This study was conducted at a high-latitude location (32°N; Kochi, Japan), where annual seawater temperatures show large fluctuations due to the meandering of the Kuroshio Current, providing a unique opportunity to examine the influence of temperature on coral reproduction. Annual spawning of individual colonies of four reef coral species-two Acropora species (Acropora hyacinthus and A. japonica) and two faviid species (Favites pentagona and Platygyra contorta)-was monitored in situ for 4 years in 2006-2009. The spawning of the four species always occurred around the last quarter moon in the local summer, July or August, irrespective of high annual variations in seawater temperatures (from 23.7 to 29.5 °C) and weather during the spawning period. However, the exact timing of spawning during the spawning period varied among the years and was correlated with the cumulative seawater temperature during the late period of gametogenesis (0-3 months before spawning). When seawater temperatures were higher, spawning occurred in the earlier spawning month (July) and vice versa, except in A. hyacinthus, which always spawned in July. In the case of the two Acropora species, higher (lower) temperatures led to spawning earlier (later) in the lunar cycle. In this article, the effects of allicin, a biological active compound of garlic, on HL60 and U937 cell lines were examined. Allicin induced growth inhibition and elicited apoptotic events such as blebbing, mitochondrial membrane depolarization, cytochrome c release into the cytosol, activation of caspase 9 and caspase 3 and DNA fragmentation. Pretreatment of HL60 cells with cyclosporine A, an inhibitor of the mitochondrial permeability transition pore (mPTP), inhibited allicin-treated cell death. HL60 cell survival after 1 h pretreatment with cyclosporine A, followed by 16 h in presence of allicin (5 microM) was approximately 80% compared to allicin treatment alone (approximately 50%). Also N-acetyl cysteine, a reduced glutathione (GSH) precursor, prevented cell death. The effects of cyclosporine A and N-acetyl cysteine suggest the involvement of mPTP and intracellular GSH level in the cytotoxicity. Indeed, allicin depleted GSH in the cytosol and mitochondria, and buthionine sulfoximine, a specific inhibitor of GSH synthesis, significantly augmented allicin-induced apoptosis. In HL60 cells treated with allicin (5 microM, 30 min) the redox state for 2GSH/oxidized glutathione shifted from EGSH -240 to -170 mV. The same shift was observed in U937 cells treated with allicin at a higher concentration for a longer period of incubation (20 microM, 2 h). The apoptotic events induced by various concentrations of allicin correlate to intracellular GSH levels in the two cell types tested (HL60: 3.7 nmol/10(6) cells; U937: 7.7 nmol/10(6) cells). The emerging mechanistic basis for the antiproliferative function of allicin, therefore, involves the activation of the mitochondrial apoptotic pathway by GSH depletion and by changes in the intracellular redox status. 相似文献

Genetic variability in individual troops of the Japanese macaque (Macaca fuscata fuscata) was quantified by the proportion of polymorphic loci and the average heterozygosity per individual from the results of starch-gel
electrophoreses of blood proteins controlled by 32 independent genetic loci. The former averaged 9.2% and the latter 1.3%,
the values being remarkably lower than those estimated for other animal populations. Geographical distribution of the genetic
variations was not uniform in the whole species but the variants occurred only in limited areas. Assuming the selective neutrality
of segregating alleles and the two-dimensional stepping-stone model of population structure, the genetic migration rate between
the local demes per generation could be estimated to average less than inverse of average effective deme size. Here, the local
deme is not a troop itself, but it consists of several troops tightly connected with each other by frequent exchanges of reproductive
males. Analyses of correlation between geographic and genetic distances between troops revealed that the gene constitutions
of two troops apart more than 100 km on an island could be regarded as practically independent of each other. These results
suggest that the population structure of the Japanese macaque species has a tendency to split into a number of local subpopulations

The mechanism of placental alkaline phosphatase (PLAP) induction by prednisolone in a uterine cervical epidermoid cancer cell line SKG-IIIa was investigated in vitro by enzyme-cytochemistry, enzyme immunoassay, Northern and Southern blot analysis, and in situ hybridization. Enzyme-cytochemical alkaline phosphatase (ALP) staining and immunoassay revealed increased levels of PLAP (heat-stable ALP) in prednisolone-treated cells. Northern blot analysis and in situ hybridization showed increased amounts of PLAP mRNA. Southern blot analysis indicated that PLAP was not a product of an amplified or rearranged gene. These findings suggest that the induction of PLAP mRNA in SKG-IIIa cells by prednisolone in turn increased the levels of PLAP. 相似文献

The fatty acid distribution pattern of lipids extracted from different subcellular components of Tetrahymena pyriformis was found to be significantly different from one type of membrane to another.The growth-temperature shift caused alterations in fatty acid composition. The ratio of palmitoleic to palmitic acid, especially, showed a sharp linear decline with increase of temperature in all of the membrane fractions.The spin labels were rapidly incorporated into Tetrahymena membranes. The order parameter of 5-nitroxide stearate spin label incorporated into various membrane fractions was found to be different for the different membrane fractions, suggesting the following order of the fluidity; microsomes > pellicles > cilia.The fluidity of the surface membranes, cilia and pellicles isolated from Tetrahymena cells grown at 15°C was noticeably higher than that of the membranes from cells grown at 34°C but was not so different with microsomal fractions.The motion of the spin label in the pellicular membrane was more restricted than in its extracted lipids, thus indicating the assumption that in Tetrahymena membranes the proteins influence the fluidity.It was also suggested that a sterol-like triterpenoid compound, tetrahymanol, which is principally localized in the surface membranes, would be involved in the membrane fluidity. 相似文献

Coupling of polyphosphoinositide breakdown with calcium efflux in formyl-methionyl-leucyl-phenylalanine-stimulated rabbit neutrophils 总被引:4,自引:0,他引:4
Exposure of rabbit neutrophils to formyl-methionyl-leucyl-phenylalanine (FMLP) induced the efflux of 45Ca2+ from pre-labeled cells which was almost complete within 30 s. On the other hand, FMLP-induced 45Ca2+ influx did not become apparent until 60 s after stimulation. When [3H]arachidonic acid-labeled neutrophils were stimulated with FMLP, the radioactivities in phosphatidylinositol 4,5-biphosphate (TPI) and phosphatidylinositol 4-phosphate (DPI) significantly decreased in parallel with the induction of 45Ca2+ efflux. In contrast, degradation of polyphosphoinositides in [3H]glycerol-labeled neutrophils was not significant until 60 s. Taken together, these results indicate that the early degradation of polyphosphoinositides, especially of those rich in arachidonic acid is closely associated with the initial efflux of calcium in FMLP-stimulated rabbit neutrophils. The study of resynthesis of polyphosphoinositides by measuring 32Pi incorporation into these lipids is also presented. 相似文献

Cell surface glycoprotein Thy-1 functions as a transformation suppressor in v-ras-transformed NIH/3T3 cells [Sugimoto et al., (1991) Cancer Res. 51, 99-104.]. In order to understand the mechanism of action of Thy-1, we examined the effect of Thy-1 expression on mitogen-induced Ca2+ oscillation which was correlated with v-ras-transformation [Fu et al., (1991) FEBS Lett. 281, 263-266.]. Forced expression of Thy-1 in v-ras-transformed cells inhibited mitogen-induced Ca2+ oscillation. Although v-Ras-free, Thy-1-positive NIH/3T3 cells (major population) did not show Ca2+ oscillation, whereas in Thy-1-negative NIH/3T3 cells (less than 1% of the population) Ca2+ oscillation was observed. Finally, replacement of the carboxyl-half of Thy-1 with that of CD4 abolished the inhibitory effect of Thy-1. These results suggest that Thy-1 directly or indirectly participates in the negative regulation of Ca2+ response by inhibiting Ca2+ oscillation. 相似文献

African non-human primates were surveyed seroepidemiologically for natural infection of human T-cell leukemia virus type I (ATLV/HTLV-I) or its closely related virus(es). Materials from three genera (Cercopithecus, Papio, and Theropithecus), four species (grivet monkey, Anubis baboon, Hamadryas baboon, and gelada), totalling 983 animals under natural conditions, were obtained in a field study in Ethiopia. Virus infection was determined by the indirect immunofluorescence test using HTLV-I specific antigens. Animals seropositive for HTLV-I were found among grivet monkeys and Anubis baboons including the hybrid offspring between Anubis and Hamadryas baboons but not pure-Hamadryas baboons and geladas. From these results, the HTLV-I family was proved to be widespread on the African continent and was regarded as a common retrovirus among catarrhines. 相似文献

Iron (Fe) is an important nutrient for phytoplankton. The low solubility of Fe in oxic waters can be a growth-limiting factor for phytoplankton. Synthetic aminopolycarboxylates (APCs) such as ethylenediaminetetraacetic acid (EDTA) and diethylenetriaminepentaacetic acid (DTPA) are widely used as Fe complexing agents for microalgae culture. However, the presence of these non-ready biodegradable APCS in aquatic systems may have serious environmental consequences. In the present study, the effects of biodegradable chelating ligands (hydroxyiminodisuccinic acid (HIDS), methylglycinediacetic acid (MGDA), and iminodisuccinate (IDS)) on Fe uptake in and growth of three coastal microalgae (Heterosigma akashiwo, Prymnesium parvum, and Skeletonema marinoi-dohrnii complex) were investigated, and the results were compared with those of non-ready biodegradable APCs (EDTA, ethylenediamine tetra-methylene phosphonic acid (EDTMP), and DTPA). The biodegradable chelating ligands did not have significant growth inhibition effect on the phytoplankton. Although the growth of the algae (except S. marinoi-dohrnii complex) was not affected substantially by 1.5 and 7.5 μM of DTPA, growth inhibition occurred by 7.5 μM of EDTMP and 150 μM of EDTA, DTPA, and EDTMP. The effect of chelating ligands on microalgal growth was likely to be associated with the intracellular Fe uptake influenced by the chelating ligands. On average, intracellular Fe concentrations for biodegradable chelating ligands were substantially higher than those for non-ready biodegradable APCs. Except H. akashiwo, the ratio of intra/extracellular Fe concentrations was highest for MGDA followed by IDS and HIDS. The results indicate that biodegradable chelating ligands are more efficient than non-ready biodegradable APCs in intracellular Fe uptake and algal growth. 相似文献
