Rotavirus RNA was detected in nasopharyngeal washings, obtained from patients with rotavirus gastroenteritis (76.3% of cases) and gastroenteritis of unknown etiology (34.7% of cases), by the method of molecular hybridization with the use of RNA of monkey virus SA11, labeled with 32P in the process of T4-ligase reaction. In 100% of cases this detection correlated with the presence of lesions of the upper respiratory ways and was probably indicative of the rotavirus nature of the catarrhal syndrome in rotavirus gastroenteritis.  相似文献

Circahoral opposite-in phase fluctuations of protein syntheses and intracellular ATP content have been observed in monolayer hepatocyte cell culture. Peculiarities of protein synthesis in hepatocytes have been studied in vitro in presence of adenylic and guanylic nucleotides. Addition of exogenous ATP leads to the decrease in the level of protein synthesis and smoothing off of the fluctuations. The presence of exogenous ADP leads to the increase in protein synthesis and retaining of the amplitude of fluctuations of this process. Effect of exogenous GTP is similar to that of ATP. Different aspects of action of exogenous NTPs on the rhythms of protein synthesis have been considered.  相似文献

In primary monolayer culture of hepatocytes, circahoralian rhythms of protein synthesis and secretion were discovered. In one of the hepatocyte populations the rhythmic cycle of protein secretion was found to be about twice as long as that of protein synthesis. Combination of different experimental variants such as the impulse and the long-term labelling of proteins or inhibition of protein synthesis, revealed that the secretion rhythms were predetermined by the secretion of both newly formed proteins and those stored in the cell for a long time. Upon the inhibition of protein synthesis with cyclohexeimide, the protein secretion in the monolayer changes its rhythmic pattern for a constant rate secretion. Possible causes of the alteration of circahoralian variations of protein synthesis are discussed.  相似文献

The enzymes of the Entner-Doudoroff pathway (EDP) and/or the oxidative branch of the pentose phosphate pathway (OBPPP) have been inactivated in the E. coli 2Δ (ΔsucAB, ΔaceAK, P L -brnQ) [pEL-IDO] strain, which is used for the biotransformation of L-isoleucine into 4-hydroxyisoleucine (4-HIL) by enzymatic hydroxylation. We established that the inactivation of the EDP in the 2Δ [pEL-IDO] strain does not virtually affect the basic parameters of the process, whereas the blocking of the OBPPP reduces the efficiency of biomass accumulation and L-isoleucine transformation in conversion to glucose. However, the simultaneous inactivation of the enzymes of the EDP and OBPPP in the 2Δ [pEL-IDO] strain allowed us to achieve a considerable improvement of the process, namely, a 20% reduction in glucose consumption and a 35% decrease in biomass yield at a constant L-isoleucine/4-HIL conversion rate at a level of 88%.  相似文献

The bovine adrenal cortex adrenodoxin gene was inserted into pTZ19 under T7 promoter control. The adrenodoxin mRNA was synthesized with T7 RNA polymerase and then translated in the reticulocyte cell-free translation system. The protein product was identified as the adrenodoxin precursor with molecular weight 22000. The import of the precursor into isolated yeast mitochondria was carried out. The protein was found to be inserted into the trypsin-insensitive compartment of mitochondria via an energy dependent way. This resulted in the processing of the precursor to the 12000-mature form. Thus, the precursor of mammalian adrenodoxin can be normally imported into yeast mitochondria.  相似文献

The nature of cells responsible for the genetic resistance of lethally irradiated CBA mice to lymphocytes of (CBA x M523)F1 hybrids was studied. Preirradiation of the hosts was shown to abolish the resistance. The latter was generally recovered by syngeneic thymocytes or splenocytes while embryonic liver and bone marrow cells or splenocytes treated with anti-Thy-I serum plus complement before injection into host were ineffective. It is postulated that some cells with T cell characteristics are responsible for the phenomenon of parental resistance. These cells differ in several respects from T cells that mediate the transplantation immunity and from M cells that control other forms of the genetic resistance.  相似文献
