Genome replication in picornaviruses is catalyzed by a virally encoded RNA-dependent RNA polymerase, termed 3D. These viruses also use a small protein primer, named VPg, to initiate RNA replication. The recent explosion of structural information on picornaviral 3D polymerases has provided insights into the initiation of RNA synthesis and chain elongation. Comparing these data with results from previous structural analyses of viral RNA-dependent RNA polymerases that catalyze de novo RNA synthesis sheds light on the different strategies that these viruses use to initiate replication. 相似文献

Since copper (Cu) is essential in key physiological oxidation reactions, organisms have developed strategies for handling Cu while avoiding its potentially toxic effects. Among the tools that have evolved to cope with Cu is a network of Cu homeostasis factors such as Cu-transporting P-type ATPases that play a key role in transmembrane Cu transport. In this work we present the functional characterization of an Arabidopsis Cu-transporting P-type ATPase, denoted heavy metal ATPase 5 (HMA5), and its interaction with Arabidopsis metallochaperones. HMA5 is primarily expressed in roots, and is strongly and specifically induced by Cu in whole plants. We have identified and characterized plants carrying two independent T-DNA insertion alleles, hma5-1 and hma5-2. Both mutants are hypersensitive to Cu but not to other metals such as iron, zinc or cadmium. Interestingly, root tips from Cu-treated hma5 mutants exhibit a wave-like phenotype at early stages and later on main root growth completely arrests whereas lateral roots emerge near the crown. Accordingly, these lines accumulate Cu in roots to a greater extent than wild-type plants under Cu excess. Finally, yeast two-hybrid experiments demonstrate that the metal-binding domains of HMA5 interact with Arabidopsis ATX1-like Cu chaperones, and suggest a regulatory role for the plant-specific domain of the CCH Cu chaperone. Based on these findings, we propose a role for HMA5 in Cu compartmentalization and detoxification. 相似文献

Arrhythmogenic right ventricular dysplasia/cardiomyopathy (ARVD/C) is a disorder characterized by fibrofatty replacement of cardiac myocytes that typically manifests in the right ventricle. It is inherited as an autosomal dominant disease with reduced penetrance, although autosomal recessive forms of the disease also occur. We identified four probands with ARVD/C caused by mutations in DSG2, which encodes desmoglein-2, a component of the cardiac desmosome. No association between mutations in this gene and human disease has been reported elsewhere. One of these probands has compound-heterozygous mutations in DSG2, and the remaining three have isolated heterozygous missense mutations, each disrupting known functional components of desmoglein-2. The protein and lipid composition of eukaryotic plasma membranes is highly dynamic and regulated according to need. The sphingolipid-responsive Pkh kinases are candidates for mediating parts of this regulation, as they affect a diverse set of plasma membrane functions, such as cortical actin patch organization, efficient endocytosis, and eisosome assembly. Eisosomes are large protein complexes underlying the plasma membrane and help to sort a group of membrane proteins into distinct domains. In this study, we identify Nce102 in a genome-wide screen for genes involved in eisosome organization and Pkh kinase signaling. Nce102 accumulates in membrane domains at eisosomes where Pkh kinases also localize. The relative abundance of Nce102 in these domains compared with the rest of the plasma membrane is dynamically regulated by sphingolipids. Furthermore, Nce102 inhibits Pkh kinase signaling and is required for plasma membrane organization. Therefore, Nce102 might act as a sensor of sphingolipids that regulates plasma membrane function. 相似文献

As the concentrations of essential vitamins and minerals and a number of other health‐promoting compounds are often low in stable crops, research is under way to understand and increase synthesis of phytochemicals in order to improve crop nutritional quality. The question addressed in this study is whether shoot genotypes or root genotypes with tolerant drought character are able to increase the fruit quality and production under moderate water stress. Zarina (drought tolerant) and Josefina (drought sensitive) were grafted self and reciprocally. The results showed that the use of cv. Zarina (drought tolerant) as rootstock (Zar × Jos) results in a greater number of fruits under water‐stress conditions, a moderate increase in some antioxidant compounds, such as ascorbic acid, phenols and flavonoids and lycopene and β‐carotene, and Zar × Jos showed high levels of sugars and organic acids, sweetness index and sugars : acids ratio, as well as important minerals, such as K and Mg, which could increase its nutritional quality under stress conditions. Overall, the use of drought‐tolerant rootstock could provide a useful tool to improve fruit yield and quality of cherry tomato fruit under moderate water stress, which in turn could mean lower costs in the use of irrigation water. 相似文献

Tetranychus urticae Koch (Acari: Tetranychidae) is a key pest of clementine mandarins, Citrus clementina Tanaka (Rutaceae), in Spain. This mite is highly polyphagous and can be easily found in clementine orchards, both in the trees and in the associated flora. In a previous study we found that the use of a cover of Festuca arundinacea Schreber (Poaceae) offered a better regulation of T. urticae populations than either bare soil or the traditional wild cover, which included a mix of weed species. We hypothesized that the selection of two host races of T. urticae, specialized in F. arundinacea and C. clementina, could partly explain the results obtained (bottom‐up regulation). Reciprocal transplant experiments show that sympatric deme × host combinations had higher mean fitness values than the allopatric combinations in clementine, but not in F. arundinacea, for most of the fitness parameters evaluated in our study. Because local adaptation implies mean deme fitness to be systematically higher for the sympatric deme × habitat combinations than for the allopatric ones, these results can be taken as indicative of occurrence of local adaptation in T. urticae. Molecular genetic analyses with microsatellite markers support this conclusion and indicate that local adaptation of T. urticae found in our system may indeed contribute to a better natural regulation of this mite. 相似文献

A phytogeographic analysis of the distributions of 454 species of trees native to the Osa Peninsula in 22 families revealed
that 4.8% of the species are endemic to the Osa Peninsula and the adjacent mainland of Costa Rica. However, nearly one-fourth
of the species might be regionally endemic to Central-South Mesoamerica (Costa Rica, Nicaragua, and Panama). Our sample suggests
that 53.6% of the species occur in some part of Mesoamerica and sometimes range into northwestern South America, and that
44.5% of the species have wide distributions throughout tropical America. There is a strong affinity with the flora of northwestern
South America, with 46.2% of the species on the Osa also found there. In addition, 50.6% of the tree species on the Osa occur
on both the Atlantic and Pacific slopes of Central America or, if they reach South America, are sometimes found on both sides
of the Andes. Major contributions to the tree flora of the Osa have been made by species arriving in the Osa by 1) dispersal
from South and North America to islands in proto Central America before the formation of a dry-land connection between the
two continents, and 2) migration from South America and North America after the closure of the Panamanian isthmus was made.
This analysis demonstrates the importance of the Osa as a regional refuge for protecting species with distributions limited
to the Osa and parts of Panama, Costa Rica, or Nicaragua. The Osa is also important because it harbors the last expanse of
tropical wet forest on the Pacific slope of Central America large enough to ensure the survival of the Central American populations

Exosomes are secreted cellular vesicles that can induce specific CD4+ T cell responses in vivo when they interact with competent antigen-presenting cells like mature dendritic cells (mDCs). The Trojan exosome hypothesis proposes that retroviruses can take advantage of the cell-encoded intercellular vesicle traffic and exosome exchange pathway, moving between cells in the absence of fusion events in search of adequate target cells. Here, we discuss recent data supporting this hypothesis, which further explains how DCs can capture and internalize retroviruses like HIV-1 in the absence of fusion events, leading to the productive infection of interacting CD4+ T cells and contributing to viral spread through a mechanism known as trans-infection. We suggest that HIV-1 can exploit an exosome antigen-dissemination pathway intrinsic to mDCs, allowing viral internalization and final trans-infection of CD4+ T cells. In contrast to previous reports that focus on the ability of immature DCs to capture HIV in the mucosa, this review emphasizes the outstanding role that mature DCs could have promoting trans-infection in the lymph node, underscoring a new potential viral dissemination pathway. 相似文献
