filtration through cotton wad and filter paper, 3) first centrifuging, resulting in the separation of the pellet into three layers containing different life cycle stages, 4) second centrifuging of the chosen layer in Percoll density gradient, 5) washing out the Percoll from the fraction under study. After centrifugation in Percoll density gradient, meronts and early sporonts form a band in the area corresponding to density 1.016 g/ml. Mature spores form the pellet at the bottom of centrifuge tube, while immature spores are distributed throughout the layer of 1.016 g/ml up to the bottom of the centrifuge tube, according to their buoyant densities. The offered technique is simple, it takes about one hour and may become a routine procedure for biochemical studies on microsporidia.  相似文献

The evaluation of panel performance was made by three methods: average of correct responses (A), comparison of distances of individual standardized judgments to the average standardized responses (D) and a principal components analysis (PCA). Thirty assessors identified water and basic tastes and discriminated different sweet stimuli in neutral or acidified vehicles using R‐index rating and ranking tests. By A and D methods 22 assessors were qualified as proficient. Composition of both panels was identical except for one judge. The output from PCA provided a graphical representation of the performance of the assessors and retained different subsets of 24–26 panelists for different proposals as discrimination of sweetness in acidified beverages, recognition of bitterness, sourness and discrimination of slight sweetness or evaluation of saltiness.  相似文献

1. Two field experiments were carried out to examine the role of patch size, host density, and complexity of the surrounding habitat, on the foraging behaviour of the parasitoid wasp Cotesia glomerata in the field. 2. First, released parasitoids were recaptured on patches of one or four Brassica nigra plants, each containing 10 hosts that were placed in a mown grassland area. Recaptures of females were higher than males, and males and females aggregated at patches with four plants. 3. In experiment 2, plants containing 0, 5 or 10 hosts were placed in unmown grassland plots that differed in plant species composition, on bare soil, and on mown grassland. Very low numbers of parasitoids were recaptured in the vegetated plots, while high numbers of parasitoids were recaptured on plants placed on bare soil or in mown grassland. Recaptures were higher on plants on bare soil than on mown grassland, and highest on plants containing 10 hosts. The host density effect was significantly more apparent in mown grassland than on bare soil. 4. Cotesia glomerata responds in an aggregative way to host density in the field. However, host location success is determined mostly by habitat characteristics, and stronger host or host‐plant cues are required when habitat complexity increases.  相似文献
