Interspecific and intraspecific comparisons of the period locus in the Drosophila willistoni sibling species

The period (per) locus has received much attention in molecular evolution studies because it is one of the best studied "behavioral genes" and because it offers insight into the evolution of repetitive sequences. We studied most of the coding region of per in Drosophila willistoni and confirmed previously observed patterns of conservation and divergence among distantly related species. Five regions are so highly diverged that they cannot be aligned, whereas a region encompassing the PAS domain is very conserved. Structural and nucleotide polymorphism patterns in the willistoni group are not the same as those observed in previously studied species. We sequenced the region homologous to the highly polymorphic threonine-glycine repeat of D. melanogaster in multiple strains of D. willistoni, as well as in other members of willistoni group, and found an unusual amount of conservation in this region. However, the next nonconserved region downstream in the sequence is quite variable and polymorphic for the number of repeated glycines. The glycine codon usage is significantly different in this glycine repeat as compared to other parts of the gene. We were able to plot the directionality of change in the glycine repeat region onto a phylogeny and find that the addition of glycines is the general trend with the diversification of the willistoni group.      

Phosphotriesters in rat liver deoxyribonucleic acid after the administration of the carcinogen NN-dimethylnitrosamine in vivo.

After treatment with NN-di[14C]methylnitrosamine, samples of DNA were isolated from rat livers by a conventional phenol procedure and examined for the presence of phosphotriesters. A method of capable of detecting relatively small amounts of 14C-labelled phosphotriesters was developed and used to establish that these products account for 10-12% of the total methylation pattern found after treatment with this agent in vitro. The significance of the presence of phosphotriesters in DNA is discussed.     

Plate Hemolysin Test for the Rapid Screening of Toxoplasma Antibodies

A plate hemolysin test was developed to screen serum specimens for the presence of toxoplasma antibodies. When we tested 130 sera by both this test and the standard toxoplasma dye test, we found the plate hemolysin test to be a rapid, sensitive, and economical method for detecting toxoplasma antibodies. In all but one instance it paralleled the dye test. A comparison of the results of testing six sera by the hemolysin, hemagglutination, and dye-test techniques suggested that the hemolytic antibodies were more closely related to hemagglutinating antibodies than to dye-test antibodies. We could not store sheep erythrocytes sensitized with toxoplasma lysate for more than 3 days without altering the sensitivity of the test. Concanavalin A proved to be an effective coupling agent for binding toxoplasma antigens to red-cell membranes, a quality attributed to its affinity for specific polysaccharide-combining sites.     

Regulation of Dopamine Release from Interplexiform Cell Processes in the Outer Plexiform Layer of the Carp Retina

The gamma-aminobutyric acid (GABA) antagonists bicuculline and picrotoxin stimulate a four- to fivefold increase in endogenous dopamine release from isolated intact carp retina. The release evoked by these agents is Ca2+ dependent, a finding suggesting a vesicular release. Using light microscopic autoradiography, we have localized the sites of dopamine release to the dopaminergic interplexiform cell processes of the outer plexiform layer, which synapse onto horizontal cells. Our findings support previous suggestions that the dopaminergic interplexiform cells receive GABAergic inhibitory input and that the effects of GABA antagonists on horizontal cells are mediated by dopamine release from the interplexiform cells.     

Effect of Varying the Ionic Concentration of a Microdialysis Perfusate on Basal Striatal Dopamine Levels in Awake Rats

In vivo microdialysis was used to study the effects of Ca2+, Mg2+, and K+ ion concentrations on basal extracellular (EC) levels of striatal DA and metabolites in awake rats on the second day (48 h) after implantation of a microdialysis probe. Basal EC striatal dopamine (DA) levels were markedly (90%) and reversibly reduced by removal and subsequent replacement of Ca2+ ions from the microdialysis perfusate. This implies that the EC DA in this preparation is primarily of synaptic origin. The addition and subsequent removal of 1.7 mM MgCl2 to the Mg2(+)-free perfusate produced a reversible decrease (20%) in basal EC DA levels. This decrease may reflect a competitive interaction between Ca2+ and Mg2+ in the process of vesicular release. Basal EC DA levels were also reduced (27%) by decreasing the K+ concentration of the perfusate from 4 mM to 3 mM. However, after restoring the K+ concentration to 4 mM, EC DA levels were slow to return to pretreatment levels. Basal EC 3,4-dihydroxyphenylacetic acid and homovanillic acid levels exhibited a parallel but diminished response to each manipulation of the ionic concentration of the perfusate. This study demonstrates that small variations in the concentrations of Ca2+, Mg2+, and K+ in the perfusate employed in microdialysis preparations will affect basal EC striatal DA and metabolite levels.
