This paper summarizes some structural characteristics of Potato virus X (PVX), the flexuous filamentous plant potexvirus. A model of PVX coat protein (CP) tertiary structure in the virion proposed on the basis of tritium planigraphy combined with predictions of the protein tertiary structure is described. A possible role of glycosylation and phosphorylation in the CP structure and function is discussed. Two forms of PVX virion disassembly are discussed: (i) the virion co-translational disassembly after PVX CP in situ phosphorylation and (ii) disassembly of PVX triggered by different factors after linear destabilization of the virion by binding of the PVX-coded movement protein (TGBp1) to one end of the polar CP-helix. Special emphasis was placed on a translational activation of encapsidated PVX RNA and rapid disassembly of TGBp1-PVX complexes into free RNA and CP. The results of experiments on the PVX CP repolymerization and PVX reconstitution are considered. In particular, the products assembled from PVX RNA, CP and TGBp1 were examined. Single-tailed particles were found with a helical, head-like structure consisting of helically arranged CP subunits located at the 5'-tail of RNA; the TGBp1 was bound to the end of the head. Translatable 'RNA-CP-TGBp1' complexes may represent the transport form of the PVX infection.  相似文献

Twelve polymorphic microsatellite loci were isolated from polyploid alpine wood‐sorrel, Oxalis alpina (Oxalidaceae), and optimized for future studies of its breeding system. The loci were screened for variability among 72 individuals from Pinos Altos, New Mexico. The primers amplified loci with allele number ranging from two to 17 per locus and with estimates of Nei's genetic diversity varying from 0.10 to 0.99. These primers provide an opportunity to use polymorphic DNA markers to study the causes of breeding system variability in this species.  相似文献

1. Thirty-six species of macrophytes (fourteen flowering plants, two quillworts, sixteen mosses and liverworts, and two algae) were collected in an extensive survey of 116 high mountain lakes in the eastern Pyrenees. Seventy per cent of the lakes showed macrophyte development. 2. The isoetids (Isoetes lacusiris, Isoetes setacea and Subularia aquatica) were the dominant growth form, although the natopotamid Sparganium angusifolium was the most widespread species. Potamids (Potamogeton spp., Ranunculus spp.), the alga Nitella gr. opaca and some mosses (Warnstorfia exannulata, Sphagnum denticulatum) were often present. 3. A multivariate ordination analysis (RDA-redundancy analysis) revealed that water chemistry, altitude and vegetation cover of the catchment, and nutrient availability are major environmental factors associated with macrophyte distribution along the eastern Pyrenean lakes. Isoetids prevailed in softwater oligotrophic lakes, potamids in relatively hardwater oligotrophic lakes, and Potamogeton natans and Callitriche palustris in small and eutrophic water bodies affected by the presence of cattle.  相似文献

Fifteen polymorphic microsatellite DNA loci were isolated from barn swallow (Hirundo rustica) and optimized for future studies of radiation‐induced mutations in populations from Ukraine. The loci were screened for variability among 25 individuals from two populations. The primers amplified loci with relatively high numbers of alleles ranging from five to 32 alleles per locus and polymorphic information content from 0.481 to 0.951. Observed heterozygosity varied from 0.458 to 0.960. None of the loci showed deviations from Hardy–Weinberg equilibrium in either population.  相似文献

Induction of melanophores was examined by the sandwich method of explantation with embryonic tissues of Xenopus laevis +/+ and the white mutant, aP/aP. Interspecific combinations of tissues of Triturus taeniatus and Xenopus borealis were also used. The ectoderm used as the reacting system was taken from embriyos at various stages and combined with various tissues known to be melanogenic inductors. The following results were obtained: 1) The sources of melanophore induction in both +/+ and ap/ap studied by sandwich explantation were the same in both retinal pigmented epithelium and dermal melanophores: 2) Melanophores were induced in epidermal material from embryos at stages from the early gastrula to the late tail bud stage: 3) The presence of melanoblasts together with other ectomesenchymal cells in the neural crest is not sine qua non for their determination and differentiation: 4) On isolation of reacting material from the late gastrula, melanophores appeared in all cases. This shows that two hours contact between inductor tissues and the ectoderm is necessary and sufficient for melanophore induction: 5) Melanophore induction is not species-specific, but occurred in Xenopus ectoderm under the action of endomesoderm of Tr. taeniatus or X. borealis , and vice versa. The shapes and structures of melanophores induced were typical for the species from which the ectoderm was taken: 6) Melanogenic activity in the late gastrula stage has a gradient of distribution with a maximum in the prechordal plate: 7) In the mutant only the primary source of melanogenic inductors, the prechordal plate (PrP1), was active in stages both before and after its invagination: 8) Despite the fact that skin melanophores and retinal melanocytes have different genesis in development, all the present data suggest the identity of the mechanisms of melanin synthesizing machinery in the two.  相似文献

The mechanisms of transdifferentiation of iris epithelial cells of Rana temporaria (Anura) in culture depending on influences from different sources were studied. In terminally differentiated iris cells, the process of transdifferentiation is initiated by dedifferentiation. Melanosomes are shed from iris cells due to cell surface activity. After depigmentation, iris epithelial cells become capable of proliferating and competent to react to the influences of various exogenous factors. Under the influence of retinal factors secreted by lentectomized tadpole eyes, both dorsal and ventral irises are converted to neural retina. Under the influence of factors from eye vesicles, the irises are converted to neural retina as well. Similar results were obtained in transfilter experiments, in which a 3-day period of transfilter interaction between the irises and eye vesicles ensured depigmentation of the iris followed by transdifferentiation into complete NR with visual receptor. Lentoid formation occurred under the influence of adult frog lens epithelium. Immunofluorescent analysis confirmed the lens nature of the lentoids. In control experiments under the conditions of the tadpole eye orbit, in which programming influences were absent, iris epithelial cells remained unaffected.
The problem of true cell-reprogramming to new differentiation in contrast to expression of inherent properties of the iris epithelial cells is discussed.  相似文献
