The adjuvant activity in DNA immunization against herpes simplex virus type 1 (HSV1) was studied for the granulocyte-macrophage colony-stimulating factor (GM-CSF) synthesized from an eukaryotic expression plasmid (pDNAGM-CSF) and for the synthetic glucosaminyl-muramyl dipeptide (GMDP). A plasmid containing the HSV gD gene (pDNAgD) was used as an immunogen. GMDP and pDNAGM-CSF each enhanced the T-cell immune response to DNA immunization. The protective effect of DNA immunization increased from 63 to 100% when the two plasmids were injected simultaneously and to 96% when pDNAgD was injected one day after injecting GMDP. Coulomb explosion of a charged, nonuniform, and spherically symmetric cluster with a finite ion temperature is investigated. The spatial distributions of the density and mean velocity of accelerated ions, as well as their energy spectra, are obtained and analyzed as functions of the initial temperature. It is shown that taking into account the finite ion temperature eliminates singularities emerging during a Coulomb explosion of a spatially nonuniform cold cluster due to multistream ion motion that arises after breaking the velocity profile of the cluster ions. The characteristic temperature is found above which the spatial distribution and energy spectrum of the expanding ions become regular.  相似文献

The regularities of the reaction of aminopolysaccharide chitosan with glutaraldehyde (GA) have been considered. The equilibrium forms of GA in water have been thoroughly studied by NMR spectroscopy. It has been established that at pH 5.6, the exchange of the protons of O=CHCH2 groups for deuterium occurs, indicating the presence of an anion, a product of the first stage of the aldol reaction; at pH > 7.2, the formation of the products of an aldol reaction and aldol condensation takes place. The kinetics of the reaction between the amino groups of chitosan and GA, the kinetics of gel formation in chitosan solutions in the presence of GA, and the kinetics of changes in the rigidity of gels formed have been studied by UV spectroscopy. IR spectra of cross-linked chitosan have been obtained. It has been shown that chitosan catalyzes the polymerization of GA to form irregular products; in this process, the length of oligomeric chains in modified or cross-linked chitosan and the concentration of conjugated bonds increase with the GA concentration and pH of the reaction medium.  相似文献

We investigated the specific pattern of major urinary proteins (MUPs) expression in 3-, 4-, and 12-week old mice of CBA/LacY and C57BL/6JY inbred strains using polyacrylamide gel electrophoresis. Quantitative evaluation of 8 protein fractions A-H with regard to sex, age, and genotype of the animals is presented for the first time. Actual problems of genetic control and neuroendocrine regulation of MUPs expression during ontogenesis are discussed. In the light of current views on MUPs as a key component in intrapopulation information exchange via pheromones, we put forward the idea that the genetically determined structure of the olfactory code of the definitive type is formed at an early ontogenetic stage on the basis of the MUPs combinatorial pattern.  相似文献

Formation of free radicals in mitochondria plays a key role in the development of apoptosis, which includes formation of superoxide by the respiratory chain, formation of radicals by cytochrome c-cardiolipin complex in the presence of hydrogen peroxide or lipids, and chain lipid peroxidation resulting in cytochrome c release from mitochondria and initiation of the apoptotic cascade. In this work the effect of taxifolin (dihydroquercetin) and some other antioxidants on these three radical-producing reactions was studied. Peroxidase activity of the complex of cytochrome c with dioleyl cardiolipin estimated by chemiluminescence with luminol decreased by 50% with quercetin, taxifolin, rutin, Trolox, and ionol at concentrations 0.7, 0.7, 0.8, 3, and 10 μM, respectively. The lipid radical production detected by coumarin C-525-activated chemiluminescence decreased under the action of rutin and taxifolin in a dose-dependent manner, so that a 50% inhibition of chemiluminescence was observed at the antioxidant concentrations of 3.7 and 10 μM, respectively. Thus, these two radical-producing reactions responsible for apoptosis onset are inhibited by antioxidants at rather low concentrations. Experiments performed on liver slices and mash showed that taxifolin, quercetin, naringenin, and Trolox have low inhibitory effect on the lucigenin-dependent chemiluminescence in the tissue only at concentrations higher than 100 μM.  相似文献

The first bystrowianids recorded in the Gamskian Horizon of the East European Platform, Vyushkoviana operta gen. et sp. nov. and Dromotectum abditum sp. nov. (Amphibia, Chroniosuchia), are described based on specimens from the Malaya Northern Dvina and Don river basins. The patterns of the dermal scute evolution observed in bystrowianids warrant the subdivision of the group into the subfamilies Bystrowianinae Vjuschkov 1957, Dromotectinae subfam. nov. and Axitectinae subfam. nov. The second and third show progressive reduction of the external (paraxial) zone of the interosteoderm articulation. New data supporting close proximity or phyletic succession of the chroniosuchid and bystrowianid types of the osteoderm design are revealed. The concept of the Chroniosuchidae as a paraphyletic group is rejected. Evolutionary significance of the key features distinguishing the bystrowianid subfamilies is analyzed.  相似文献

According to materials of trap, long-line, and trawl fishing, specific features of distribution of sablefish Anoplopoma fimbria and some of its biological characteristics in Pacific waters off the southeastern coast of Kamchatka, continental slope of the western part of the Bering Sea, Shirshov Underwater Ridge, and off the Commander Islands are considered. Maximum density concentrations according to data of trap fishing was noted along southeastern coast of Kamchatka and the data of trawl fishing indicated most frequent catches in the western part of the Bering Sea in the area of Koryak coast up to Cape Navarin. The pattern of vertical distribution in different areas considerably differs. The magnitude of trap catches in different areas is different and determined by the type of trap and the period of soaking. The size composition, fatness, and the sex ratio are different in catches of different fishing gear and differ between regions. On the whole, in Russian Far Eastern waters, females mature in mass at a body length of 71 cm and males at 57 cm.  相似文献

A search for poly(ADP-ribose) polymerase-1 inhibitors by virtual screening of a chemical compound database and a subsequent experimental verification of their activities have been done. It was shown that the most efficient method to predict inhibitory properties implies a combinatorial approach joining molecular docking capabilities with structural filtration. Among more than 300000 database chemicals 9 PARP1 inhibitors were revealed; the most active ones, namely: STK031481, STK056130, and STK265022,--displayed biological effect at a micro-molar concentration (IC50 = 2.0 microM, 1.0 microM and 2.6 microM, respectively).  相似文献
