New chromatographic material based on tryptophil-threonil-tirosine was prepared. This sorbent effectively binds human, sheep, goat and cow immunoglobulins G. New sorbent shows high selectivity for removing immunoglobulins from blood plasma. Effective sorption capacity is 15-25 mg of immunoglobulin G per ml of matrix. Optimal method of covalent attachment ligand to polysaccharide matrix allows achieving high stability of the sorbents in terms of use and storage. This sorbent can be used in medicine and biotechnology.  相似文献

A polysaccharide was isolated from the opportunistic human pathogen Providencia alcalifaciens O45:H26 by extraction with aqueous phenol and studied by sugar and methylation analyses along with 1H and 13C NMR spectroscopy, including two-dimensional ROESY and H-detected 1H,13C HSQC experiments. The polysaccharide contains N-acetylglu-cosamine and N-acetylmuramic acid (D-GlcpNAc3Rlac) amidated with L-alanine and has the following structure:

The polysaccharide possesses a remarkable structural similarity to the bacterial cell wall peptidoglycan. It is not unique to the strain studied but is common to strains of at least four P. alcalifaciens O-serogroups (O3, O24, O38, and O45). No evidence was obtained that the polysaccharide is associated with the LPS, and hence it might represent a bacterial capsule component.

Influenza A virus matrix M1 protein is membrane associated and plays a crucial role in virus assembly and budding. The N-terminal two thirds of M1 protein was resolved by X-ray crystallography. The overall 3D structure as well as arrangement of the molecule in relation to the viral membrane remains obscure. Now a proteolytic digestion of virions with bromelain was used as an instrument for the in situ assessment of the M1 protein structure. The lipid bilayer around the subviral particles lacking glycoprotein spikes was partially disrupted as was shown by transmission electron microscopy. A phenomenon of M1 protein fragmentation inside the subviral particles was revealed by SDS-PAGE analysis followed by in-gel trypsin hydrolysis and MALDI-TOF mass spectrometry analysis of the additional bands. Putative bromelain-digestion sites appeared to be located at the surface of the M1 protein globule and could be used as landmarks for 3D molecular modeling.  相似文献

The authors present the results of study of the virulence of shigellae isolated from carriers and patients suffering from acute dysentery, on a continuous culture of E1 and Hep-2 cells. The virulence of shigellae isolated from carriers displayed no significant difference from the virulence of shigellae isolated from patients with a mild and moderately severe forms of dysentery. In the patient's organism shigellae were capable of retaining the initial virulence for a long time, despite the treatment and the influence of the macroorganism's protective factors. The authors believe that this was connected with the capacity of Shigellae to parasitic life in the cells of human intestinal epithelium, and, apparently, played a definite role in the formation of carrier state.  相似文献

The influence of adenosine on hemodynamic changes induced by electrostimulation of reflexogenic zone of aorta was studied in this article. It was shown that 3-hour electrostimulation increased the total peripheral resistance and decreased cardiac output and aortic pressure. Intravenous infusion of adenosine during electrostimulation led to the normalization of main indices of hemodynamics.  相似文献

The correlation between the kinetic stability of molecules against temperature and variations in their geometric structure under optical excitation is investigated by the example of different organic pheromone molecules sensitive to temperature or ultraviolet radiation using the density functional theory. The kinetic stability is determined by the previously developed method based on the calculation of the probability of extension of any structural bond by a value exceeding the limit value Lмах corresponding to the breaking of the bond under temperature excitation. The kinetic stability calculation only requires the eigenfrequencies and vibrational mode vectors in the molecule ground state to be calculated, without determining the transition states. The weakest bonds in molecules determined by the kinetic stability method are compared with the bond length variations in molecules in the excited state upon absorption of light by a molecule. Good agreement between the results obtained is demonstrated and the difference between them is discussed. The universality of formulations within both approaches used to estimate the stability of different pheromone molecules containing strained cycles and conjugated, double, and single bonds allows these approaches to be applied for studying other molecules.

1. Resource density can regulate the area that animals use. At low resource density, there is a conflict in terms of balance between costs of foraging and benefits acquired. The foraging of the higher termite Nasutitermes aff. coxipoensis consists of searching throughout trails and a building galleries phase. 2. In this study, a manipulative field experiment was used to test the hypothesis that colonies of N. aff. coxipoensis forage towards a more profitable balance between the establishment of trails and gallery construction at low resource density. 3. The experiment was conducted in north‐eastern Brazil. Seven experimental plots were established with a continuous increase in resource density (sugarcane baits). Entire colonies of N. aff. coxipoensis were transplanted from their original sites to the experimental plot, totalling 35 nests. The number, branches and total length of trails and galleries were quantified. 4. The results show that N. aff. coxipoensis optimises its foraging output, intensifying the establishment of trails at the cost of gallery construction when resource density is low. The number of trails, the number of trail branches and the total length of trails decreased with increasing resource density. Interestingly, at low resource density, the search effort was concentrated on forming longer and a greater number of trails, a small proportion of which were converted into galleries. The opposite relationship was observed at high resource density. 5. These results suggest an optimisation of search efforts during foraging depending on resource density, a mechanism that may help researchers to understand the use of space by higher termite species.  相似文献

Females of certain aphidophagous ladybirds in the absence of natural protein food (aphids) enter reproductive diapause. Reactivation of diapausing beetles is possible only after consuming the food which is suitable for oogenesis of females and development of larvae. The influence of diet and photoperiod on the dynamics of weight and on the rate of reproductive maturation of reactivating females of Harmonia axyridis was studied under laboratory conditions. The experiments were conducted at combinations of two day lengths (12 and 18 h) with 6 trophic regimes with the following mean numbers of daily consumed aphids: 0 (aphids were absent over the whole experiment), 0.1 (1 aphid was provided every 10 days), 0.5 (1 aphid every second day), 1 (1 aphid every day), 10 (10 aphids every day), and 50 (about 50 aphids every day). Judging from the state of ovarian development estimated by dissection 20 days after the beginning of the experiment, the threshold of the trophic termination of reproductive diapause under the long day conditions lies between the regimes of 0.1 and 0.5 aphids per day, which is approximately equal to the earlier investigated threshold of the trophic induction of diapause. Short day slowed reactivation and has increased the threshold of the trophic response up to 1-10 aphids per day. Based on these data, we conclude that under natural conditions females which entered reproductive diapause because of the absence of aphids are capable of reactivation in the presence of even a minimal amount of natural protein food. Moreover, in spring and summer (when the probability of the increase in aphid abundance is relatively high) reactivation starts at lower prey population density than in autumn, when the appearance of the stable aphid colony (which is necessary to complete the development of the emerged larvae) is less probable.  相似文献
