The present study has been performed to elucidate a possiblerole of cell volume in low-density lipoprotein (LDL) binding andinternalization (LDLb+i). Asshown previously, increase of extracellular osmolarity (OSMe) andK+ depletion, both known to shrinkcells, interfere with the formation of clathrin-coated pits and thuswith LDLb+i. On the other hand,alterations of cell volume have been shown to modify lysosomal pH,which is a determinant of LDLb+i.LDLb+i have been estimated fromheparin-releasable (binding) or heparin-insensitive (internalization)uptake of 125I-labeled LDL. OSMewas modified by alterations of extracellular concentrations of ions,glucose, urea, or raffinose. When OSMe was altered by varying NaClconcentrations, LDLb+i decreased (by 0.5 ± 0.1%/mM) with increasing OSMe andLDLb+i increased (by 1.2 ± 0.1%/mM) with decreasing OSMe, an effect mainly due to alteredaffinity; the estimated dissociation constant amounted to 20.6, 48.6, and 131.6 µg/ml at 219, 293, and 435 mosM, respectively. A 25%increase of OSMe increased cytosolic (by 0.46 ± 0.03) and decreasedlysosomal (by 0.14 ± 0.02) pH. Conversely, a 25% decrease of OSMedecreased cytosolic (by 0.28 ± 0.02) and increased lysosomal (by0.17 ± 0.02) pH. Partial replacement of extracellularNa+ withK+ had little effect onLDLb+i, although it swelledhepatocytes and increased lysosomal and cytosolic pH. Hypertonicglucose, urea, or raffinose did not exert similar effects despite ashrinking effect of hypertonic raffinose. Monensin, which completelydissipates lysosomal acidity, virtually abolishedLDLb+i. In conclusion, theobservations reveal a significant effect of ionic strength onLDLb+i. The effect is, however,not likely to be mediated by alterations of cell volume or alterationsof lysosomal pH.

Zusammenfassung In der vorliegenden Arbeit wird ein einfaches Modell abgeleitet. Es dient der Sch?tzung additiver, dominanz- und interaktionsbedingter Beitr?ge definierter Gene zu einem metrischen Merkmal. Die zur Sch?tzung ben?tigten Formeln werden für 2 und 3 Loci mit je 2 Allelen gegeben. Voraussetzung für die Anwendung ist ein genetisches Material, in dem bein untersuchten Loci alle 3 n Genotypen vor einem m?glichst isogenen Hintergrund vorliegen. Werden die Versuche unter verschiedenen Umweltbedingungen wiederholt, so k?nnen auch die Interaktionen zwischen der Umwelt und den verschiedenen genetischen Komponenten erfa?t werden. Die Ph?notypen sind mit Hilfe der ermittelten Sch?tzwerte eindeutig beschreibbar.
Simulation of quantitative characters by genes with biochemically definable action
Summary The present paper contains the derivation of a simple model for estimating additive, dominant and epistatic contributions to a metric character by clearly defined genes. Formulas for 2 and 3 loci with 2 alleles each are given. If n loci are investigated, the model presupposes genetic material comprising all of the 3n possible genotypes on an isogenic background. Interactions between genotypic components and environment are recognizable when the experiments are repeated in different environments. By means of the estimated parameters the phenotypes can be clearly described.

Antigen structure modulation represents an approach towards designing subunit malaria vaccines. A specific epitope's alpha carbon stereochemistry, as well as its backbone topochemistry, was assessed for obtaining novel malarial immunogens. A variety of MSP-1(38-61) Plasmodium falciparum epitope pseudopeptides derived were synthesised, based on solid-phase pseudopeptide chemistry strategies; these included all-L, all-D, partially-D substituted, all-Psi-[NH-CO]-Retro, all-Psi-[NH-CO]-Retro-inverso, and Psi-[CH2NH] reduced amide surrogates. We demonstrate that specific recombinant MSP-1(34-469) fragment binding to red blood cells (RBCs) is specifically inhibited by non-modified MSP-1(42-61), as well as by its V52-L53, M51-V52 reduced amide surrogates and partial-D substitutions in K48 and E49. In vivo tests revealed that reduced amide pseudopeptide-immunised Aotus monkeys induced neutralising antibodies specifically recognising the MSP-1 N-terminus region. These findings support the role of molecular conformation in malaria vaccine development.  相似文献

A mutant blocked in anthocyanin synthesis leads to an accumulation of 4,2,4,6-tetrahydroxy-chalcone-2-glucoside (isosalipurposide) in blossoms of Callistephus chinesis (L.) Nees, whereas in geno-types with the wild-type allele, higher oxidized flavonoids and anthocyanins are synthesized. Measurements of chalcone-flavanone isomerase activity of 18 lines of Callistephus chinensis showed a clear correlation between accumulation of chalcone in the recessive genotypes (ch ch) and deficiency of this enzyme activity. Both the chemogenetic and the enzymologic evidence lead to the following conclusions: 1. The first product of the synthesis of the flavonoid skeleton should be tetrahydroxychalcone.-2. Osmotic cell swelling activates Cl channels to achieve anion efflux. In this study, we find that both the tyrosine kinase inhibitor herbimycin A and genetic knockout of p56lck, a src-like tyrosine kinase, block regulatory volume decrease (RVD) in a human T cell line. Activation of a swelling-activated chloride current (ICl−swell) by osmotic swelling in whole-cell patch-clamp experiments is blocked by herbimycin A and lavendustin. Osmotic activation of ICl−swell is defective in p56lck-deficient cells. Retransfection of p56lck restores osmotic current activation. Furthermore, tyrosine kinase activity is sufficient for activation of ICl−swell. Addition of purified p56lck to excised patches activates an outwardly rectifying chloride channel with 31 pS unitary conductance. Purified p56lck washed into the cytoplasm activates ICl−swell in native and p56lck-deficient cells even when hypotonic intracellular solutions lead to cell shrinkage. When whole-cell currents are activated either by swelling or by p56lck, slow single-channel gating events can be observed revealing a unitary conductance of 25–28 pS. In accordance with our patch-clamp data, osmotic swelling increases activity of immunoprecipitated p56lck. We conclude that osmotic swelling activates ICl−swell in lymphocytes via the tyrosine kinase p56lck.  相似文献

Summary Investigations made on diploid and experimentally produced autotetraploidSilene armeria have brought about the result, that the red flower colour of the diploid heterozygotes, first described byCorrens (1912), in contrast to the whitish and pink homozygotes, is likewise to be observed in the three tetraploid heterozygotes.By means of colorimetric measurements it was possible to find a gene dosage effect on the tetraploid heterozygotes (vide Figs. 1 and 2, and Table 3). Paper chromatography of the flower colour pigments of the three diploid phenotypes did not yield any difference in the quality of the pigments. With biochemical methods, however, we found that the nearly colourless albida-types contain leuco-cyanidin, whereas in coloured flowers cyanidin glycosides occur instead of leuco-cyanidin.The question is discussed, whether this phenomenon of heterosis depends on alleles, pseudoalleles or a complex-locus. The latter possibility is the most probable one.

Among the more than one hundred formally described human papillomavirus (HPV) types, 18 are referred to as high-risk HPV types due to their association with anogenital cancer. Despite pathogenic similarities, these types form three remotely related taxonomic groups. One of these groups is called HPV species 9 and is formed by HPV-16, the most common and best-studied type, together with HPV-31, -33, -35, -52, -58, and -67. Previous worldwide comparisons of HPV-16 samples showed about 2% nucleotide diversity between isolates, which were subsequently termed variants. The distribution of divergent variants has been found to correlate frequently with the geographic origin and the ethnicity of the infected patients and led to the concept of unique African, European, Asian, and Native American HPV-16 variants. In the current study, we address the question of whether geography and ethnicity also correlate with sequence variations found for HPV-31, -35, -52, and -58. This was done by sequencing the long control region in samples derived from Europe, Asia, and Africa, and from immigrant populations in North and South America. We observed maximal divergence between any two variants within each of these four HPV types ranging from 1.8 to 3.6% based on nucleotide exchanges and, occasionally, on insertions and deletions. Similar to the case with HPV-16, these mutations are not random but indicate a relationship between the variants in form of phylogenetic trees. An interesting example is presented by a 16-bp insert in select variants of HPV-35, which appears to have given rise to additional variants by nucleotide exchanges within the insert. All trees showed distinct phylogenetic topologies, ranging from dichotomic branching in the case of HPV-31 to star phylogenies of the other three types. No clear similarities between these types or between these types and HPV-16 exist. While variant branches in some types were specific for Europe, Africa, or East Asia, none of the four trees reflected human evolution and spread to the extent illustrated by HPV-16. One possible explanation is that the rare HPV types that we studied spread and thereby diversified more slowly than the more abundant HPV-16 and may have established much of today's variant diversity already before the worldwide spread of humans 100,000 years ago. Most variants had prototypic amino acid sequences within the E6 oncoprotein and a segment of the L1 capsid protein. Some had one, two, or three amino acid substitutions in these regions, which might indicate biological and pathogenic diversity between the variants of each HPV type.  相似文献
