Keratinocyte differentiation and stratification are complex processes involving multiple signaling pathways, which convert a basal proliferative cell into an inviable rigid squame. Loss of attachment to the basement membrane triggers keratinocyte differentiation, while in other epithelial cells, detachment from the extracellular matrix leads to rapid programmed cell death or anoikis. The potential role of AKT in providing a survival signal necessary for stratification and differentiation of primary human keratinocytes was investigated. AKT activity increased during keratinocyte differentiation and was attributed to the specific activation of AKT1 and AKT2. Targeted reduction of AKT1 expression, but not AKT2, by RNA interference resulted in an abnormal epidermis in organotypic skin cultures with a thin parabasal region and a pronounced but disorganized cornified layer. This abnormal stratification was due to significant cell death in the suprabasal layers and was alleviated by caspase inhibition. Normal expression patterns of both early and late markers of keratinocyte differentiation were also disrupted, producing a poorly developed stratum corneum. 相似文献

Question: What are the importance of elevation and fire in maintaining ecotones of Florida scrub assemblages along a gradual topographic gradient? Location: Archbold Biological Station (ABS), 12 km south of Lake Placid, Florida, USA. Methods: Vegetation cover of upland Florida shrublands was quantified using the line‐intercept method along 20 transects traversing similar elevation gradients, stratified by time since fire (TSF). We objectively identified shrubland ecotones using a split moving windows boundary analysis (SMW) with three different window widths. Non‐metric multidimensional scaling ordination was used to determine relationships among plant assemblages defined by SMW. Results: We located up to four ecotones per transect, the majority of which were wide, highly heterogeneous zones. Relative elevation controlled the distribution of plant assemblages in upland Florida shrublands. Ecotones in shrublands > 30 years TSF had relatively low dissimilarity values in SMW, indicating that previously discrete plant assemblages with longer TSF were becoming more similar with time. Conclusions Split Moving Windows (SMW) analysis identified ecotones relatively well although patches generated by oak clonal growth were sometimes identified as ecotones. Fire suppression caused ecotones to become more diffuse, suggesting that without fire at least every 30 years, discrete plant assemblages within upland Florida shrublands will be more continuous. 相似文献

In the present work, the experimental and the theoretical vibrational spectra of trifluorothymine were investigated. The FT-IR (400-4000?cm(-1)) and μ-Raman spectra (100-4000?cm(-1)) of trifluorothymine in the solid phase were recorded. The geometric parameters (bond lengths and bond angles) and vibrational frequencies of the title molecule in the ground state were calculated using ab initio Hartree-Fock (HF) method and density functional theory (B3LYP) method with the 6-31++G(d,p) and 6-311++G(d,p) basis sets for the first time. The optimized geometric parameters and the theoretical vibrational frequencies were found to be in good agreement with the corresponding experimental data and with results found in the literature. Vibrational frequencies were assigned based on the potential energy distribution using the VEDA 4 program. The dimeric form of trifluorothymine was also simulated to evaluate the effect of intermolecular hydrogen bonding on the vibrational frequencies. It was observed that the stretching modes shifted to lower frequencies, while the in-plane and out-of-plane bending modes shifted to higher frequencies due to the intermolecular N-H?O hydrogen bonds. 相似文献

Empirical studies for different life histories have shown an inverse relationship between elasticity (i.e. the proportional contribution to population growth rate) and temporal variation in vital rates. It is accepted that this relationship indicates the effect of selective pressures in reducing variation in those life‐history traits with a major impact on fitness. In this paper, we sought to determine whether changes in environmental conditions affect the relationship between elasticity of vital rates and their temporal variation, and whether vital rates with simultaneously large elasticity and temporal variation might represent a characteristic life‐history strategy. We used demographic data on 13 populations of the short‐lived Hypericum cumulicola over 5–6 years, in three time‐since‐fire classes. For each population of each time‐since‐fire, we computed the mean matrix over years and its respective elasticity matrix, and the coefficients of variation in matrix entries over study years as an estimate of temporal variability. We found that mean elasticity negatively significantly correlated with temporal variation in vital rates in populations (overall eight out of 13) included in each time‐since‐fire. However, seedling recruitment exhibited both high elasticity and high temporal variation in almost all study populations. These results indicated that (1) the general relationship between elasticity and temporal variation in vital rates was not modified by environmental changes due to time‐since‐fire, and (2) high elasticity and high temporal variation in seedling recruitment in H. cumulicola is a particular trait of the species' life history. After seed survival in the soil seed bank, seedling recruitment represents the most important life‐history trait influencing H. cumulicola population growth rate (and fitness). The high temporal variability in seedling recruitment suggests that this trait is determined by environmental cues, leading to an increase in population size and subsequent replenishment of the seed bank in favorable years. 相似文献

Novel imidazopyridine derivatives were synthesized according to a very simple protocol and then subjected to cytotoxicity testing against LN-405 cells. Two of the compounds exhibited antiproliferative effects on LN-405 cells at 10 and 75?µM and were selected as lead compounds for further study. Safety experiment for lead compounds on WS1 was carried out and IC50 values were calculated as 480 and 844?µM. LN-405 cell line were incubated with the lead compounds and then tested for DNA damage by comet assay and effects on cell cycle using flow cytometry. The results of these two tests showed that both lead compounds affected the G0/G1 phase and did not allow the cells to reach the synthesis phase. The log BB (blood–brain barrier) and Caco-2 permeability of the synthesized molecules were calculated and it was shown that imidazopyridine derivatives taken orally are likely to pass through gastrointestinal membrane and the blood–brain barrier. 相似文献

Eryngium cuneifolium Small. (Apiaceae) is a narrowly distributed endemic found only in Ceratiola ericoides (Florida rosemary)-dominated Florida scrub, a periodically burned, shrub-dominated habitat. Multivariate analyses using 22 ∗∗∗microhabitat characteristics indicated significant microhabitat and time-since-fire effects on survival, growth, and fecundity of 1287 individuals over a 4-yr period. Survival increased with distance to the nearest shrub, and plants in larger open patches had greater survival rates. Neighboring shrubs of Ceratiola ericoides and Calamintha ashei were associated with a higher mortality of E. cuneifolium than other neighboring shrub species. Survival was reduced by two-thirds over 4 yr (14% vs. 42%) for E. cuneifolium near C. ericoides. Sand accretion increased growth and fecundity. With greater time since fire, woody shrubs increasingly dominate and open patches shrink, significantly reducing survival, growth, and fecundity of E. cuneifolium. Effects were particularly dramatic between 2 and 7 yr postfire, when annual mortality increased from <10% to >30% (r = 0.74). This herbaceous species is dependent on an open habitat maintained by periodic fire. Belowground competition or allelopathy from shrubs probably restricts E. cuneifolium to recently burned, open patches within the most xeric parts of Florida scrub. 相似文献

Temporal variations in the endothelium-dependent relaxant effects of acetylcholine (ACh) in mice and histamine (HA) in rat thoracic aorta have been studied. The relaxations induced by higher concentrations of ACh and HA were significantly dependent on the time the tissues were obtained. However, neither EC 50 (the concentration inducing half of the maximum response) values for ACh and HA, nor K B (antagonist dissociation constant) values for atropine and diphenhydramine were found to be statistically significant depending upon the time of obtaining aorta preparations. These results show that the in vitro responsiveness of mice and rat thoracic aortas to endothelium-dependent relaxant effects of ACh and HA, respectively, changes over a 24-h period. These variations might be dependent on a temporal rhythm in post-receptor events, i.e., guanylate cyclase-cGMP-phosphodiesterase system which mediates responses to endothelium-derived relaxing factor(s). 相似文献

The objective of this study was to evaluate the effect of lactose and dried whey supplementation as dietary component on growth performance and histology of lymphoid organs and ileum in broilers. A total of 480 day-old chicks were utilised for 42 days. Animals were assigned randomly to one of three treatments: control, lactose (2.5%), and dried whey (3.85%). Body weight was greater for animals supplemented with lactose or dried whey than for those not supplemented. There were no effects of treatments on feed intake and feed efficiency. In general, the effects of lactose or dried whey supplementation on histology of lymphoid organs and ileum were variable. Plasma cell counts were lower for animals supplemented with lactose than for those supplemented with dried whey. However, the length of intestinal villi during the starter period was greater for experimental groups than for control group. 相似文献
