The mechanical stimuli resulting from weight loading play an important role in mature bone remodeling. However, the effect of weight loading on the developmental process in young bones is less well understood. In this work, chicks were loaded with bags weighing 10% of their body weight during their rapid growth phase. The increased load reduced the length and diameter of the long bones. The average width of the bag-loaded group's growth plates was 75 +/- 4% that of the controls, and the plates showed increased mineralization. Northern blot analysis, in situ hybridization, and longitudinal cell counting of mechanically loaded growth plates showed narrowed expression zones of collagen types II and X compared with controls, with no differences between the relative proportions of those areas. An increase in osteopontin (OPN) expression with loading was most pronounced at the bone-cartilage interface. This extended expression overlapped with tartarate-resistant acid phosphatase staining and with the front of the mineralized matrix in the chondro-osseous junction. Moreover, weight loading enhanced the penetration of blood vessels into the growth plates and enhanced the gene expression of the matrix metalloproteinases MMP9 and MMP13 in those growth plates. On the basis of these results, we speculate that the mechanical strain on the chondrocytes in the growth plate causes overexpression of OPN, MMP9, and MMP13. The MMPs enable penetration of the blood vessels, which carry osteoclasts and osteoblasts. OPN recruits the osteoclasts to the cartilage-bone border, thus accelerating cartilage resorption in this zone and subsequent ossification which, in turn, contributes to the observed phenotype of narrower growth plate and shorter bones. 相似文献

A method for calculating and optimizing the composition of materials for soft X-ray sources used in research on inertial confinement fusion is described. For a target-converter, a material composition is determined with which the conversion of laser light into X radiation is highly efficient. A comparative analysis is carried out of the efficiencies of generation of soft X-ray emission in the plasmas of some composite materials of thin conductors (wires) used as loads in X-and Z-pinches. Numerical calculations of the optical plasma properties are reported whose results make it possible to judge the emissivity of plasmas of different materials. Calcareous nannofossils originated in the Triassic, radiated in the Jurassic and became a dominant component of the marine
biosphere from the earliest Jurassic onward. They can be considered as one of the most important “innovations” of the Mesozoic
oceans. Their basic morphology allows the differentiation of three different groups: coccoliths, nannoliths and calcispheres
(= calcareous dinocysts). Only coccoliths and nannoliths are discussed in this article in some detail. Coccoliths and nannoliths
have contributed greatly in the Interpretation of Mesozoic marine Systems through biostratigraphy and palaeoecology/palaeoceanography.
Ever since the late 1960s both coccoliths and nannoliths have proven to be useful and reliable zonal markers for biostratigraphic
schemes, allowing detailed zonations for the Jurassic and Cretaceous. Though affected by palaeobiogeographic provincialism,
coccoliths and nannoliths have supplied many cosmopolitan biostratigraphic markers. These allow a global correlation of marine
sedimentary units both from onshore sections in the classical European and North American areas and pelagic sequences recovered
in the course of the DSDP/ODP drilling from the world’s oceans. Thus research on calcareous nannofossils Covers both, regional
and global aspects. Research in the last 15 years concentrated on palaeoecological aspects. Apart from dinoflagellates, coccolithophores
were the most important primary producers in Mesozoic oceans. As such they heavily relied on autecological factors such as
light, nutrients and temperature. Variations in the assemblage composition of these groups may thus be viewed as a key for
understanding palaeoecological, palaeoceanographic and palaeoclimatic changes of the past.

We have examined the indirect effects of UV-A and UV-B on cypris attachment of the barnacle Balanus amphitrite Darwin through their effects on microbial films. Specifically, we tested the hypothesis that both UV-A and UV-B radiation can indirectly affect the larval attachment of barnacles by altering the microbial film bioactivity. Microbial films were developed from mid-intertidal region (∼1 m above Mean Low Water Level) for 6 days and subjected to ambient levels of ultraviolet radiation. Response of cyprids to untreated and UV-treated microbial films was investigated using double-dish still water choice bioassay. Results showed that both UV-A and UV-B caused a decrease in the percentage of respiring bacterial cells in microbial films and this effect increased with UV energy. With the same UV energy, UV-B caused a greater decrease in respiring bacterial cells than UV-A. However, despite strong UV radiation, the bioactivities of microbial films (i.e., stimulation of cypris attachment) remain unchanged. Results of this study suggest that increased UV radiation, which might occur due to ozone depletion, may not significantly affect the barnacle recruitment by means of affecting the inductive larval attachment cues of microbial films. 相似文献

Passive integrated transponder (PIT) tags were surgically implanted in 118 sea lamprey Petromyzon marinus ammocoetes which were left to recover in the laboratory for 2 months. During this period 55 individuals started to metamorphose. In the late summer of 2002 the tagged animals were released in a small tributary of the River Mondego, Portugal, and were regularly monitored for a period of 2 months using a portable PIT tag reader. The distribution of the released animals changed from an initial uniform pattern to a random distribution, and then to an aggregated pattern. At the end of the first week 60% of the tagged sea lampreys had already left the study area, indicating their dynamic behaviour. Ammocoetes were more active than metamorphosing sea lampreys, and downstream movements were more frequent when compared to the upstream ones, which were usually a short distance. In order to determine the influence of the dark‐light cycle in the diel activity rhythms, 10 tagged sea lamprey ammocoetes were released in a tank (2000 l capacity) and their position monitored twice a day, for a period of 1 month. Ammocoetes locomotor activity appeared to be conditioned by circadian rhythms, and they were particularly active during darkness. 相似文献

Experiments on the transfection of cultured SKOV3 tumor cells of human ovarian adenocarcinoma and HeLa cells of human cervical carcinoma with gene Bax have demonstrated that SKOV3 cells are highly sensitive to the protein product of this gene, whereas the sensitivity of HeLa cells is substantially lower. HeLa cells obtained as a result of Bax transfection and subsequent selection are characterized by an extremely high Bax protein content and a hypersensitivity to doxorubicin. All Bax-transfected SKOV3 cells with an increased Bax content have died. In the SKOV3 cell line, a Bax exon 3 mutation has been found that corresponds to genotype G7/G9, whereas the native type of the Bax gene corresponds to genotype G8/G8. A search for fucoidan-degrading enzymes and other O-glycosylhydrolases has been performed among 51 strains of marine bacteria of the family Flavobacteriaceae isolated from red, green, and brown algae, as well as from the sea urchin Strongylocentrotus intermedius and the holothurian Apostichopus japonicus. Over 40% of the studied strains synthesized fucoidanases. The marine bacteria Mesonia algae KMM 3909T (an isolate from green alga Acrosiphonia sonderi), as well as Maribacter sp. KMM 6211 and Gramella sp. KMM 6054 (associants of the sea urchin S. intermedius), were the best producers of fucoidanases. Xylose effectively induced the biosynthesis of fucoidanases in these strains. None of the 15 strains of marine bacteria belonging to the genus Arenibacter produced polysaccharide hydrolases. 相似文献

The effect of denervation on the contractile activity of the saphenous artery in normotensive rats and rats with regional hypotension was studied. Hypotension was caused by partial occlusion of the abdominal aorta distally from the renal arteries, and then, in four weeks, to denervate the saphenous artery, a portion of the femoral nerve in one of the limbs was resected. In two more weeks, the contractile responses of ring preparations of the saphenous artery (after removal of the endothelium and block of neuronal uptake and β-adrenoreceptors) were investigated under isometric conditions. In normotensive rats, the denervation led to an increase in the vascular sensitivity to norepinephrine, phenylephrine, serotonin, and KCl. Similar changes in contraction were caused by chronic hypotension; however, rats with hypotension exhibited no additional denervation-induced increase in the vascular sensitivity. After treatment with glyoxylic acid, the fluorescence intensity of the vascular adrenergic fibers adapted to a reduced pressure was lower than that in the norm.
