Invasive plants that fix nitrogen can alter nutrient availability and thereby community dynamics and successional trajectories of native communities they colonize. Morella cerifera (Myricaceae) is a symbiotic nitrogen fixer originally from the southeastern U.S. that is colonizing native-dominated vegetation on a young lava flow near Hilo, Island of Hawai‘i, where it increases total and biologically available soil nitrogen and increases foliar nitrogen concentrations in associated individuals of the native tree Metrosideros polymorpha. This invasion has the potential to alter the few remaining native-dominated lowland forest ecosystems in windward Hawai‘i.  相似文献

Nurses are the largest group of healthcare providers and will be at the forefront during a response to a bioterrorism attack in the U.S. However, nurses' bioterrorism risk perceptions and their participation in bioterrorism preparedness activities, such as bioterrorism-related exercises or drills, have not been evaluated. We mailed a survey to all members of the Missouri Nurses Association in July 2006, consisting of 1,528 registered nurses. The instrument measured risk perception, perceived susceptibility, perceived seriousness, bioterrorism education received, participation in exercises/drills, and personal response plan thoroughness. The response rate was 31% (474/1,528). Most respondents believe that a bioterrorism attack will occur in the U.S. (82.3%; n = 390), but few (21.3%; n = 101) believe that one will occur in their community. The majority of nurses reported that they believe that a bioterrorism attack would have serious consequences (96.1%, n = 448), including having a serious impact on U.S. citizens' safety (90.7%, n = 446) and on their own safety (84.3%, n = 379). Most (60%, n = 284) reported that they had not received any bioterrorism-related education nor participated in any drills/exercises (82.7%, n = 392). Of those who had received education, most had participated in 3 or fewer programs and in only 1 drill. Few nurses (3.6%, n = 15) reported having all aspects of a personal bioterrorism response plan; approximately 20% (19.4%, n = 81) did not have any components of a plan. Most of the registered nurses in Missouri who were surveyed are not receiving bioterrorism education, participating in bioterrorism exercises, or developing thorough personal response plans. Nurses need to be aware of and encouraged to participate in the many education and training opportunities on bioterrorism and infectious disease disasters.  相似文献

Microbial infections of medical implants occur in more than 2 million surgical cases each year in the United States alone. These increase patient morbidity and mortality, as well as patient cost and recovery time. Many treatments are available, but none are guaranteed to remove the infection. In many cases, the device infections are caused by the adhesion of microbes to the implant, ensuing growth, pathogenesis, and dissemination. The purpose of this work is to examine the initial events in microbial adhesion by simulating the approach and contact between a planktonic cell, immobilized on an atomic force microscope (AFM) cantilever, and a biomaterial or biofilm substrate. The two model microbes used in this study, Candida parapsilosis (ATCC 90018) and Pseudomonas aeruginosa (ATCC 10145), were chosen for both their clinical relevance and their ease of acquisition and handling in the laboratory setting. Attractive interactions exist between C. parapsilosis and both unmodified silicone rubber and P. aeruginosa biofilms. Using C. parapsilosis cells immobilized on AFM cantilevers with a silicone substrate, we have measured attractive forces of 4.3 ± 0.25 nN in the approach portion of the force cycle. On P. aeruginosa biofilms, the magnitude of the attractive force decreases to 2.0 ± 0.40 nN and is preceded by a 2.0-nN repulsion at approximately 75 nm from the cell surface. These data suggest that C. parapsilosis may adhere to both silicone rubber and P. aeruginosa biofilms, possibly contributing to patient morbidity and mortality. Characterization of cell-biomaterial and cell-cell interactions allows for a quantitative link between the physicomechanical and physicochemical properties of implant materials and the nanoscale interactions leading to microbial colonization and infection.  相似文献

Patterns of fecal reproductive steroid metabolites and adrenal corticoids were characterized for 12‐ to 24‐month periods in black (n = 10 male, 16 female) and white (n = 6 male, 13 female) rhinoceroses at 14 institutions. All black rhinoceros females exhibited at least some ovarian cyclicity on the basis of fecal progestogen analysis (range, 2–12 cycles/yr). However, cycles often were erratic, with many being shorter (<20 days; 18% of cycles) or longer (>32 days; 21%) than the average of 26.8 ± 0.5 days (n = 104 cycles). Five females exhibited periods of acyclicity of 2–10‐month duration that were unrelated to season. One complete and seven partial pregnancies were evaluated in the black rhinoceros. Fecal progestogens increased over luteal phase concentrations after 3 months of gestation. Females resumed cyclicity within 3 months postpartum, before calves were weaned (n = 5). Approximately half of white rhinoceros females (6 of 13) showed no evidence of ovarian cyclicity. Of the cycles observed, 5 were “short” (32.8 ± 1.2 days) and 24 were “long” (70.1 ± 1.6 days). Only two females cycled continuously throughout the study. One had both long (n = 9) and short (n = 2) cycles, whereas the other exhibited long cycles only (n = 5). Fecal estrogen excretion was variable, and profiles were not useful for characterizing follicular activity or diagnosing pregnancy in either species. Males of both species showed no evidence of seasonality on the basis of fecal androgen profiles. Androgen metabolite concentrations were higher (P < 0.05) in the black (27.6 ± 6.9 ng/g) than in the white (16.8 ± 3.1 ng/g) rhinoceros. An adrenocorticotropin hormone challenge in four black rhinoceros males demonstrated that the clearance rate of corticoid metabolites into feces was ~24 hours. Fecal corticoid concentrations did not differ between males and females, but overall means were higher in the black (41.8 ± 3.1 ng/g) than in the white (31.2 ± 1.7 ng/g) rhinoceros. In summary, fecal steroid analysis identified a number of differences in hormonal secretory dynamics between the black and white rhinoceros that may be related to differences in reproductive rates in captivity. Most black rhinoceros females exhibited some cyclic ovarian activity. In contrast, few white rhinoceroses demonstrated evidence of regular estrous cyclicity, and those females that were active had comparatively long cycles. Results also suggest that fecal corticoid concentrations reflect adrenal activity and may be species specific. Continued studies are needed to determine whether fecal corticoid measurements will be useful for understanding the cause of inconsistent gonadal activity in these two species. Because all but three (15.8%) of the white rhinoceroses evaluated in this study were less than 20 years of age compared to 73.1% (19 of 26) of the black rhinoceroses, the impact of age on reproductive and adrenal activity also needs to be evaluated further. The reproductive rates of Russian wheat aphid, Diuraphis noxia (Kurdjumov) (Hemiptera: Aphididae), Biotype 1 (RWA 1) and Biotype 2 (RWA 2) were compared in the laboratory at three temperature regimes on a Russian wheat aphid resistant cultivar ('Prairie Red') and a susceptible cultivar ('TAM 107'). The objective of this study was to expose RWA 1 and RWA 2 to three temperature regimes and two levels of resistance to find whether there were reproductive differences that may occur within each biotype as well as differences in reproduction between biotypes. In addition, temperature effects of the Dn4 gene on biotype reproduction were noted. Differences in reproductive rates between the two biotypes seem to be driven by temperature. For both biotypes, longevity and reproductive rate parameters, except for intrinsic rate of increase, were lower at the 24-29 degree C temperature regime than the 13-18 degree C and 18-24 degree C temperature regimes. The intrinsic rate of increase was higher for both biotypes at the 18-24 degree C and 24-29 degree C temperature regimes than at the 13-18 degree C temperature regime. Reproductive rates between biotypes were similar at the two higher temperature regimes, but the fecundity for RWA 1 was less than RWA 2 at the 13-18 degree C temperature. The change in fecundity rates between RWA 1 and RWA 2 at lower temperatures could have ecological and geographical implications for RWA 2.  相似文献

1. In this study we predict stream sensitivity to non‐point source pollution based on the non‐linear responses of hydrological regimes and associated loadings of non‐point source pollutants to catchment properties. We assessed two hydrologically based thresholds of impairment, one for catchment storage (5–10%) and one for mature forest (<50% versus >60% of catchment in mature forest cover) across two different hydrogeomorphic regions within the Northern Lakes and Forest (NLF) ecoregion: the North Shore [predominantly within the North Shore Highlands Ecological Unit] and the South Shore (predominantly within the Lake Superior Clay Plain Ecological Unit). Water quality samples were collected and analysed during peak snowmelt and baseflow conditions from 24 second‐order streams grouped as follows: three in each region × catchment storage × mature forest class. 2. Water quality was affected by a combination of regional influences, catchment storage and mature forest. Regional differences were significant for suspended solids, phosphorus, nitrogen: phosphorus ratios, dissolved organic carbon (DOC) and alkalinity. Catchment storage was significantly correlated with dissolved silica during the early to mid‐growing season, and with DOC, specific conductance and alkalinity during all seasons. Total nitrogen and dissolved nitrogen were consistently less in low mature forest than in high mature forest catchments. Catchment storage interacted with the influence of mature forest for only two metrics: colour and the soluble inorganic nitrogen : phosphorus ratio. 3. Significant interaction terms (region by mature forest or region by storage) suggest differences in regional sensitivity for conductance, alkalinity, total organic carbon, and colour, as well as possible shifts in thresholds of impact across region or mature forest class. 4. Use of the NLF Ecoregion alone as a basis for setting regional water quality criteria would lead to the misinterpretation of reference condition and assessment of condition. There were pronounced differences in background water quality between the North and South Shore streams, particularly for parameters related to differences in soil parent material and glacial history. A stratified random sampling design for baseflow and snowmelt stream water quality based on both hydrogeomorphic region and catchment attributes improves assessments of both reference condition and differences in regional sensitivity.  相似文献

We present results from an ongoing field study conducted in Kansas grassland to examine correlates of invasibility and community stability along a natural gradient of plant diversity. Invasibility was evaluated by sowing seeds of 34 plant species into 40 experimental plots and then measuring colonization success after two growing seasons. Compositional stability, defined as resistance to change in species relative abundances over two growing seasons and in response to experimental disturbance, was measured in a separate set of 40 plots.
We found that community susceptibility to invasion was greatest in high diversity microsites within this grassland. Multiple regression analyses suggested that the positive correlation between invasibility and plant diversity was due to the direct influences of the extrinsic factors that contribute to spatial variation in diversity (soil disturbances; light availability), not to any direct impact of diversity. In addition, we found that compositional stability in response to disturbance was greatest within low diversity microsites and was strongly related to the dominance (evenness) component of diversity.  相似文献

The objective of this study was to generate bison x cattle hybrid embryos by in vitro fertilization, to assess their developmental potential, to determine the pattern of secretion of the embryonic signaling molecule interferon-tau (IFN-tau), and to identify novel IFN-tau mRNA polymorphism in the American plains bison. A total of 600 bovine oocytes were inseminated with frozen-thawed bison semen. Of these, 40.7% cleaved and 14.8% proceeded to the blastocyst stage. Individual blastocysts were cultured on a basement membrane (Matrigel) and their ability to attach and form outgrowths was monitored. A total of 36 blastocysts were cultured of which 22 formed outgrowths. During individual culture, medium samples were collected and their IFN-tau concentration was measured. On day 6 after onset of individual culture, attached outgrowths produced significantly more IFN-tau than unattached viable or degenerate blastocysts. At this time, female conceptuses also produced significantly more IFN-tau than their male cohorts. However, by day 12 this difference had disappeared. Total mRNA was extracted from three individual outgrowths and analyzed by RT-PCR. Subsequent sequencing of 28 clones showed several known bovine IFN-tau sequences as well as two novel sequences termed bisIFN-tau1 and 2. To determine the origin of these, DNA was extracted from bison semen and analyzed by PCR. One bovine IFN-tau sequence (bovIFN-tau1d) as well as bisIFN-tau2 and a third novel sequence bisIFN-tau3 were detected. This study demonstrates the feasibility of using hybrid embryos for the analysis of developmentally regulated gene expression in species where embryos may not be available.  相似文献

NADP-dependent isocitrate dehydrogenase is a low-copy nuclear gene family. We have sequenced two regions from an idh gene (idhB) near the 3' terminal end. The first fragment encodes 4 exons and 3 introns and is between approximately 600 and 950 bp in length. The second fragment includes three additional exons and introns and is between approximately 1200 and 1500 bp in length. The phylogenetic utility of the two sequence regions was evaluated in Polemoniaceae with a focus on Saltugilia, an incipient species complex that lacks phylogenetic resolution among these same taxa based on nuclear ribosomal ITS and chloroplast trnL. Multiple sequences from several individuals, multiple individuals from several populations, and multiple populations from all Saltugilia species were sampled to evaluate the taxonomic level at which idhB was useful as a phylogenetic marker in this clade. Phylogenies based on idhB sequences were compared with topological resolution and clade composition in ITS and trnL phylogenies. Phylogenies based on idhB and idhB in combination with ITS and trnL are better resolved than any other phylogenies for Saltugilia published to date, and character evolution within Saltugilia is explored.  相似文献

Background

DNA methylation occurs at preferred sites in eukaryotes. In Arabidopsis, DNA cytosine methylation is maintained by three subfamilies of methyltransferases with distinct substrate specificities and different modes of action. Targeting of cytosine methylation at selected loci has been found to sometimes involve histone H3 methylation and small interfering (si)RNAs. However, the relationship between different cytosine methylation pathways and their preferred targets is not known.

Results

We used a microarray-based profiling method to explore the involvement of Arabidopsis CMT3 and DRM DNA methyltransferases, a histone H3 lysine-9 methyltransferase (KYP) and an Argonaute-related siRNA silencing component (AGO4) in methylating target loci. We found that KYP targets are also CMT3 targets, suggesting that histone methylation maintains CNG methylation genome-wide. CMT3 and KYP targets show similar proximal distributions that correspond to the overall distribution of transposable elements of all types, whereas DRM targets are distributed more distally along the chromosome. We find an inverse relationship between element size and loss of methylation in ago4 and drm mutants.

Conclusion

We conclude that the targets of both DNA methylation and histone H3K9 methylation pathways are transposable elements genome-wide, irrespective of element type and position. Our findings also suggest that RNA-directed DNA methylation is required to silence isolated elements that may be too small to be maintained in a silent state by a chromatin-based mechanism alone. Thus, parallel pathways would be needed to maintain silencing of transposable elements.  相似文献
