The mutagenic effect of chrysotile asbestos fibers and zeolite and latex particles on human lymphocytes in whole blood has been studied. It was concluded that their mutagenic activities were mediated by oxygen radicals because they were inhibited by antioxidant enzymes (SOD and catalase) and oxygen radical scavengers (rutin, ascorbic acid, and bemitil). It was proposed that oxygen radicals were released by phagocytes activated upon exposure to mineral dusts and fibers. The study of lucigenin- and luminol-amplified chemiluminescence of peritoneal macrophages stimulated by chrysotile fibers and zeolite and latex particles has shown that their mutagenic action is probably mediated by different oxygen species, namely, by the iron-oxygen complexes (perferryl ions) plus hydrogen peroxide, hydrogen peroxide, and superoxide ion, respectively. From the oxygen radical scavengers studied, rutin was the most effective inhibitor of the mutagenic effect of mineral fibers and dusts.  相似文献

Adult male rats were maintained on 0, 5, 10, 15, and 20% casein diets to produce a series of animals having serum alpha 2u-globulin levels varying linearly from a normal of 31 micrograms/ml to a minimum of 13 micrograms/ml. In this way, it was possible to titrate endogenously the renal reabsorption and urinary excretion of this low molecular weight protein. The average maximal reabsorption rate (Tm) was established to be 9.7 micrograms/min and was reached at a renal filtered load (F alpha 2u) of 13.6 micrograms/min. These data were expressed in terms of a Tm:F alpha 2u ratio of 0.71. Below this value, the reabsorption declined from 70% to 50% of the F alpha 2u. Above 0.71, where F alpha 2u is less than the Tm, the reabsorption increased to 80-90%. It was observed that the fractional renal uptake of the alpha 2u-globulin varied linearly with the filtered load.  相似文献

The number of osteoporotic females in Japan with vertebral bone mineral density measured by dual energy x-ray absorptiometry, defined as less than -3 SD of the peak bone mass, is approximately 10,000,000, corresponding to 8% of the whole population of Japan. While this value approximately corresponds to the prevalence of low bone mineral density in the United States, the incidence of hip fracture appears to be much less in Japan than in the United States, 50,000 per 125,000,000 per year compared with 250,000 for a population twice as large. This seems to be paradoxical because of the lower bone mineral density and lower calcium intake in Japan, with 400-500 mg/day mainly as soybean products, small fish with bones, and vegetables. The difference in hip fracture incidence, however, may not actually be as wide as it seems when the larger number of bedridden elderly subjects in Japan is taken into consideration. In these bedridden subjects with severe immobilization osteoporosis, hip fracture is only prevented by the fact that they are not ambulatory. Life-style difference may also offer an explanation. Sitting on a tatami mattress on completely flexed knees with frequent standing up, along with other household work, in a narrow home space may ensure a marked development of hip musculature and also provide skill in balancing oneself to prevent fails. The difference in fracture incidence should be analyzed from various angles to obtain a firm ground for the future prevention of hip fracture due to osteoporosis. Although osteoporosis universally affects all races and nationalities, conspicuous differences may be encountered in the severity of its manifestations and complications.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Activity of redox-enzymes of AA system and of catalase was measured in two near-isogenic tomato lines, respectively resistant and susceptible to Tobacco Mosaic Virus infection. AFR reductase, DHA reductase and catalase showed quite similar activities in both lines, whereas AA peroxidase activity in resistant plants was 75% higher than in susceptible ones, with Km values about 4-fold lower. These data suggest that hydrogen peroxide scavenging operated by AA peroxidase could play an important role in the development of biological defence mechanisms against pathogens.  相似文献

Chromatin structure has been studied with a method of optico-structural machine analysis for interphase nuclei of peripheral blood lymphocytes in healthy donors, a girl with the Shereshevski?-Turner syndrome (45, XO) and her parents, and for peripheral blood lymphocytes of a cow, Bos taurus L., in the last trimester on the background of preeclamptic syndrome and with normal cow pregnancy of compared terms. A significant change was revealed both in human heteroploidy and in developing preeclamptic syndrome in animals as concerns such indexes as the nucleus area, integral optical density, the amount and area of condensed, decondensed chromatin. The profile of histograms, according to the given parameters, is distinctly changing. The analysis based on constructing a two-measured field in the given coordinate system showed changes in the lymphatic population structure with the increase in the rate of "heavy", enriched DNA cells.  相似文献

A pUC19-derived plasmid was constructed that coded for a hybrid cellulase with the Thermomonospora fusca E2 cellulose-binding domain at its C terminus joined to the Prevotella ruminicola 40.5-kDa carboxymethyl cellulase (CMCase). The hybrid enzyme was purified and characterized enzymatically. It bound tightly to cellulose, and its specific activities on carboxymethyl cellulose, amorphous cellulose, and ball-milled cellulose were 1.5, 10, and 8 times that of the 40.5-kDa CMCase, respectively. Furthermore, the modified enzyme gave synergism with an exocellulase in the degradation of filter paper, while the 40.5-kDa CMCase did not.  相似文献

Glycosylation of trimethylsilyl derivatives of 5-benzyloxymethyl- and 5-hydroxymethyluracil with 3-azido-2,3-dideoxy-5-O-benzoyl-D-ribofuranosyl chloride (prepared from ethyl 3-azido-2,3-dideoxy-5-O-benzoyl-D-ribofuranoside) and subsequent deacylation gave in both cases a mixture of anomeric 3'-azido-2',3'-dideoxy-5-benzyloxymethyl-or 5-hydroxymethyluridines. The anomers were separated by preparative TLC and their structures were studied by UV, IR and 1H-NMR spectroscopy. It is shown that 1-(3-azido-2,3-dideoxy-alpha-D-ribofuranosyl)-5-benzyloxymethyluracil has cytotoxic activity in vitro: in 10(-5)-10(-4) M concentrations it inhibits the thymidine incorporation into DNA of CaOv cells on 78.6-95.2%.  相似文献
